    Castanea sativa Bur: An undervalued by-product but a promising cosmetic ingredient

    Castanea sativa fruit processing generates high amounts of by-products, mostly bur. Currently, the cosmetic industry has a great interest in natural extracts as antioxidant sources. In the present study, C. sativa bur extract was used as the active ingredient, in different amounts, in topical hydrogels. The formulations were characterized regarding total phenolic and flavonoid contents (TPC and TFC, respectively), antioxidant activity (DPPH radical scavenging capacity and ferric reducing antioxidant power (FRAP)) and technological and microbiological properties. The same parameters were evaluated after 30 days of storage at 4 °C (T 30/4 °C) and 20 °C (T 30/20 °C). At time 0 (T 0 ), the TPC ranged between 0.79 and 9.65 mg of gallic acid equivalents (GAE)/g gel, while TFC varied from 0.05 to 1.23 mg of catechin equivalents (CAE)/g gel. Antioxidant activity was high for both assays, with values at T0 ranging between 98.41 and 1013.43 mol of ferrous sulphate equivalents (FSE)/g gel and varying between 431.96 and 990.84 g of Trolox equivalents (TE)/g gel for FRAP and DPPH assays, respectively. No formulation exceeded the defined criteria in microbiological counts. All formulations showed similar technological profiles but particular attention should be given to pH.     Are coffee silverskin extracts safe for topical use? An in vitro and in vivo approach

    Recent changes in regulatory requirements and social views on animal testing have incremented the development of reliable alternative tests for predicting skin and ocular irritation potential of products based on new raw materials. In this regard, botanical ingredients used in cosmetic products are among those materials, and should be carefully reviewed concerning the potential presence of irritant constituents. In particular, cosmetic products used on the face, in vicinity of the eyes or that may come in contact with mucous membranes, should avoid botanical ingredients that contain, or are suspected to contain, such ingredients. In this study, we aimed to evaluate the effect of a new cosmetic ingredient, namely, coffee silverskin (CS), with an in vitro skin and ocular irritation assay using reconstructed human epidermis, EpiSkinâ„¢, and human corneal epithelial model, SkinEthicsâ„¢ HCE, and an in vivo assay. Three different extracts of CS were evaluated. The histology of the models after extracts applications was analysed. The in vitro results demonstrated that extracts were not classified as irritant and the histological analyses proved that extracts did not affect both models structure. The content of caffeine, 5-hydroxymethyl furfural and chlorogenic acid was quantified after the epidermal assay. The in vivo test carried out with the most promising extract (hydroalcoholic) showed that, with respect to irritant effects, these extracts can be regarded as safe for topical application

    Phenylketonuria: Protein content and amino acids profile of dishes for phenylketonuric patients. The relevance of phenylalanine

    Phenylketonuria is an inborn error of metabolism, involving, in most cases, a deficient activity of phenylalanine hydroxylase. Neonatal diagnosis and a prompt special diet (low phenylalanine and natural-protein restricted diets) are essential to the treatment. The lack of data concerning phenylalanine contents of processed foodstuffs is an additional limitation for an already very restrictive diet. Our goals were to quantify protein (Kjeldahl method) and amino acid (18) content (HPLC/fluorescence) in 16 dishes specifically conceived for phenylketonuric patients, and compare the most relevant results with those of several international food composition databases. As might be expected, all the meals contained low protein levels (0.67–3.15 g/100 g) with the highest ones occurring in boiled rice and potatoes. These foods also contained the highest amounts of phenylalanine (158.51 and 62.65 mg/100 g, respectively). In contrast to the other amino acids, it was possible to predict phenylalanine content based on protein alone. Slight deviations were observed when comparing results with the different food composition databases
