    Repression of osteoblast maturation by ERRalpha accounts for bone loss induced by estrogen deficiency

    ERRalpha is an orphan member of the nuclear receptor family, the complete inactivation of which confers resistance to bone loss induced by ageing and estrogen withdrawal to female mice in correlation with increased bone formation in vivo. Furthermore ERRalpha negatively regulates the commitment of mesenchymal cells to the osteoblast lineage ex vivo as well as later steps of osteoblast maturation. We searched to determine whether the activities of ERRalpha on osteoblast maturation are responsible for one or both types of in vivo induced bone loss. To this end we have generated conditional knock out mice in which the receptor is normally present during early osteoblast differentiation but inactivated upon osteoblast maturation. Bone ageing in these animals was similar to that observed for control animals. In contrast conditional ERRalphaKO mice were completely resistant to bone loss induced by ovariectomy. We conclude that the late (maturation), but not early (commitment), negative effects of ERRalpha on the osteoblast lineage contribute to the reduced bone mineral density observed upon estrogen deficiency

    Formation sur le repérage et la prise en charge des victimes de violences conjugales : Impact sur la pratique des internes de médecine générale

    Médecine généraleIntroduction : les violences conjugales sont peu dépistées en médecine générale. La mise en place d'une formation à ce sujet au cours de l'Internat de Médecine Générale permet-elle d'améliorer ce dépistage chez les professionnels concernés ? Matériel et Méthode : une étude observationnelle prospective monocentrique avant/après a été réalisée de novembre 2017 à mars 2019 auprès des Internes en Médecine Générale (IMG) de la Faculté de Médecine de Strasbourg inscrits à la formation. Les participants ont été invités par mail à répondre à un premier questionnaire avant la formation, puis à un second deux mois après la formation. Résultats : 66 IMG étaient inscrits à la formation. Parmi ceux-ci, 52 ont répondu au premier questionnaire, et 47 ont répondu au deuxième questionnaire. La formation a permis la mise en place d'un dépistage systématique des violences lors d'une première consultation chez 42% (n=15) des participants, alors qu'aucun (n=44) ne le faisaient avant, p=0,0005. Conclusion : la formation des IMG au repérage et à la prise en charge des victimes de violences conjugales permet une réelle amélioration des pratiques, et lève un certain nombre de freins empêchant d'aborder le sujet en consultation. Le développement de ce type de formation dans les différentes facultés de médecine semble important pour améliorer la prise en charge des victimes de violences conjugales

    <p>The aim of this work is to study the regulation of intratesticular during spermatogenesis ex vivo. To highlight the progress of spermatogenesis ex vivo, we developed two cell culture systems of seminiferous tubules to study the role of local factors that control the proliferation and differentiation of male germ cells. Our studies are based on two main techniques: RT-PCR and RNA extraction to examine changes in the expression of some growth factors in the culture of seminiferous tubules as the SCF, c- Kit and TGFĂź. The results show, using RT-PCR, that expression of SCF, c-Kit and TGFb is probably not involved in the alterations of spermatogenesis ex vivo. Indeed, their expressions are not modified during three weeks of culture, and their expressions depend on the proportion of cells where they are expressed. Our results also show that clusterin is a marker of Sertoli cells in the culture of seminiferous tubules and its expression is not altered by the presence of germ cells.</p
