    Analysis of Flavor Quality of Two Cherry Tomatoes After Refrigeration

    To investigate the differences in the types and relative contents of refrigerated volatile flavor substances between 'Chitose' and 'Ji Tian No.1' cherry tomatoes, the effect of cold storage at 4 ℃ for 16 d on the flavor quality of cherry tomatoes ('Chitose' and 'Ji Tian No.1') was analyzed by using electronic nose and headspace solid-phase microextraction-gas chromatography. The results showed that electronic nose analysis results displayed, the volatile substances that undergo significant changes before and after refrigeration in two cherry tomatoes were inorganic sulfides, nitrogen oxides, and aromatic substances, a total of 93 volatile substances were detected by HS-SPME-GC-MS technique, including 27 aldehydes, 23 alcohols, 4 esters, 11 ketones, 3 furans, 9 alkanes, 12 olefins and 4 other substances. Compared to 0 d, the relative content of total volatile substances in 'Chitose' and 'Ji Tian No.1' after cold storage decreased by 7.08% and 3.68% respectively. The relative content of the main volatile compounds hexanal and trans-2-hexenal in two cherry tomatoes decreased after refrigeration, the relative content of trans 2-pentenal, heptanal, 1-pentanol, 6-methyl-5-hepten-2-one, and 1-penten-3-one increased after refrigeration. Compared with 'Chitose', 'Ji Tian No.1' could retain higher contents of main volatile substances of cherry tomato such as aldehydes, esters and ketones after cold storage. Therefore, 'Ji Tian No.1' was more suitable for post-harvest storage and transportation and low-temperature refrigeration, which would provide a theoretical basis for high-quality cherry tomato post-harvest storage and transportation technology

    Infrastructure provision in backward areas has been proposed by many studies as a cure for widening income gaps in developing countries like China. However, little has been said regarding infrastructure’s role as an unpaid production factor or the issue of optimal resource allocation. This paper examines the efficiency side of infrastructure provision in different Chinese provinces and its spillover effect on the non-infrastructure sector with an adapted dual-sector model. Our findings suggest that while the spillover effect on non-infrastructure sector is considerable across different Chinese regions, non-optimality of resource allocation still exists in regional attempts of developing public facilities.Dual-sector model, Infrastructure, Regional gap, Spillover effect

    Development of a Conventional RT-PCR Assay for Rapid Detection of Porcine Deltacoronavirus with the Same Detection Limit as a SYBR Green-Based Real-Time RT-PCR Assay

    Porcine deltacoronavirus (PDCoV) is a newly discovered coronavirus, which belongs to the family Coronaviridae. It causes watery diarrhea, vomiting, and dehydration in newborn piglets. A sensitive RT-PCR method is urgently required to detect PDCoV infection. In this study, we developed and evaluated a conventional RT-PCR assay and a SYBR green-based real-time RT-PCR assay that targeted the PDCoV n gene. Both assays are specific and have the same limit of detection at 2 × 101 copies of RNA molecules per reaction. Eighty-four clinical samples were subjected to both conventional RT-PCR and real-time RT-PCR, and the same positive rate (41.7%) was achieved, which was much higher than the positive rate (26.2%) using a previously described one-step RT-PCR technique. In summary, a conventional RT-PCR technique was successfully established for the detection of PDCoV with the same detection limit as a SYBR green-based real-time RT-PCR assay

    Vascular neointimal hyperplasia and remodeling arising from local inflammation are characteristic pathogeneses of proliferative cardiovascular diseases, such as atherosclerosis and post angioplasty restenosis. The molecular mechanisms behind these pathological processes have not been fully determined. The adipokine chemerin is associated with obesity, metabolism, and control of inflammation. Recently, chemerin has gained increased attention as it was found to play a critical role in the development of cardiovascular diseases. In this study, we investigated the effects of chemerin on the regulation of vascular smooth muscle cells and carotid neointimal formation after angioplasty. We found that circulating chemerin levels increased after carotid balloon injury, and that knockdown of chemerin significantly inhibited the proliferative aspects of vascular smooth muscle cells induced by platelet-derived growth factor-BB and pro-inflammatory chemokines in vitro as well as prohibited carotid neointimal hyperplasia and pro-inflammatory chemokines in vivo after angioplasty. Additionally, inhibition of chemerin down-regulated the expression of several proteins, including phosphorylated p38 mitogen-activated protein kinase, phosphorylated extracellular signal regulated kinase 1/2, nuclear factor-kappa B p65, and proliferation cell nuclear antigen. The novel finding of this study is that chemerin stimulated vascular smooth muscle cells proliferation and carotid intimal hyperplasia through activation of the mitogen-activated protein kinase signaling pathway, which may lead to vascular inflammation and remodeling, and is relevant to proliferative cardiovascular diseases

    Multicentric hepatic EBV-associated smooth muscle tumors in an AIDS patient: a case report, investigation of mTOR activation and review of the literature

    Epstein-Barr virus (EBV) - associated smooth muscle tumors (EBV-SMT) are a rare, recently recognized distinct group of mesenchymal tumors that develop exclusively in patients with immunosuppression. It is believed that tumorigenesis is, at least in part, through the activation of the Akt/mammalian target of rapamycin (mTOR) signal pathway. We describe the clinicopathologic and immunohistochemical features of a multifocal hepatic EBV-SMT in a 34-year-old acquired immunodeficiency syndrome (AIDS) patient and investigate the activation status of the mTOR signal pathway in this tumor. In addition, we provide a review of the literature on the clinicopathologic findings of hepatic EBV-SMT in adult AIDS patients, and discuss their biologies and possible therapeutic strategies

    Development and evaluation of a broadly reactive reverse transcription recombinase polymerase amplification assay for rapid detection of murine norovirus

    Abstract Background Murine norovirus (MNV) is recognized as the most prevalent viral pathogen in captive mouse colonies. The rapid detection assay for MNV would be a useful tool for monitoring and preventing MNV infection. A recombinase polymerase amplification (RPA) assay was established in this study to provide a solution for rapid and sensitive detection of MNV. Results The detection limit of the RT-RPA assay for the detection of MNV was 1 × 102 copies of RNA molecules per reaction. The assay was specific since there was no cross-reaction with other common murine viruses. In addition, the broad reactivity of the RT-RPA assay was validated using the synthesized template carrying seven point mutations among several MNV strains. The MNV RT-RPA assay could detect as few as 1 × 102 copies of the mutant per reaction, suggesting the assay could be broadly reactive against a large diversity of MNV strains. Forty eight clinical samples including 16 gastric tissue specimens, 16 cecal tissue specimens and 16 fecal specimens were tested for the validation of the new developed RT-RPA assay. The detection results of RT-RPA and RT-qPCR for clinical samples were very similar, except that a gastric tissue sample which was positive by RT-qPCR, with a RNA titer of 27 copies, was negative by RT-RPA. Conclusions A broadly reactive RT-RPA assay was successfully established for MNV detection

    Rapid detection of three rabbit pathogens by use of the Luminex x-TAG assay

    Abstract Background Domestic rabbits especially New Zealand white rabbits play an important role in biological research. The disease surveillance and quality control are essential to guarantee the results of animal experiments performed on rabbits. Rabbit hemorrhagic disease virus, rabbit rotavirus and Sendai virus are the important pathogens that needed to be eliminated. Rapid and sensitive method focus on these three viruses should be established for routine monitoring. The Luminex x-TAG assay based on multiplex PCR and fluorescent microsphere is a fast developing technology applied in high throughput detection. Specific primers modified with oligonucleotide sequence/biotin were used to amplify target fragments. The conjugation between oligonucleotide sequence of the PCR products and the MagPlex-TAG microspheres was specific without any cross-reaction, and the hybridization products could be analyzed using the Luminex 200 analyzer instrument. Recombinant plasmids were constructed to estimate the detection limit of the viruses. Furthermore, 40 clinical samples were used to evaluate the efficiency of this multiplex PCR based Luminex x-TAG assay. Results According to the results, this new method showed high specificity and good stability. Assessed by the recombinant plasmids, the detection limit of three viruses was 100copies/μl. Among 40 clinical specimens, 18 specimens were found positive, which was completely concordant with the conventional PCR method. Conclusions The new developed Luminex x-TAG assay is an accurate, high throughput method for rapid detection of three important viruses of rabbits

    High-Throughput Phenotyping of Cross-Sectional Morphology to Assess Stalk Mechanical Properties in Sorghum

    Lodging is one of the major constraints in attaining high yield in crop production. Major factors associated with stalk lodging involve morphological traits and anatomical features along with the chemical composition of the stem. However, little relevant research has been carried out in sorghum, particularly on the anatomical aspects. In this study, with a high-throughput procedure newly developed by our research group, the nine parameters related to stem regions and vascular bundles were generated in 58 sorghum germplasm accessions grown in two successive seasons. Correlation analysis and principal component analysis were conducted to investigate the relationship between anatomical aspects and stalk mechanical traits (breaking force, stalk strength and lodging index). It was found that most vascular parameters were positively associated with breaking force and lodging index with the correlation coefficient r varying from −0.46 to 0.64, whereas stalk strength was only associated with rind area with the r = 0.38. The germplasm resources can be divided into two contrasting categories (classes I with 23 accessions and II with 30 accessions). Compared to class II, the class I was characterized by a larger number (+40.7%) and bigger vascular bundle (+30%), thicker stem (+19.6%) and thicker rind (+36.0%) but shorter internode (plant) (−91.0%). This study provides the methodology and information for the studies of the stem anatomical parameters in crops and facilitates the selective breeding of sorghum
