    Fundamentals of Earth Observation Policy: Examples for German and European Missions

    Several European countries have developed their national high resolution earth observation systems. Some of them are operated in close cooperation with industrial partners, others are dual-use missions earmarked to fulfil the needs of national security. In addition, the European Space Agency and the European Commission have initiated the Global Monitoring for Environment and Security (GMES) project. Therein, a fleet of satellites (SENTINELs) will deliver data for European wide information services, augmented by data from national and non-European earth observation systems. This new scenario needs clear guidance and regulations. Besides the principles for operations of earth observation missions – as set out in UN principles on earth observation – the operators of very high resolution missions require clear governmental acts which international users can be served and which data might be restricted in distribution. For national science and the SENTINEL-missions, a policy for free and open access is being developed to guarantee a maximum use of the data. Exemplified on the German national missions and the European GMES scenario, data policies and regulations for existing and new earth observation missions will be explained

    Lunar Rover with Multiple Science Handling Capability

    A rover design study was undertaken for exploration of the Moon. Rovers that have been launched in the past carried a suite of science payload either onboard its body or on the robotic arm’s end. No rover has so far been launched and tasked with “carrying and deploying” a payload on an extraterrestrial surface. This paper describes a lunar rover designed for deploying payload as well as carrying a suite of instruments onboard for conventional science tasks. The main consideration during the rover design process was the usage of existing, in-house technology for development of some rover systems. The manipulation subsystem design was derived from the technology of Light Weight Robot, a dexterous arm originally developed for terrestrial applications. Recent efforts have led to definition of a mission architecture for exploration of the Moon with such a rover.     Directed evolution of Vibrio fischeri LuxR for improved response to butanoyl-homoserine lactone

    LuxR is the 3-oxohexanoyl-homoserine lactone (3OC6HSL) dependent transcriptional activator of the prototypical acyl-homoserine lactone (AHL) quorum sensing system of Vibrio fischeri. Wild-type LuxR exhibits no response to butanoyl-HSL (C4HSL) in quantitative bioassays at concentrations of up to 1 µM; a previously described LuxR variant (LuxR-G2E) exhibits a broadened response to diverse AHLs, including pentanoyl-HSL (C5HSL), but not to C4HSL. Here, two rounds of directed evolution of LuxR-G2E generated variants of LuxR that responded to C4HSL at concentrations as low as 10 nM. One variant, LuxR-G4E, had only one change, I45F, relative to the parent LuxR-G2E, which itself differs from wild-type at three residues. Dissection of the four mutations within LuxR-G4E demonstrated that at least three of these changes were simultaneously required to achieve any measurable C4HSL response. The four changes improved both sensitivity and specificity towards C4HSL relative to any of the other 14 possible combinations of those residues. These data confirm that LuxR is evolutionarily pliable and suggest that LuxR is not intrinsically asymmetric in its response to quorum sensing signals with different acyl-side chain lengths

    Compositional Strategies for Large-Scale Works Around 1600

    Giovanni Gabrieli’s posthumously published motets In ecclesiis and Benedictus es, Dominus (Sacrae symphoniae ... liber secundus, 1615) show obvious characteristics of self-quotation and reworking. An analysis of the works focuses on a characteristic ascending-fifths sequence which has been called “Monte Romanesca” by Robert Gjerdingen who introduced the term for the eighteenth century. In this study, the term is expanded to include music from the end of the sixteenth century for the first time. Further analysis concentrates on motivic and modal connections as well as special harmonic features such as chromatic-third relationships. Additionally, several other works of the composer are considered, showing that Gabrieli built on highly sophisticated harmonic, motivic, and formal compositional strategies in his large-scale oeuvre. Also, apparent processes of imitation in the Missa sine nomine of Gabrieli’s pupil Alessandro Tadei suggest that the younger composer used either or both of his teacher's motets as a model. A comparative analysis of all three works provides further insight into compositional methods at the “dawn of the Baroque” era and sheds new light on what must have been a special teacher-student relationship

    3D cell cultures, as a surrogate for animal models, enhance the diagnostic value of preclinical in vitro investigations by adding information on the tumour microenvironment : a comparative study of new dual-mode HDAC inhibitors

    Anchorage-independent 3D-cultures of multicellular tumour spheroids (MCTS) and in vitro microtumours of cancer cells can provide upfront information on the effects of anticancer drug candidates, tantamount to that obtained from animal xenograft studies. Unlike 2D cancer cell cultures, 3D-models take into account the influence of the tumour microenvironment and the location dependence of drug effects and accumulation. We exemplified this by comparison of the effects of two new dual-mode anticancer agents, Troxbam and Troxham, and their monomodal congeners SAHA (suberoylanilide hydroxamic acid) and CA-4 (combretastatin A-4). We assessed the growth of MCTS of HCT116(wt) human colon carcinoma cells exposed to these compounds, as well as the spatial distribution of dead HCT116(wt) cells in these MCTS. Also, fluorescence imaging of live and fixed MCTS was used to assess the type of cellular death induced by test compounds. Furthermore, an innovative perfusion bioreactor system was used to grow microtumours in the presence or absence of test compounds. Both new investigational compounds led to significant reductions of the size of such MCTS and also of corresponding in vitro microtumours by inducing caspase-9 dependent apoptosis and elevated levels of reactive oxygen species. 3D multicellular tumour spheroids are easy to grow and employ for compound tests in the familiar well-plate set-up. Together with 3D microtumours grown at scaffolds in continuously perfused bioreactors they allow to study, early on in the course of drug evaluations, the communication of tumour cells with their microenvironment to an extent hitherto available only in animal experiments.     Stake Shift in Major Cryptocurrencies: An Empirical Study

    In the proof-of-stake (PoS) paradigm for maintaining decentralized, permissionless cryptocurrencies, Sybil attacks are prevented by basing the distribution of roles in the protocol execution on the stake distribution recorded in the ledger itself. However, for various reasons this distribution cannot be completely up-to-date, introducing a gap between the present stake distribution, which determines the parties' current incentives, and the one used by the protocol. In this paper, we investigate this issue, and empirically quantify its effects. We survey existing provably secure PoS proposals to observe that the above time gap between the two stake distributions, which we call stake distribution lag, amounts to several days for each of these protocols. Based on this, we investigate the ledgers of four major cryptocurrencies (Bitcoin, Bitcoin Cash, Litecoin and Zcash) and compute the average stake shift (the statistical distance of the two distributions) for each value of stake distribution lag between 1 and 14 days, as well as related statistics. We also empirically quantify the sublinear growth of stake shift with the length of the considered lag interval. Finally, we turn our attention to unusual stake-shift spikes in these currencies: we observe that hard forks trigger major stake shifts and that single real-world actors, mostly exchanges, account for major stake shifts in established cryptocurrency ecosystems.Comment: 20 pages, 8 figures, 2 tables, paper accepted for publication at Financial Cryptography and Data Security 2020 (FC 2020, see https://fc20.ifca.ai
