    ESTIMATION OF SOME SECONDARY METABOLITES FROM THE IN VITRO CULTURES OF CHLOROPHYTUM BORIVILIANUM SANT. Et. FERN

    Objective: To perform phytochemical screening, estimate total saponins and to study the secondary metabolite profiling of Chlorophytum borivilianum by ESI-MS (Electron spray ionisation-mass spectroscopy) and GC-MS (Gas chromatography-mass spectroscopy) techniques.Methods: The powdered samples of leaves and roots (nature grown and freshly harvested and hardened in vitro regenerated) and callus of Chlorophytum borivilianum (2 g) were extracted exhaustively by soxhlet by refluxing with petroleum ether (60-80 °C) and then with ethanol (85%). Alternatively, the methanol extract was prepared for the samples by the cold maceration method and the filtered extract was used. This extract was further analyzed by gas chromatography-mass spectrometry to identify and characterize the chemical compounds present in the crude extracts. Vanillin sulphuric acid assay was done to estimate the total saponin content of extracts.Results: Phytochemical analysis of ethanolic extract showed the presence of major classes of phytochemicals. The spectrophotometric analysis as well as the GC-MS results revealed the remarkably high percentage of saponin content in the in vitro roots. Besides this, many important secondary metabolites viz. β-Sitosterol and Taraxerone were also revealed by GC-MS.Conclusion: Results of this study showed enhanced production of many important secondary metabolites, especially saponins in tubers of in vitro regenerated plantlets vis a vis that of the nature grown plants establishing the fact that micropropagation can pave new vistas for the research in phytopharmaceuticals and bioprospecting of rare and important medicinal plants.Â

    The study of forecasting identifies the urgent need for special attention in evolving effective energy policies to alleviate an energy famine in the near future. Since power demand is increasing day by day in entire world and it is also one of the fundamental infrastructure input for the development, its prospects and availability sets significant constraints on the socio-economic growth of every person as well as every country. A care full long-term power plan is imperative for the development of power sector. This need assumes more importance in the state of Uttar Pradesh where the demand for electrical energy is growing at a rapid pace. This study analyses the requirement of electricity with respect to the future population for the major forms of energy in the Lucknow city in Uttar Pradesh state of India. A model consisting of significant key energy indicators have been used for the estimation. Model wherever required refined in the second stage to remove the effect of auto-correlation. The accuracy of the model has been checked using standard statistical techniques and validated against the past data by testing for 'expost' forecast accuracy

    Broad inhibition of transmission frequency in multilayered dielectric one dimensional photonic crystal nanostructure

    We report the omnidirectional reflection (ODR) in one dimensional photonic crystal (PC) structure consisting of alternate layers of Cryolite (Na3AlF6) as material of low refractive index and Germanium (Ge) as material of high refractive index. The effects of the thickness of layers and incidence angles on the spectral reflectance have been investigated using transfer matrix method (TMM). The proposed structure gives 100% reflection within a wide range of wavelengths in the visible-near IR region (600 nm- 850 nm) which can be tuned according to the design parameters. We observe that cryolite based photonic crystal structure can be used as a good candidate for wavelength filter or broad reflector in the near infrared spectrum which is very useful in many imaging sensors in the field of optical technology

    Silicon amendment induces synergistic plant defense mechanism against pink stem borer (Sesamia inferens Walker.) in finger millet (Eleusine coracana Gaertn.).

    Silicon (Si) uptake and accumulation in plants can mitigate various biotic stresses through enhanced plant resistance against wide range of herbivores. But the role of silicon in defense molecular mechanism still remains to be elucidated in finger millet. In the present study, we identified three silicon transporter genes viz. EcLsi1, EcLsi2, and EcLsi6 involved in silicon uptake mechanism. In addition, the study also identified and characterized ten different Si transporters genes from finger millet through transcriptome assembly. The phylogenetic study revealed that EcLsi1 and EcLsi6 are homologs while EcLsi2 and EcLsi3 form another pair of homologs. EcLsi1 and EcLsi6 belong to family of NIP2s (Nod26-like major intrinsic protein), bona fide silicon transporters, whereas EcLsi2 and EcLsi3, an efflux Si transporter, belong to an uncharacterized anion transporter family having a significant identity with putative arsB transporter proteins. Further, the phylogenetic and topology analysis suggest that EcLsi1 and EcLsi2 co-evolved during evolution while, EcLsi2 and EcLsi3 are evolved from either EcLsi1 and/or EcLsi6 by fusion or duplication event. Moreover, these silicon transporters are predicted to be localized in plasma membrane, but their structural differences indicate that they might have differences in their silicon uptake ability. Silicon amendment induces the synergistic defense mechanism by significantly increasing the transcript level of silicon transporter genes (EcLsi1, EcLsi2 and EcLsi6) as well as defense hormone regulating genes (EcSAM, EcPAL and EcLOX) at 72 hpi (hours of post infestation) in both stem and roots compared to non-silicon treated plants against pink stem borer in finger millet plants. This study will help to understand the molecular defense mechanism for developing strategies for insect pest management

    Dataplane Specialization for High-performance OpenFlow Software Switching

    OpenFlow is an amazingly expressive dataplane program- ming language, but this expressiveness comes at a severe performance price as switches must do excessive packet clas- sification in the fast path. The prevalent OpenFlow software switch architecture is therefore built on flow caching, but this imposes intricate limitations on the workloads that can be supported efficiently and may even open the door to mali- cious cache overflow attacks. In this paper we argue that in- stead of enforcing the same universal flow cache semantics to all OpenFlow applications and optimize for the common case, a switch should rather automatically specialize its dat- aplane piecemeal with respect to the configured workload. We introduce ES WITCH , a novel switch architecture that uses on-the-fly template-based code generation to compile any OpenFlow pipeline into efficient machine code, which can then be readily used as fast path. We present a proof- of-concept prototype and we demonstrate on illustrative use cases that ES WITCH yields a simpler architecture, superior packet processing speed, improved latency and CPU scala- bility, and predictable performance. Our prototype can eas- ily scale beyond 100 Gbps on a single Intel blade even with complex OpenFlow pipelines
