Drosophila olfactory receptors as classifiers for volatiles from disparate real world applications
Olfactory receptors evolved to provide animals with ecologically and behaviourally relevant information. The resulting extreme sensitivity and discrimination has proven useful to humans, who have therefore co-opted some animals' sense of smell. One aim of machine olfaction research is to replace the use of animal noses and one avenue of such research aims to incorporate olfactory receptors into artificial noses. Here, we investigate how well the olfactory receptors of the fruit fly, Drosophila melanogaster, perform in classifying volatile odourants that they would not normally encounter. We collected a large number of in vivo recordings from individual Drosophila olfactory receptor neurons in response to an ecologically relevant set of 36 chemicals related to wine ('wine set') and an ecologically irrelevant set of 35 chemicals related to chemical hazards ('industrial set'), each chemical at a single concentration. Resampled response sets were used to classify the chemicals against all others within each set, using a standard linear support vector machine classifier and a wrapper approach. Drosophila receptors appear highly capable of distinguishing chemicals that they have not evolved to process. In contrast to previous work with metal oxide sensors, Drosophila receptors achieved the best recognition accuracy if the outputs of all 20 receptor types were used
Prescription Patterns, Recurrence, and Toxicity Rates of Adjuvant Treatment for Stage III/IV Melanoma-A Real World Single-Center Analysis.
Approved adjuvant treatment options for stage III melanoma are the immune checkpoint inhibitors (ICI) pembrolizumab and nivolumab, and in presence of a BRAF V600E/K mutation additionally dabrafenib in combination with trametinib (BRAFi/MEKi). This study aims to describe prescription patterns and recurrence and toxicity rates of adjuvant-treated melanoma patients from the Cancer Center of the University Hospital Bern, Switzerland. One hundred and nine patients with an indication for adjuvant treatment were identified. Five (4.6%) had contraindications and, as such, were not proposed any adjuvant treatment, while 10 patients (9.2%) declined treatment. BRAF status was known for 91 (83.5%) patients. Of 40 (36.7%) patients with BRAF V600E/K melanoma, pembrolizumab was prescribed to 18 (45.0%), nivolumab to 16 (40.0%), and dabrafenib/trametinib to three (7.5%) patients. Grade 3-4 toxicity was reported in 18.9% and 16.7% of all the patients treated with pembrolizumab and nivolumab, respectively. No toxicities were observed for dabrafenib/trametinib. Thirty-eight percent of the patients treated with pembrolizumab and 40.0% of those treated with nivolumab relapsed. No relapses were reported for dabrafenib/trametinib. Prescription patterns indicate a clear preference for adjuvant ICI treatment
Impact of hypnosis on psychophysiological measures: A scoping literature review.
Exploring psychophysiological changes during hypnosis can help to better understand the nature and extent of the hypnotic phenomenon by characterizing its influence on the autonomic nervous system (ANS), in addition to its central brain effects. Hypnosis is thought to induce a relaxation response, yet studies using objective psychophysiological measures alongside hypnosis protocols show various results. We review this literature and clarify the effects of hypnosis on psychophysiological indices of ANS activity and more specifically of the stress/relaxation response, such as heart rate variability and electrodermal activity. Studies reporting psychophysical measures during hypnosis were identified by a series of Pubmed searches. Data was extracted with an interest for the influence of hypnotizability and effects of specific suggestions or tasks on the findings. We found 49 studies comprising 1315 participants, 45 concerning healthy volunteers and only 4 on patients. Sixteen compared high vs. low hypnotizable people; 30 measured heart rate, 18 measured heart rate variability, 25 electrodermal activity, and 23 respiratory signals as well as other physiological parameters. Globally, results converge to show reductions in sympathetic responses and/or increases in parasympathetic tone under hypnosis. Several methodological limitations are underscored, such as older studies (N = 16) using manual analyses, small sample sizes (<30, N = 31), as well as uncontrolled multiple comparisons. Nevertheless, we confirm that hypnosis leads to a physiological relaxation response and highlight promising avenues for this research. Suggestions are made for guiding future work in this field
Toxicity, disease management and outcome of treatment with immune checkpoint inhibitors by sex in patients with cancer and preexisting autoimmune disease.
Female sex is associated with a higher risk for autoimmune diseases (ADs) and immune-related adverse events (irAEs) from immune checkpoint inhibitors (ICIs). While the safety of ICIs in AD cohorts has been reported, sex-segregated data on patient characteristics and outcomes are lacking. In the present study, the disease and treatment characteristics of 51 patients with cancer and preexisting AD (PAD) treated with ICIs at Bern University Hospital Cancer Center (Bern, Switzerland) between January 2017 and June 2021 were analyzed by sex. Rheumatic (n=12/27, 44.4%) and endocrine (n=11/24, 45.8%) PADs were most common among male and female patients, respectively. At the time of ICI initiation, 29.6% (n=8/27) of male and 20.8% (n=5/24) of female patients received immunosuppression for their PAD. Female patients were more likely to experience an irAE (58.3 vs. 48.1%), and less likely to encounter an exacerbation of their PAD (38.5 vs. 14.3%) compared with male patients. Multiple-site irAEs (46.2 vs. 21.4%), implication of an organ specialist for irAEs (100.0 vs. 57.1%) and use of additional immunosuppressive drugs (38.4 vs. 7.7%) were more common in male patients. IrAEs were resolved and ICIs were discontinued in 69.2% (n=9/13) and 71.4% (n=10/14) of the total male and female patients, respectively. Median progression-free survival was higher in male than female patients with irAEs (19.9 vs. 10.7 months) and without irAEs (4.4 vs. 1.8 months). The median overall survival time was higher in male than female patients with irAEs (not estimable vs. 22.5 months) and without irAEs (10.1 vs. 7.4 months). Taken together, these results suggested that sex-related differences existed regarding the clinical presentation of irAEs and treatment outcome
The Role of Immune Checkpoint Inhibitors in Metastatic Pancreatic Cancer: Current State and Outlook.
Pancreatic ductal adenocarcinoma (PDAC) is one of the deadliest tumors, characterized by its aggressive tumor biology and poor prognosis. While immune checkpoint inhibitors (ICIs) play a major part in the treatment algorithm of various solid tumors, there is still no evidence of clinical benefit from ICI in patients with metastatic PDAC (mPDAC). This might be due to several reasons, such as the inherent low immunogenicity of pancreatic cancer, the dense stroma-rich tumor microenvironment that precludes an efficient migration of antitumoral effector T cells to the cancer cells, and the increased proportion of immunosuppressive immune cells, such as regulatory T cells (Tregs), cancer-associated fibroblasts (CAFs), and myeloid-derived suppressor cells (MDSCs), facilitating tumor growth and invasion. In this review, we provide an overview of the current state of ICIs in mPDAC, report on the biological rationale to implement ICIs into the treatment strategy of pancreatic cancer, and discuss preclinical studies and clinical trials in this field. Additionally, we shed light on the challenges of implementing ICIs into the treatment strategy of PDAC and discuss potential future directions
Universality of the helimagnetic transition in cubic chiral magnets: Small angle neutron scattering and neutron spin echo spectroscopy studies of FeCoSi
We present a comprehensive Small Angle Neutron Scattering (SANS) and Neutron
Spin Echo Spectroscopy (NSE) study of the structural and dynamical aspects of
the helimagnetic transition in FeCoSi with = 0.30. In contrast
to the sharp transition observed in the archetype chiral magnet MnSi, the
transition in FeCoSi is gradual and long-range helimagnetic
ordering coexists with short-range correlations over a wide temperature range.
The dynamics are more complex than in MnSi and involve long relaxation times
with a stretched exponential relaxation which persists even under magnetic
field. These results in conjunction with an analysis of the hierarchy of the
relevant length scales show that the helimagnetic transition in
FeCoSi differs substantially from the transition in MnSi and
question the validity of a universal approach to the helimagnetic transition in
chiral magnets
Strong HI Lyman- variations from the 11 Gyr-old host star Kepler-444: a planetary origin ?
Kepler-444 provides a unique opportunity to probe the atmospheric composition
and evolution of a compact system of exoplanets smaller than the Earth. Five
planets transit this bright K star at close orbital distances, but they are too
small for their putative lower atmosphere to be probed at optical/infrared
wavelengths. We used the Space Telescope Imaging Spectrograph instrument
onboard the Hubble Space Telescope to search for the signature of the planet's
upper atmospheres at six independent epochs in the Ly- line. We detect
significant flux variations during the transits of both Kepler-444e and f
(~20%), and also at a time when none of the known planets was transiting
(~40%). Variability in the transition region and corona of the host star might
be the source of these variations. Yet, their amplitude over short time scales
(~2-3 hours) is surprisingly strong for this old (11.2+-1.0Gyr) and apparently
quiet main-sequence star. Alternatively, we show that the in-transits
variations could be explained by absorption from neutral hydrogen exospheres
trailing the two outer planets (Kepler-444e and f). They would have to contain
substantial amounts of water to replenish such hydrogen exospheres, which would
reveal them as the first confirmed ocean-planets. The out-of-transit
variations, however, would require the presence of a yet-undetected Kepler-444g
at larger orbital distance, casting doubt on the planetary origin scenario.
Using HARPS-N observations in the sodium doublet, we derived the properties of
two Interstellar Medium clouds along the line-of-sight toward Kepler-444. This
allowed us to reconstruct the stellar Ly- line profile and to estimate
the XUV irradiation from the star, which would still allow for a moderate mass
loss from the outer planets after 11.2Gyr. Follow-up of the system at XUV
wavelengths will be required to assess this tantalizing possibility.Comment: Accepted for publication in A&A Name of the system added to the title

Physical Activity and Hippocampal Sub-Region Structure in Older Adults with Memory Complaints.
BackgroundPhysical activity (PA) plays a major role in maintaining cognition in older adults. PA has been shown to be correlated with total hippocampal volume, a memory-critical region within the medial temporal lobe (MTL). However, research on associations between PA and MTL sub-region integrity is limited.ObjectiveTo examine the relationship between PA, MTL thickness, and its sub-regions, and cognitive function in non-demented older adults with memory complaints.MethodsTwenty-nine subjects aged â„60 years, with memory complaints were recruited for this cross-sectional study. PA was tracked for 7 days using accelerometers, and average number of steps/day determined. Subjects were categorized into two groups: those who walked â€4000 steps/day (lower PA) and those with >4000 steps/day (higher PA). Subjects received neuropsychological testing and 3T MRI scans. Nonparametric ANCOVAs controlling for age examined differences between the two groups.ResultsTwenty-six subjects aged 72.7(8.1) years completed the study. The higher PA group (nâ=â13) had thicker fusiform gyrus (median differenceâ=â0.11âmm, effect size (ES)â=â1.43, pâ=â0.001) and parahippocampal cortex (median differenceâ=â0.12âmm, ESâ=â0.93, pâ=â0.04) compared to the lower PA group. The higher PA group also exhibited superior performance in attention and information-processing speed (median differenceâ=â0.90, ESâ=â1.61, pâ=â0.003) and executive functioning (median differenceâ=â0.97, ESâ=â1.24, pâ=â0.05). Memory recall was not significantly different between the two groups.ConclusionOlder non-demented individuals complaining of memory loss who walked >4000 steps each day had thicker MTL sub-regions and better cognitive functioning than those who walked â€4000 steps. Future studies should include longitudinal analyses and explore mechanisms mediating hippocampal related atrophy
