    Underlying molecular mechanism in the modulation of the ram sperm acrosome reaction by progesterone and 17ß-estradiol

    Steroid hormones progesterone (P4) and 17ß-estradiol (E2) not only have important functions in regulation of reproductive processes in mammals but also have direct effects on spermatozoa. There can be induction of the acrosome reaction in ram spermatozoa by P4 and E2 and, in the present study, there was further investigation of mechanisms underlying this effect. In a medium containing agents that increase cAMP, the presence of both P4 and E2 led to changes in the localization of proteins phosphorylated in tyrosine residues evaluated by indirect immunofluorescence. The inclusion of P4 at 1 µM in the media induced an increase in Ca2+i and mobilization in the area of the acrosome (Fluo-4 and Rhod-5 staining, respectively), an increase in ROS (H2DCFDA staining) and a substantial disruption of the acrosome (evaluated using RCA), while E2 did not have these effects. There were no effects on cAMP concentrations or PKA activity with inclusion of these hormones in the media. The inclusion of P4 at 100 pM in the media led to changes in values for sperm kinematic variables which could indicate there was an inhibition of the hyperactivation caused by agents that induce an increase in cAMP concentrations. In conclusion, results from the present study indicate that P4 and E2 promote mechanisms regulating the acrosome reaction in ram spermatozoa, however, these effects on mechanisms are different for the two hormones, and for E2, require further clarification

    High-temperature anti-Invar behavior of gamma-Fe precipitates in Fe_xCu_(100-x) solid solutions: Ferromagnetic phases

    High-temperature magnetization and neutron diffraction measurements on metastable Fe_xCu_(100-x) solid solutions have recently shown to imply that γ-Fe precipitates present ferromagnetic anti-Invar behavior. For this reason, we have studied the ferromagnetic phases of γ-Fe in moment-volume parameter space, using the general potential linearized-augmented plane-wave method and the fixed spin moment procedure in order to calculate the corresponding total energy. We find that only two ferromagnetic phases (one related to a low- spin state and the other to a high-spin state) can exist and even coexist in limited volume ranges (3.55-3.59 Å). Hence, our results provide a "revisited" version of the local spin density calculations used in the early article by Moruzzi [Phys. Rev. B 34, 1784 (1986)]. In addition, the fixed spin moment method-using an energy-moment-volume space representation-allows us to conclude that the high-spin state is the ground state of the gamma-Fe precipitates, as the anti-Invar behavior is an intrinsic property of these states. This simple scenario seems to adequately describe the perplexing phenomenology recently observed on Fe_xCu_(100-x) solid solutions

    Changes in melatonin concentrations in seminal plasma are not correlated with testosterone or antioxidant enzyme activity when rams are located in areas with an equatorial photoperiod

    In temperate climates, photoperiod and melatonin regulate ram reproduction, modulating hormonal secretions, sperm quality, and seminal plasma composition. Information on the effect of an equatorial photoperiod (12L:12D) on ram reproduction, however, is scarce, and no data on hormonal concentrations and antioxidant enzyme activity in seminal plasma have been reported. Thus, the variation was investigated of melatonin and its relationship with testosterone and antioxidant enzyme activity in the seminal plasma of three sheep breeds in Colombia, when there was a consistent photoperiod during two dry and two rainy seasons per year. Semen was collected once a week from 12 mature rams (four of each breed: Colombian Creole, Hampshire, and Romney Marsh). Seminal plasma was obtained by centrifugation. The concentration of melatonin and testosterone were quantified along with the enzymatic activity of glutathione peroxidase (GPx), glutathione reductase (GRD), and catalase (CAT). Correlation analyses between melatonin and testosterone concentrations or enzymatic activity were also performed. Melatonin concentration was affected by season (P < 0.05) but not breed, with lesser concentrations in the first rainy season. Testosterone concentration, however, was affected by breed and season, with greater concentrations (P < 0.01) in the Hampshire and Romney Marsh rams during the second dry season. Regarding antioxidant enzyme activity, there was only seasonal variation in GPx activity (P < 0.05). When correlation analyses were used for data assessments, there was a negative correlation between melatonin and testosterone concentrations in Hampshire rams. In conclusion, melatonin concentrations in seminal plasma of rams that were located in an area with an equatorial photoperiod was affected by the climatological season but there was no positive correlation with testosterone concentration or antioxidant enzyme activity

    Influencia del riego deficitario controlado precosecha sobre la calidad de la cereza ´Prime Giant`

    El uso de redes de sensores para el manejo del riego deficitario controlado (RDC) en cerezo puede incidir en la mejora de la productividad del agua, calidad de la fruta y en su comportamiento poscosecha, al permitir el conocimiento y control del estado hídrico del suelo y árbol en todo momento. En el primer año de ensayo, el riego deficitario aplicado en precosecha para satisfacer el 85% de las necesidades máximas del cultivo (ETcg) permitió un ahorro de agua del 17% (380 m3 ha-1) respecto al tratamiento control, que se regó al 110% de la ETcg. Este déficit hídrico ligero no afectó a la producción (16,13 t ha-1) e incluso mejoró la calidad del fruto. Así, en el momento de la cosecha (t0) los frutos bajo déficit ligero presentaron matices más rojos y mayor acidez que los de riego completo. Esta mayor acidez no afectó al índice de madurez debido a la compensación por sólidos solubles totales (SST). El carácter de mayor acidez perduró tras 30 días de conservación en frio (t1) y 5 días de simulación de las condiciones de comercialización (t2). Igualmente, los frutos bajo déficit hídrico presentaron una tendencia a menores pérdidas de peso por deshidratación a finales de los periodos de conservación en frio y de simulación de la comercialización.Ministerio de Economía y Competitividad AGL2013-49047-C2-1

    NGC 6705 a young α\alpha-enhanced Open Cluster from OCCASO data

    The stellar [α\alpha/Fe] abundance is sometimes used as a proxy for stellar age, following standard chemical evolution models for the Galaxy, as seen by different observational results. In this work we show that the Open Cluster NGC6705/M11 has a significant α\alpha-enhancement [α\alpha/Fe]>0.1>0.1 dex, despite its young age (\sim300 Myr), challenging the current paradigm. We use high resolution (R>65,000>65,000) high signal-to-noise (\sim70) spectra of 8 Red Clump stars, acquired within the OCCASO survey. We determine very accurate chemical abundances of several α\alpha elements, using an equivalent width methodology (Si, Ca and Ti), and spectral synthesis fits (Mg and O). We obtain [Si/Fe]=0.13±0.050.13\pm0.05, [Mg/Fe]=0.14±0.070.14\pm0.07, [O/Fe]=0.17±0.070.17\pm0.07, [Ca/Fe]=0.06±0.050.06\pm0.05 and [Ti/Fe]=0.03±0.030.03\pm0.03. Our results place these cluster within the group of young [α\alpha/Fe]-enhanced field stars recently found by several authors in the literature. The ages of our stars have an uncertainty of around 50 Myr, much more precise than for field stars. By integrating the cluster's orbit in several non-axisymmetric Galactic potentials, we establish the M11's most likely birth radius to lie between 6.8-7.5 kpc from the Galactic center, not far from its current position. With the robust Open Cluster age scale, our results prove that a moderate [α\alpha/Fe]-enhancement is no guarantee for a star to be old, and that not all α\alpha-enhanced stars can be explained with an evolved blue straggler scenario.     Acute unilateral vestibulopathy (AUV) is characterized by acute vertigo, nausea, and imbalance without neurological deficits or auditory symptomatology. Here, we explore the effect of glucocorticoid treatment on the degree of canal paresis in patients with AUV, and critically, establish its relationship with dizziness symptom recovery. We recruited consecutive patients who were retrospectively assigned to one of the two groups according to whether they received glucocorticoid treatment (n = 32) or not (n = 44). All patients underwent pure-tone audiometry, bithermal caloric testing, MRI brain imaging, and were asked to complete a dizziness handicap inventory on admission to hospital and just prior to hospital discharge. In the treatment group, the canal paresis at discharge was significantly lower than in the control group (mean ± SD % 38.04 ± 21.57 versus 82.79 ± 21.51, p < 0.001). We also observed a significant reduction in the intensity of nystagmus in patients receiving glucocorticoid treatment compared to the non-treatment group (p = 0.03). DHI test score was significantly lower at discharge in the treatment group (mean ± SD % 23.15 ± 12.40 versus 64.07 ± 12.87, p < 0.001), as was the length of hospital stay (2.18 ± 1.5 days versus 3.6 ± 1.7 days, p = 0.002). Glucocorticoid treatment leads to acute symptomatic improvement, with a reduced hospital stay and reduction in the intensity of acute nystagmus. Our findings suggest that glucocorticoids may accelerate vestibular compensation via a restoration of peripheral vestibular function, and therefore has important clinical implications for the treatment of AUV

    Vasectomy and photoperiodic regimen modify the protein profile, hormonal content and antioxidant enzymes activity of ram seminal plasma

    This work aimed to determine the contribution of the testis and epididymis and the effect of the photoperiodic regimen on ram seminal plasma (SP). Semen was collected from 15 mature rams located in an equatorial (Colombian Creole and Romney Marsh, eight intact and two vasectomized) or a temperate climate (Rasa Aragonesa, three intact and two vasectomized). SP proteins were analyzed by Bradford, SDS-PAGE and difference gel electrophoresis (DIGE). Melatonin and testosterone concentrations were quantified by ELISA, and activity of glutathione peroxidase (GPx), glutathione reductase (GRD), and catalase by enzymatic assays. Vasectomy increased protein concentration and the intensity of high molecular weight bands (p < 0.001), with no differences between breeds. DIGE revealed the absence of six proteins in vasectomized rams: angiotensin-converting enzyme, lactotransferrin, phosphoglycerate kinase, sorbitol dehydrogenase, epididymal secretory glutathione peroxidase and epididymal secretory protein E1. Vasectomy also decreased melatonin concentrations in seasonal rams, and testosterone in all of them (p < 0.001), but did not affect antioxidant enzyme activity. Equatorial rams showed lower melatonin and testosterone concentration (p < 0.01) and catalase, but higher GPx activity (p < 0.05). In conclusion, vasectomy modifies the protein profile and hormonal content of ram seminal plasma, whereas the exposure to a constant photoperiod affects hormonal concentration and antioxidant enzymes activity

    Sperm survival and heterogeneity are correlated with fertility after intrauterine insemination in superovulated ewes

    Abstract Efficient animal production involves accurate estimations of fertilizing ability. One key factor is the plasma membrane of the sperm cell, which is actively involved in the cascade of events before oocyte fusion. Many methods are used to analyze the characteristics of this membrane, including partition in aqueous two-phase systems which is an efficient method to analyze sperm surface changes accounting for loss of viability and different functional states. Centrifugal countercurrent distribution (CCCD) analysis can also be used in an aqueous two-phase system to determine the relationship between sperm parameters and in vivo fertility in ewes. In a previous work, we found a significant correlation between two post-CCCD parameters (heterogeneity and recovered viability) and field fertility when the same sample was used after cervical AI. The present study was intended to find out whether the control of several external factors that affect reproductive efficiency is able to increase the correlation coefficient between post-CCCD parameters and fertility. Thus, 90 Rasa aragonesa ewes were controlled on the same farm and received intrauterine inseminations using the same technical equipment. The fertilizing ability of the raw semen and sperm samples selected by a dextran/swim-up process was compared using a low number of spermatozoa per insemination (7 Â 10 7 ) to enhance possible fertility differences. A new post-CCCD parameter was considered; the loss of viability (LV) occurred during the CCCD process. This variable denotes the sperm surviving ability and corresponds to the difference between the total number of viable cells loaded and recovered after the CCCD run. The mean fertility of eight sperm control samples was 60% (range: 25-76%), and there was no significant correlation between standard parameters and in vivo fertility. LV ranged from 2 to 69% (average 27%) and was negatively correlated with fertility (r ¼ À0.914, P &lt; 0.01). Ejaculate heterogeneity (H) ranged from 20 to 47% and was positively, but not significantly, correlated with fertility (r ¼ 0.391). A predictive equation for fertility was deduced by multiple analysis with a very high correlation coefficient (r ¼ 0.967), and level of significance (P &lt; 0.005): predictive fertility PF ¼ 52.546 À 0.594 LV þ 0.665 H. The mean fertility of 13 swim-up selected samples was 63% (range: 25-86%). Again, only parameters derived from the CCCD analysis were highly correlated with fertility, especially LV and H (P &lt; 0.05)

    The role of snow in scavenging aerosol particles: A physical-chemical characterization

    [EN] The below cloud scavenging of aerosols by snow has been analysed in León (NW Spain). Six snow events were registered over the course of one year of study. Ultrafine and accumulation aerosol particles were measured using a scanning mobility particle sizer spectrometer, while hydrometeors were characterized using a disdrometer. Furthermore, the chemical composition of the melted snow-water samples (soluble and insoluble fractions) was analysed. The scavenging coefficient (λ) showed a great variability among events. An effective washing of particles was observed during the first 30 min of snowfall. The mean change in the scavenging efficiency (%ΔC) of particle number concentration (PNC) and λ coefficient during this time interval were: i) nucleation mode: 36.3 % and 3.02 · 10−4 s−1; ii) Aitken mode: 30.4 % and 2.37 · 10−4 s−1 and iii) accumulation mode: 22.4 % and 1.77 · 10−4 s−1. The range of particle sizes that is less efficiently scavenged by snowfall was observed between 400 and 600 nm. When analyzing the whole snow event, an increase of PNC was observed. Two possible explanations underlie this behaviour: it could be caused by changes in air masses or by the resuspension of aerosol particles scavenged by snowflakes upon reaching the ground. A clear relationship was observed between Ca2+, SO42− and NO3− concentrations of aerosol particles before the snow event and the concentrations registered in the melted snow-water. The largest and smallest changes in aerosol number concentrations were caused by snowflakes of 3 and 6 mm in diameter, respectively. The particle size distributions (PSD) were fitted to log-normal distributions and the parameters were compared before and after snowfall.S
