    Temporal dynamics of Prochlorococcus cells with the potential for nitrate assimilation in the subtropical Atlantic and Pacific oceans

    Utilization of nitrate as a nitrogen source is broadly conserved among marine phytoplankton, yet many strains of Prochlorococcus lack this trait. Among cultured strains, nitrate assimilation has only been observed within two clades of Prochlorococcus: the high-light adapted HLII clade and the low-light adapted LLI clade. To better understand the frequency and dynamics of nitrate assimilation potential among wild Prochlorococcus, we measured seasonal changes in the abundance of cells containing the nitrate reductase gene (narB) in the subtropical North Atlantic and North Pacific oceans. At the Atlantic station, the proportion of HLII cells containing narB varied with season, with the highest frequency observed in stratified waters during the late summer, when inorganic nitrogen concentrations were lowest. The Pacific station, with more persistent stratification and lower N : P ratios, supported a perennially stable subpopulation of HLII cells containing narB. Approximately 20–50% of HLII cells possessed narB under stratified conditions at both sites. Since HLII cells dominate the total Prochlorococcus population in both ecosystems, nitrate potentially supports a significant fraction of the Prochlorococcus biomass in these waters. The abundance of LLI cells containing narB was positively correlated with nitrite concentrations at the Atlantic station. These data suggest that Prochlorococcus may contribute to the formation of primary nitrite maxima through incomplete nitrate reduction and highlight the potential for interactions between Prochlorococcus and sympatric nitrifying microorganisms.     Investigating Estimation: Influences of Time and Confidence of Urban Middle School Students

    This study sought to determine the estimation processes used by 10 urban middle school students for solving computational estimation problems, and if there was a difference in the estimation processes utilized for straight computation and application problems. An adapted model of the Accessing Computational Estimation Test (ACE) was used to determine the estimation strategies employed by the subjects within timed and un-timed settings. Qualitative methods were used to analyze the data. The timed ACE tests were administered using an interview format and included computation and application problems. The findings indicated that there were no differences in estimation processes for straight computation and application problems; however, students performed better on timed tests for application problems

    The occurrence of ionospheric signatures of plasmaspheric plumes over different longitudinal sectors

    Plasmaspheric plumes have ionospheric signatures and are observed as storm-enhanced density (SED) in global positioning system (GPS) total electron content (TEC). These ionospheric signatures have been primarily observed over the American sector and in a few limited examples over the European sector. This study examines the longitudinal occurrence frequency of plasmaspheric plumes. We analyzed all images from the Imager for Magnetopause-to-Aurora Global Exploration (IMAGE) Extreme Ultraviolet Imager (EUV) databases for the first half of 2001 and identified a total of 31 distinct plume intervals observed during different storm events. Out of the total IMAGE EUV plumes that we identified, 12 were projected over North America, 10 over Asia, and the remaining 9 were over Europe and the Atlantic Ocean. Using ground-based GPS TEC from MIT\u27s Madrigal database, we searched for corresponding SED/TEC plumes at different longitudinal sector and found 12 ionospheric SED plume signatures over North America, 4 over Europe, and 2 over Asia. This indicates that the observation probability of an ionospheric SED plume when a plasmaspheric plume is seen is 100% in the American sector, 50% in the European sector, and 20% in the Asian sector. This could be due to the fact that the plumes may be either positioned beyond the limit of the ground-based GPS field of view, which happens mainly when there is less plasmaspheric erosion, or are too weak to be detected by the sparse number of GPS receivers over Asia. The in situ plasma densities from the available coincident defense metrological satellite program (DMSP) satellites were also used to study the characteristics of SED/TEC plume at DMSP orbiting altitude (i.e., ∼870 km). The TOPographic EXplorer (TOPEX) altimeter TEC also is used to identify the conjugate SED/plume signature over the Southern Hemisphere

    Survey of research approaches utilised in the scholarship of teaching and learning publications

    The Scholarship of Teaching and Learning (SoTL) has been described as the fastest growing academic development movement in higher education. As this field of inquiry matures, there is a need to understand how SoTL research is conducted. The purpose of our study was to inform this debate by investigating research approaches used in SoTL publications. We analysed 223 empirical research studies published from 2012 to 2014 in three explicitly focused SoTL journals. We classified the studies as either qualitative, quantitative, or mixed methods using an analytical framework devised from existing literature on research methods. We found that the use of the three research designs was fairly evenly distributed across the papers examined: qualitative (37.2%), quantitative (29.6%), and mixed methods (33.2%). However, there was an over-reliance on data collection from a single source in 83.9% of papers analysed, and this source was primarily students. There was some, but limited, evidence of the use of triangulation through the use of multiple data collection instruments (e.g. survey, assessment tasks, grade databases). Similarly, only one-third of publications classified as mixed methods integrated the analysis and interpretation of the qualitative and quantitative data equally within the study. We conclude that current SoTL research is characterised by methodological pluralism but could be advanced through inclusion of more diverse approaches, such as close reading, and adoption of strategies known to enhance the quality of research, for example, triangulation and visual representation

    Roadmap Towards Communitywide Intercalibration and Standardization of Ocean Nucleic Acids ‘Omics Measurements

    In January 2020, the US Ocean Carbon & Biogeochemistry (OCB) Project Office funded the Ocean Nucleic Acids 'omics Intercalibration and Standardization workshop held at the University of North Carolina in Chapel Hill. Thirty-two participants from across the US, along with guests from Canada and France, met to develop a framework for standardization and intercalibration (S&I) of ocean nucleic acid ‘omics (na’omics) approaches (i.e., amplicon sequencing, metagenomics and metatranscriptomics). During the three-day workshop, participants discussed numerous topics, including: a) sample biomass collection and nucleic acid preservation for downstream analysis, b) extraction protocols for nucleic acids, c) addition of standard reference material to nucleic acid isolation protocols, d) isolation methods unique to RNA, e) sequence library construction, and f ) integration of bioinformatic considerations.     Investigating the physical properties of transiting hot Jupiters with the 1.5-m Kuiper Telescope

    We present new photometric data of 11 hot Jupiter transiting exoplanets (CoRoT-12b, HAT-P-5b, HAT-P-12b, HAT-P-33b, HAT-P-37b, WASP-2b, WASP-24b, WASP-60b, WASP-80b, WASP-103b, XO-3b) in order to update their planetary parameters and to constrain information about their atmospheres. These observations of CoRoT-12b, HAT-P-37b and WASP-60b are the first follow-up data since their discovery. Additionally, the first near-UV transits of WASP-80b and WASP-103b are presented. We compare the results of our analysis with previous work to search for transit timing variations (TTVs) and a wavelength dependence in the transit depth. TTVs may be evidence of a third body in the system and variations in planetary radius with wavelength can help constrain the properties of the exoplanet's atmosphere. For WASP-103b and XO-3b, we find a possible variation in the transit depths that may be evidence of scattering in their atmospheres. The B-band transit depth of HAT-P-37b is found to be smaller than its near-IR transit depth and such a variation may indicate TiO/VO absorption. These variations are detected from 2-4.6σ\sigma, so follow-up observations are needed to confirm these results. Additionally, a flat spectrum across optical wavelengths is found for 5 of the planets (HAT-P-5b, HAT-P-12b, WASP-2b, WASP-24b, WASP-80b), suggestive that clouds may be present in their atmospheres. We calculate a refined orbital period and ephemeris for all the targets, which will help with future observations. No TTVs are seen in our analysis with the exception of WASP-80b and follow-up observations are needed to confirm this possible detection.Comment: 18 pages, 7 figures, 9 Tables.
