    Gas chromatography-mass spectrometry identification of anticancer phytochemicals in Aframomum danielli (LB579)

    Aframomum danielli is one of the African spices used in folklore medicine for the management of several diseases. This study identified the phytochemical components present in the n-hexane seed extract of the A. danielli by gas chromatography-mass spectrometry (GC-MS) analysis and also evaluated the anti-cancer potential of the identified phytochemicals by performing molecular docking against human Vascular Endothelial Growth Factor (VEGF) using Molegro Virtual Docker. The GC-MS analysis identified the presence of phytochemical components caryophyllene (RT: 18.479), humulene (RT: 19.189), 2-butanone (RT: 22.976), benzenesulfonamide (RT: 31.651) and 2-pyridine acetic acid (RT: 32.446). 2-Butanone was the strongest binding ligand (-65.744 kcal/mol) while caryophyllene was the weakest bind lingand (-56.311 kcal/mol). These compounds showed relative strong docking to VEGF with docking energies comparable to an anticancer drug, bevacizumab (-77.883 kcal/mol). This in silico molecular docking study has shown that these phytochemical components could be responsible for anti-cancer properties of A. danielli

    Background Sickle cell disease (SCD) is a genetic disease caused by an individual inheriting an allele for sickle cell hemoglobin from both parents and is associated with unusually large numbers of immature blood cells, containing many long, thin, crescent-shaped erythrocytes. It is a disease prevalent throughout many populations. The use of medicinal plants and nutrition in managing SCD is gaining increasing attention. Methods The antisickling effects of Solenostemon monostachyus (SolMon), Carica papaya seed oil (Cari-oil) and Ipomoea involucrata (Ipocrata) in male (HbSSM) and female (HbSSF) human sickle cell blood was examined in vitro and compared with controls, or cells treated with glutathione or an antisickling plant (Vernonia amygdalina; VerMyg). Results Levels of sickle blood cells were significantly reduced (P < 0.05) in all the plant-extract treated SCD patients’ blood compared with that of untreated SCD patients. RBCs in SolMon, Ipocrata, and Cari-oil treated samples were significantly higher (P < 0.05) compared with VerMyg-treated samples. The Fe2+/Fe3+ ratio was significantly reduced (P < 0.05) in all plant extract-treated HbSSM samples compared with controls. Hemoglobin concentration was significantly increased (P < 0.05) by SolMon treatment in HbSSF compared with VerMyg. Sickle cell polymerization inhibition exhibited by SolMon was significantly higher (P < 0.05) compared with that of VerMyg in HbSSF blood. Sickle cell polymerization inhibition in SolMon and Ipocrata were significantly higher (P < 0.05) compared with VerMyg in HbSSM blood. All plant extracts significantly reduced (P < 0.05) lactate dehydrogenase activity in both HbSSM and HbSSF-treated blood. Catalase activity was significantly increased (P < 0.05) in HbSSF blood treated with Ipocrata compared with glutathione. Cari-oil treated HbSSM and HbSSF blood had significantly increased (P < 0.05) peroxidase activity compared with controls. Conclusions Methanolic extracts from S. monostachyus, C. papaya seed oil and I. involucrata exhibited particular antisickling properties coupled with the potential to reduce stress in sickle cell patients. (4-Hydroxy- 3-methoxyphenyl)ethyl methyl ketone were the strongest binding ligand (-65.744 kcal/mol for VEGF) and (- 99.7836kcal/mol for MMP) while β-Caryophyllene was the weakest binding ligand. These compounds showed relative strong docking to VEGF with docking energies comparable to an anticancer drug, bevacizumab (-77.883kcal/mol for VEGF) and (-109.021kcal/mol for MMP).     Benzyl isothiocyanate (BITC) is toxic in high concentration. The capacity of Aspergillus niger, microwave and ultraviolet radiations to reduce the BITC levels in Carica papaya Linn seed oil were assessed in vitro. BITC at different concentrations were periodically exposed to microwave and ultraviolet radiations for 30 min and 10 h, respectively; and to identify Aspergillus niger for 4 days. Microwave radiation significantly reduced (p < 0.05) BITC levels (0.0272, 0.0544, and 0.0816 μmol) to 12.19, 8.99 and 27.5 % respectively within 15 min. Ultraviolet radiation significantly reduced (p < 0.05) BITC levels at all the concentrations. A. niger significantly increased (p < 0.05) BITC degradation on days 2 and 4 at 0.816, 1.36 and 2.72 nmol. Glutathione activity was significantly increased (p < 0.05) while glutathione S-transferase activity significantly reduced (p < 0.05) at all concentrations on days 3 and 4 respectively. The three techniques are possible models for improving the dietary consumption of the oil

    Dominant modifiable risk factors for stroke in Ghana and Nigeria (SIREN): a case-control study

    Summary: Background: Sub-Saharan Africa has the highest incidence, prevalence, and fatality from stroke globally. Yet, only little information about context-specific risk factors for prioritising interventions to reduce the stroke burden in sub-Saharan Africa is available. We aimed to identify and characterise the effect of the top modifiable risk factors for stroke in sub-Saharan Africa. Methods: The Stroke Investigative Research and Educational Network (SIREN) study is a multicentre, case-control study done at 15 sites in Nigeria and Ghana. Cases were adults (aged ≥18 years) with stroke confirmed by CT or MRI. Controls were age-matched and gender-matched stroke-free adults (aged ≥18 years) recruited from the communities in catchment areas of cases. Comprehensive assessment for vascular, lifestyle, and psychosocial factors was done using standard instruments. We used conditional logistic regression to estimate odds ratios (ORs) and population-attributable risks (PARs) with 95% CIs. Findings: Between Aug 28, 2014, and June 15, 2017, we enrolled 2118 case-control pairs (1192 [56%] men) with mean ages of 59·0 years (SD 13·8) for cases and 57·8 years (13·7) for controls. 1430 (68%) had ischaemic stoke, 682 (32%) had haemorrhagic stroke, and six (<1%) had discrete ischaemic and haemorrhagic lesions. 98·2% (95% CI 97·2–99·0) of adjusted PAR of stroke was associated with 11 potentially modifiable risk factors with ORs and PARs in descending order of PAR of 19·36 (95% CI 12·11–30·93) and 90·8% (95% CI 87·9–93·7) for hypertension, 1·85 (1·44–2·38) and 35·8% (25·3–46·2) for dyslipidaemia, 1·59 (1·19–2·13) and 31·1% (13·3–48·9) for regular meat consumption, 1·48 (1·13–1·94) and 26·5% (12·9–40·2) for elevated waist-to-hip ratio, 2·58 (1·98–3·37) and 22·1% (17·8–26·4) for diabetes, 2·43 (1·81–3·26) and 18·2% (14·1–22·3) for low green leafy vegetable consumption, 1·89 (1·40–2·54) and 11·6% (6·6–16·7) for stress, 2·14 (1·34–3·43) and 5·3% (3·3–7·3) for added salt at the table, 1·65 (1·09–2·49) and 4·3% (0·6–7·9) for cardiac disease, 2·13 (1·12–4·05) and 2·4% (0·7–4·1) for physical inactivity, and 4·42 (1·75–11·16) and 2·3% (1·5–3·1) for current cigarette smoking. Ten of these factors were associated with ischaemic stroke and six with haemorrhagic stroke occurrence. Interpretation: Implementation of interventions targeting these leading risk factors at the population level should substantially curtail the burden of stroke among Africans.
