    Prior exercise and antioxidant supplementation: effect on oxidative stress and muscle injury

    <p>Abstract</p> <p>Background</p> <p>Both acute bouts of prior exercise (preconditioning) and antioxidant nutrients have been used in an attempt to attenuate muscle injury or oxidative stress in response to resistance exercise. However, most studies have focused on untrained participants rather than on athletes. The purpose of this work was to determine the independent and combined effects of antioxidant supplementation (vitamin C + mixed tocopherols/tocotrienols) and prior eccentric exercise in attenuating markers of skeletal muscle injury and oxidative stress in resistance trained men.</p> <p>Methods</p> <p>Thirty-six men were randomly assigned to: no prior exercise + placebo; no prior exercise + antioxidant; prior exercise + placebo; prior exercise + antioxidant. Markers of muscle/cell injury (muscle performance, muscle soreness, C-reactive protein, and creatine kinase activity), as well as oxidative stress (blood protein carbonyls and peroxides), were measured before and through 48 hours of exercise recovery.</p> <p>Results</p> <p>No group by time interactions were noted for any variable (P > 0.05). Time main effects were noted for creatine kinase activity, muscle soreness, maximal isometric force and peak velocity (P < 0.0001). Protein carbonyls and peroxides were relatively unaffected by exercise.</p> <p>Conclusion</p> <p>There appears to be no independent or combined effect of a prior bout of eccentric exercise or antioxidant supplementation as used here on markers of muscle injury in resistance trained men. Moreover, eccentric exercise as used in the present study results in minimal blood oxidative stress in resistance trained men. Hence, antioxidant supplementation for the purpose of minimizing blood oxidative stress in relation to eccentric exercise appears unnecessary in this population.</p

    Nonlocal restoration of two-mode squeezing in the presence of strong optical loss

    We present the experimental realization of a theoretical effect discovered by Olivares and Paris, in which a pair of entangled optical beams undergoing independent losses can see nonlocal correlations restored by the use of a nonlocal resource correlating the losses. Twin optical beams created in an entangled Einstein-Podolsky-Rosen (EPR) state by an optical parametric oscillator above threshold were subjected to 50% loss from beamsplitters in their paths. The resulting severe degradation of the signature quantum correlations observed between the two beams was then suppressed when another, independent EPR state impinged upon the other input ports of the beamsplitters, effectively entangling the losses inflicted to the initial EPR state. The additional EPR beam pair was classically coherent with the primary one but had no quantum correlations with it.     A 21 day Daniel Fast improves selected biomarkers of antioxidant status and oxidative stress in men and women

    <p>Abstract</p> <p>Background</p> <p>Dietary modification via both caloric and nutrient restriction is associated with multiple health benefits, some of which are related to an improvement in antioxidant status and a decrease in the production of reactive oxygen species. The Daniel Fast is based on the Biblical book of Daniel, is commonly partaken for 21 days, and involves food intake in accordance with a stringent vegan diet. The purpose of the present study was to determine the effect of a 21 day Daniel Fast on biomarkers of antioxidant status and oxidative stress.</p> <p>Methods</p> <p>43 subjects (13 men; 30 women; 35 ± 1 yrs; range: 20-62 yrs) completed a 21 day Daniel Fast following the guidelines provided by investigators. Subjects reported to the lab in a 12 hour post-absorptive state both pre fast (day 1) and post fast (day 22). At each visit, blood was collected for determination of malondialdehyde (MDA), hydrogen peroxide (H<sub>2</sub>O<sub>2</sub>), nitrate/nitrite (NOx), Trolox Equivalent Antioxidant Capacity (TEAC), and Oxygen Radical Absorbance Capacity (ORAC). Subjects recorded dietary intake during the 7 day period immediately prior to the fast and during the final 7 days of the fast.</p> <p>Results</p> <p>A decrease was noted in MDA (0.66 ± 0.0.03 vs. 0.56 ± 0.02 μmol L<sup>-1</sup>; p = 0.004), while H<sub>2</sub>O<sub>2 </sub>demonstrated a trend for lowering (4.42 ± 0.32 vs. 3.78 ± 0.21 μmol L<sup>-1</sup>; p = 0.074). Both NOx (18.79 ± 1.92 vs. 26.97 ± 2.40 μmol L<sup>-1</sup>; p = 0.003) and TEAC (0.47 ± 0.01 vs. 0.51 ± 0.01 mmol L<sup>-1</sup>; p = 0.001) increased from pre to post fast, while ORAC was unchanged (5243 ± 103 vs. 5249 ± 183 μmol L<sup>-1 </sup>TE; p = 0.974). As expected, multiple differences in dietary intake were noted (p < 0.05), including a reduction in total calorie intake (2185 ± 94 vs. 1722 ± 85).</p> <p>Conclusion</p> <p>Modification of dietary intake in accordance with the Daniel Fast is associated with an improvement in selected biomarkers of antioxidant status and oxidative stress, including metabolites of nitric oxide (i.e., NOx).</p

    Effect of betaine supplementation on plasma nitrate/nitrite in exercise-trained men

    Background: Betaine, beetroot juice, and supplemental nitrate have recently been reported to improve certain aspects of exercise performance, which may be mechanistically linked to increased nitric oxide. The purpose of the present study was to investigate the effect of betaine supplementation on plasma nitrate/nitrite, a surrogate marker or nitric oxide, in exercise-trained men.Methods: We used three different study designs (acute intake of betaine at 1.25 and 5.00 grams, chronic intake of betaine at 2.5 grams per day for 14 days, and chronic [6 grams of betaine per day for 7 days] followed by acute intake [6 grams]), all involving exercise-trained men, to investigate the effects of orally ingested betaine on plasma nitrate/nitrite. Blood samples were collected before and at 30, 60, 90, and 120 min after ingestion of 1.25 and 5.00 grams of betaine (Study 1); before and after 14 days of betaine supplementation at a dosage of 2.5 grams (Study 2); and before and after 7 days of betaine supplementation at a dosage of 6 grams, followed by acute ingestion of 6 grams and blood measures at 30 and 60 min post ingestion (Study 3).Results: In Study 1, nitrate/nitrite was relatively unaffected and no statistically significant interaction (p = 0.99), dosage (p = 0.69), or time (p = 0.91) effects were noted. Similar findings were noted in Study 2, with no statistically significant interaction (p = 0.57), condition (p = 0.98), or pre/post intervention (p = 0.17) effects noted for nitrate/nitrite. In Study 3, no statistically significant changes were noted in nitrate/nitrite between collection times (p = 0.97).Conclusion: Our data indicate that acute or chronic ingestion of betaine by healthy, exercise-trained men does not impact plasma nitrate/nitrite. These findings suggest that other mechanisms aside from increasing circulating nitric oxide are likely responsible for any performance enhancing effect of betaine supplementation. © 2011 Bloomer et al; licensee BioMed Central Ltd

    Cultural considerations at end of life in a geriatric inpatient rehabilitiation setting

    Aim: To explore the impact of cultural factors on the provision of end-of-life care in a geriatric inpatient rehabilitation setting. Background: Australia’s ageing population is now also one of the most culturally diverse. Individuals from culturally and linguistically diverse backgrounds may have specific care needs at the end of life according to various aspects of their culture. Design: A mixed method approach using a retrospective audit of existing hospital databases, deceased patients’ medical records, and in-depth interviews with clinicians. Findings: Patients’ and families’ cultural needs were not always recognised or facilitated in end-of-life care, resulting in missed opportunities to tailor care to the individual’s needs. Clinicians identified a lack of awareness of cultural factors, and how these may influence end-of-life care needs. Clinicians expressed a desire for education opportunities to improve their understanding of how to provide patient-specific, culturally sensitive end-of-life care. Conclusion: The findings highlight that dying in geriatric inpatient rehabilitation settings remains problematic, particularly when issues of cultural diversity further compound end-of-life care provision. There is a need for recognition and acceptance of the potential sensitivities associated with cultural diversity and how it may influence patients’ and families’ needs at the end of life. Health service organisations should prioritise and make explicit the importance of early referral and utilisation of existing support services such as professional interpreters, specialist palliative care and pastoral care personnel in the provision of end-of-life care. Furthermore, health service organisations should consider reviewing end-of-life care policy documents, guidelines and care pathways to ensure there is an emphasis on respecting and honouring cultural diversity at end of life. If use of a dying care pathway for all dying patients was promoted, or possibly mandated, these issues would likely be addressed. © 2018 Published by Elsevier Ltd on behalf of Australian College of Nursing Ltd

    Predictors of Cardiometabolic Disease Risk Factors in Professional Firefighters

    The leading cause of mortality (~50% of deaths) among on-duty firefighters is sudden cardiac death. Firefighters with adequate cardiorespiratory fitness levels (among other muscular fitness metrics) and lower body fat percentage (BF%) are likely to better combat on-duty cardiac events and cardiovascular disease (CVD) risk. Individuals with higher VO2max values may be at lower risk noted by lower oxidative stress and inflammatory biomarkers, which may be more ideal predictors of CVD risk compared to traditional biomarkers (i.e., blood glucose and lipids) among the fire community; however, more research is needed to elucidate. PURPOSE: This study examined the relationship between fitness metrics, body fat percentage, and traditional and non-traditional biomarkers among professional firefighters. METHODS: Ninety-eight professional, male firefighters (age = 36.26 ± 9.08 yrs; height = 179.53 ± 7.06 cm; waist circumference = 37.99 ± 4.13 cm; BF% = 24.39 ± 5.21; years of experience = 11.81 ± 8.00 yrs) from a local fire department were studied VO2max was estimated from total treadmill exercise time using the Foster equation.. Fitness metrics including muscular strength, endurance, and power variables were also measured. Fasted blood samples were analyzed for concentrations of lipids, glucose, insulin resistance (HOMA-IR), advanced oxidation protein products (AOPP), and ultra-sensitive C-reactive protein (CRP). To assess the relationship between fitness metrics and AOPP, CRP, and HOMA-IR, ordinary least square regression analyses were used. RESULTS: VO2max values were inversely (p \u3c 0.05) related to AOPP, HOMA-IR, and CRP. Waist circumference was positively correlated to AOPP concentrations and HOMA-IR, while increased BF% was significantly related to increased CRP concentrations. CONCLUSION: High VO2max values were significantly related to lower AOPP, CRP, and HOMA-IR. Additionally, high waist circumference and BF% values were related to increases in oxidative stress, inflammation, and insulin resistance. Firefighters are encouraged to maintain high cardiorespiratory fitness (VO2max) and lower BF% to reduce CVD and, ultimately, on-duty sudden cardiac death risk

    Real-Gas Effects and Phase Separation in Underexpanded Jets at Engine-Relevant Conditions

    A numerical framework implemented in the open-source tool OpenFOAM is presented in this work combining a hybrid, pressure-based solver with a vapor-liquid equilibrium model based on the cubic equation of state. This framework is used in the present work to investigate underexpanded jets at engine-relevant conditions where real-gas effects and mixture induced phase separation are probable to occur. A thorough validation and discussion of the applied vapor-liquid equilibrium model is conducted by means of general thermodynamic relations and measurement data available in the literature. Engine-relevant simulation cases for two different fuels were defined. Analyses of the flow field show that the used fuel has a first order effect on the occurrence of phase separation. In the case of phase separation two different effects could be revealed causing the single-phase instability, namely the strong expansion and the mixing of the fuel with the chamber gas. A comparison of single-phase and two-phase jets disclosed that the phase separation leads to a completely different penetration depth in contrast to single-phase injection and therefore commonly used analytical approaches fail to predict the penetration depth.Comment: Preprint submitted to AIAA Scitech 2018, Kissimmee, Florid
