Effects of Piptoporus betulinus ethanolic extract on the proliferation and viability of melanoma cells and models of their cell membranes
Piptoporus betulinus is a fungus known for its medicinal properties. It possesses antimicrobial, anti-inflammatory, and anti-cancer activity. In this study, several tests were performed to evaluate the cytotoxic effect of the ethanolic extract of Piptoporus betulinus on two melanoma human cell lines, WM115 primary and A375 metastatic cell lines, as well as Hs27 human skin fibroblasts. The extract proved to affect cancer cells in a dose-dependent manner, and at the same time showed a low cytotoxicity towards the normal cells. The total phenolic content (TPC) was determined spectrophotometrically by the Folin-Ciocalteu method (F-C), and the potential antioxidant activity was measured by ferric-reducing antioxidant power (FRAP) assay. One of the active compounds in the extract is betulin. It was isolated and then its cytotoxic activity was compared to the results obtained from the Piptoporus betulinus extract. To further understand the mechanism of action of the extract’s anticancer activity, tests on model cell membranes were conducted. A model membrane of a melanoma cell was designed and consisted of 1,2-dimyristoyl-sn-glycero-3-phosphocholine, disialoganglioside-GD(1a) and cholesterol: DMPC:GD(1a):chol (5:2:3 mole ratio). Changes in a Langmuir monolayer were observed and described based on Π-A(mol) isotherm and compressibility modulus changes. LB lipid bilayers were deposited on a hydrophilic gold substrate and analyzed by IR and X-ray photoelectron spectroscopy. Our study provides new data on the effect of Piptoporus betulinus extract on melanoma cells and its impact on the model of melanoma plasma membranes
Zastosowanie metody Warda do oceny poziomu bezrobocia w ujęciu przestrzennym
One of the largest economic and social problems of modern world is phenomenon of unemployment. It appeared together with development of free market economy and is integrally bounded with technological progress, consequently which is to replace work of human hands with work of machines. In this paper is undertaken an effort to characterise population of unemployed in powiats of the Małopolskie voivodeship in spatial sense. To separate concentrations of objects it was used agglomeration method of grouping – Ward’s method. Function of criterion of joining of units into groups is sum of squares of distances of particular units from groups’ centres of gravity, of which they belong to. Unemployment rate in powiats of the Małopolskie voivodeship showed large spatial diversification, varying between 16,4% in dąbrowski powiat and 5,6% in bocheński powiat in year 2008. Bocheński powiat was distinguished by the highest interest of unemployed women and people with University education, against the background of remaining powiats of the Małopolskie voivodeship. Separation of group of powiats in voivodeship characterizing by similar scale and intensity of certain problems, occurring on local labour market, indicates possibility of exchanging experience, cooperation between institutions servicing labour markets and local authorities. Despite changes, which have taken place over a span of years, still most endangered with unemployment in powiats of the Małopolskie voivodeship are: women, young people (under 24 years old) and people with low education
The studies on waste biodegradation by Tenebrio molitor
As cities are growing in size with a rise in the population, the amount of plastic waste generated is increasing and becoming unmanageable. The treatment and disposal of plastic waste is an urgent need of our present and future. It has been proved recently that mealworms, the larvae of Tenebrio molitor Linnaeus, are able eat styrofoam, a common polystyrene product. Polystyrene is one of the most widely used plastics, the scale of its production being several million tons per year. Tenebrio molitor is one of the largest pests found in stored-grain products. The insect is indigenous to Europe, but is currently cosmopolitan in distribution. The styrofoam is efficiently degraded in the larval gut by microorganisms. We have used the larvae of T. molitor to biodegrade three types of food packaging plastics: polystyrene (PS), polyvinyl chloride (PVC) and polylactide (PLA). PVC is a thermoplastic made of 57% chlorine (derived from industrial grade salt) and 43% carbon (derived predominantly from oil /gas via ethylene). It is the world's third-most widely produced synthetic plastic polymer, which is not biodegradable easily. On the other hand, PLA is an easily biodegradable and bioactive thermoplastic aliphatic polyester derived from corn and tapioca starch or sugarcane. Three groups of larvae were fed selected types of polymers as an only food, while a control population was fed on oatmeal. The mass loss, dry matter content and biochemical composition of mealworms were assessed in the performed laboratory experiments. The protein concentration in homogenates of the larvae was determined by the Bradford method. To determine the level of hydrolized carbohydrates we used anthrone method. The classical sulfo-phospho-vanillin assay (SPVA) was used to quantitate total lipids in mealworms. The results allowed to compare the decomposition efficiency of selected polymer materials by mealworms and to recognize the mechanism of decomposition contributing to the future use of these animals for the treatment and disposal of plastic waste

We tested: raw material (PS), processed polystyrene (PSr), building insulation material (EPS) and food packaging boxes (PSp). Materials were characterized by means melt flow ratio (MFR), shore hardness and gloss. The biochemical assessment of macromolecules (proteins, lipids and sugars) in the mealworms organisms fed with tested forms of polystyrene allowed us to set how efficient and beneficial the biodegradation of types of polystyrene is. We also evaluated the variability of bacterial community in larval guts by the use of denaturing gradient gel electrophoresis (DGGE) on the bacterial DNA of 16S rRNA genes amplified in polymerase chain reaction (PCR). The results suggest that EPS and PSp polystyrene are the most digestible for T. molitor larvae. The metabolic degradation of polystyrene is probably strictly connected with the changes in biodiversity of gut bacteria

Cases of fatal electrocution due to contact between carbon fibre fishing rods and overhead power lines
Abstract Background Electrocution caused by electricity conducted via overhead power lines carries a high risk to health and life. With the introduction and proliferation of conductive carbon fibre fishing tackle, severe and fatal electrocution occurs due to accidental contact with overhead transmission lines. Case presentation The paper presents three cases of men who died due to electrocution from a conductive fishing rod, which occurred in the same fishery over several years. The deaths occurred on the spot in two cases despite rapid rescue efforts. One of the men died in the hospital the following day due to developing severe complications. Additionally, all of the victims were under the influence of alcohol at the time of the incident. During postmortem diagnosis, skin lesions were identified as signs of electric shock on the hands and feet in each case. To the best of the authors’ knowledge, there is no similar case series available in the literature where several almost identical fatal electrocutions occurred in the same place as a result of contact of a fishing rod with overhead power lines. Conclusions Action is needed to avoid similar incidents and to increase safety in the vicinity of power lines
Prognostic Value of the Modified Atherogenic Index of Plasma during Body Mass Reduction in Polish Obese/Overweight People
Although weight loss is recommended for obese patients, it remains questionable how much weight loss is optimal. A novel index that accurately determines the risk of cardiovascular diseases (CVDs) in terms of weight loss is needed. The modified Atherogenic Index of Plasma (AIP), presented here is unique in the literature. It is calculated based on data for anti-atherogenic, high-density lipoprotein cholesterol (HDL-C) fractions, instead of the total HDL-C. This study investigates whether weight loss correlates with CVD risk, and whether the modified AIP allows more accurate diagnostics in obese/overweight people. According to the increase or decrease of AIP during weight loss, 52 Polish patients were subdivided into two groups: group I (increased AIP; n = 16) and group II (decreased AIP; n = 36). The patients’ body mass composition and fasting serum lipid parameters (total cholesterol, triglycerides, HDL-C, and LDL-C (low-density lipoprotein cholesterol)), and cholesterol in 21 lipoprotein sub-fractions were determined. Over six months, all patients reduced their body mass by about 10%. There were no significant differences in anthropometric measures between groups. Increases in large and intermediate HDL-C fractions 1 to 6 and decreases in smaller fractions 7 to 10 were observed in group II. In group I, HDL-C fractions 1 and 10 decreased, while cholesterol in other fractions increased. Increases were observed in the antiatherogenic HDL-C of 52% of group II and 4% of group I. As for atherogenic HDL-C, a decrease of 24% was observed in group II and an increase of 9% in group I. In group I, increases of very-low-density lipoprotein (VLDL), intermediate-density lipoprotein (IDL), and large LDL fractions were noticed, and the reverse in group II. The results show that the modified AIP is a more accurate indicator of CVD risk than existing indices, and that uncontrolled weight reduction does not necessarily have a beneficial influence, and may adversely affect the cardiovascular system
