Differential Impact of Tetratricopeptide Repeat Proteins on the Steroid Hormone Receptors
Tetratricopeptide repeat (TPR) motif containing co-chaperones of the chaperone Hsp90 are considered control modules that govern activity and specificity of this central folding platform. Steroid receptors are paradigm clients of Hsp90. The influence of some TPR proteins on selected receptors has been described, but a comprehensive analysis of the effects of TPR proteins on all steroid receptors has not been accomplished yet.We compared the influence of the TPR proteins FK506 binding proteins 51 and 52, protein phosphatase-5, C-terminus of Hsp70 interacting protein, cyclophillin 40, hepatitis-virus-B X-associated protein-2, and tetratricopeptide repeat protein-2 on all six steroid hormone receptors in a homogeneous mammalian cell system. To be able to assess each cofactor's effect on the transcriptional activity of on each steroid receptor we employed transient transfection in a reporter gene assay. In addition, we evaluated the interactions of the TPR proteins with the receptors and components of the Hsp90 chaperone heterocomplex by coimmunoprecipitation. In the functional assays, corticosteroid and progesterone receptors displayed the most sensitive and distinct reaction to the TPR proteins. Androgen receptor's activity was moderately impaired by most cofactors, whereas the Estrogen receptors' activity was impaired by most cofactors only to a minor degree. Second, interaction studies revealed that the strongly receptor-interacting co-chaperones were all among the inhibitory proteins. Intriguingly, the TPR-proteins also differentially co-precipitated the heterochaperone complex components Hsp90, Hsp70, and p23, pointing to differences in their modes of action.The results of this comprehensive study provide important insight into chaperoning of diverse client proteins via the combinatorial action of (co)-chaperones. The endothelial glycocalyx: composition, functions, and visualization
This review aims at presenting state-of-the-art knowledge on the composition and functions of the endothelial glycocalyx. The endothelial glycocalyx is a network of membrane-bound proteoglycans and glycoproteins, covering the endothelium luminally. Both endothelium- and plasma-derived soluble molecules integrate into this mesh. Over the past decade, insight has been gained into the role of the glycocalyx in vascular physiology and pathology, including mechanotransduction, hemostasis, signaling, and blood cell–vessel wall interactions. The contribution of the glycocalyx to diabetes, ischemia/reperfusion, and atherosclerosis is also reviewed. Experimental data from the micro- and macrocirculation alludes at a vasculoprotective role for the glycocalyx. Assessing this possible role of the endothelial glycocalyx requires reliable visualization of this delicate layer, which is a great challenge. An overview is given of the various ways in which the endothelial glycocalyx has been visualized up to now, including first data from two-photon microscopic imaging
What scans we will read: imaging instrumentation trends in clinical oncology
Oncological diseases account for a significant portion of the burden on public healthcare systems with associated
costs driven primarily by complex and long-lasting therapies. Through the visualization of patient-specific
morphology and functional-molecular pathways, cancerous tissue can be detected and characterized non-
invasively, so as to provide referring oncologists with essential information to support therapy management
decisions. Following the onset of stand-alone anatomical and functional imaging, we witness a push towards
integrating molecular image information through various methods, including anato-metabolic imaging (e.g., PET/
CT), advanced MRI, optical or ultrasound imaging.
This perspective paper highlights a number of key technological and methodological advances in imaging
instrumentation related to anatomical, functional, molecular medicine and hybrid imaging, that is understood as
the hardware-based combination of complementary anatomical and molecular imaging. These include novel
detector technologies for ionizing radiation used in CT and nuclear medicine imaging, and novel system
developments in MRI and optical as well as opto-acoustic imaging. We will also highlight new data processing
methods for improved non-invasive tissue characterization. Following a general introduction to the role of imaging
in oncology patient management we introduce imaging methods with well-defined clinical applications and
potential for clinical translation. For each modality, we report first on the status quo and point to perceived
technological and methodological advances in a subsequent status go section. Considering the breadth and
dynamics of these developments, this perspective ends with a critical reflection on where the authors, with the
majority of them being imaging experts with a background in physics and engineering, believe imaging methods
will be in a few years from now.
Overall, methodological and technological medical imaging advances are geared towards increased image contrast,
the derivation of reproducible quantitative parameters, an increase in volume sensitivity and a reduction in overall
examination time. To ensure full translation to the clinic, this progress in technologies and instrumentation is
complemented by progress in relevant acquisition and image-processing protocols and improved data analysis. To
this end, we should accept diagnostic images as “data”, and – through the wider adoption of advanced analysis,
including machine learning approaches and a “big data” concept – move to the next stage of non-invasive tumor
phenotyping. The scans we will be reading in 10 years from now will likely be composed of highly diverse multi-
dimensional data from multiple sources, which mandate the use of advanced and interactive visualization and
analysis platforms powered by Artificial Intelligence (AI) for real-time data handling by cross-specialty clinical experts
with a domain knowledge that will need to go beyond that of plain imaging

Validation and clinical significance of the childhood myositis assessment scale for assessment of muscle function in the juvenile idiopathic inflammatory myopathies
Objective. To examine the measurement characteristics of the Childhood Myositis Assessment Scale (CMAS) in children with juvenile idiopathic inflammatory myopathy (juvenile IIM), and to obtain preliminary data on the clinical significance of CMAS scores. Methods. One hundred eight children with juvenile IIM were evaluated on 2 occasions, 7-9 months apart, using various measures of physical function, strength, and disease activity. Interrater reliability, construct validity, and responsiveness of the CMAS were examined. The minimum clinically important difference (MID) and CMAS scores corresponding to various degrees of physical disability were estimated. Results. The intraclass correlation coefficient for 26 patients assessed by 2 examiners was 0.89, indicating very good interrater reliability. The CMAS score correlated highly with the Childhood Health Assessment Questionnaire (C-HAQ) score and with findings on manual muscle testing (MMT) (r(s) = -0.73 and 0.73, respectively) and moderately with physician-assessed global disease activity and skin activity, parent-assessed global disease severity, and muscle magnetic resonance imaging (rs = -0.44 to -0.61), thereby demonstrating good construct validity. The standardized response mean was 0.81 (95% confidence interval 0.53, 1.09) in patients with at least 0.8 cm improvement on a 10-cm visual analog scale for physician-assessed global disease activity, indicating strong responsiveness. In bivariate regression models predicting physician-assessed global disease activity, MMT remained significant in models containing the CMAS (P = 0.03) while the C-HAQ did not (P = 0.4). Estimates of the MID ranged from 1.5 to 3.0 points on a 0-52-point scale. CMAS scores corresponding to no, mild, mild-to-moderate, and moderate physical disability, respectively, were 48, 45, 39, and 30. Conclusion. The CMAS exhibits good reliability, construct validity, and responsiveness, and is therefore a valid instrument for the assessment of physical function, muscle strength, and endurance in children with juvenile IIM. Preliminary data on MID and corresponding levels of disability should aid in the clinical interpretation of CMAS scores when assessing patients with juvenile IIM

Prevalência de transtornos depressivos e ansiosos em uma amostra ambulatorial brasileira de mulheres na menopausa Prevalence of depressive and anxiety disorders in a Brazilian outpatient sample of menopausal women
OBJETIVO: Determinar a prevalência dos transtornos depressivo-ansiosos entre mulheres atendidas em um ambulatório de menopausa. METODOLOGIA: Avaliamos, através da entrevista semi-estruturada Mini International Neuropsychiatric Interview, 86 mulheres que encontravam-se em tratamento no ambulatório de menopausa do Instituto de Ginecologia da Universidade Federal do Rio de Janeiro. RESULTADOS: A maioria das mulheres apresentava algum diagnóstico psiquiátrico (57%) sendo mais prevalentes o transtorno de ansiedade generalizada (34,9%) e a depressão maior (31,4%). O grupo com algum diagnóstico foi representado por mulheres mais jovens, casadas, com menor escolaridade e história familiar para transtornos psiquiátricos. CONCLUSÃO: Observamos uma grande prevalência de transtornos mentais entre mulheres em atendimento ambulatorial na menopausa em nosso estudo, em relação às mulheres em atendimento em outros ambulatórios segundo a literatura. Há também uma alta taxa de prevalência de comorbidades (55,5% dos pacientes com algum transtorno) complicadoras do transtorno primário, o que pode representar a evolução para pior prognóstico pela ausência de tratamento precoce e específico.<br>OBJETIVE: To determine the prevalence of depressive and anxiety disorders in women receiving care in a menopause clinic. METHODS: Eighty-six women receiving care in the menopause clinic at Instituto de Ginecologia da Universidade Federal do Rio de Janeiro were assessed using the Mini-International Neuropsychiatric Interview. RESULTS: Most women had a psychiatric diagnosis (57%); generalized anxiety disorder (34.9%) and major depression (31.4%) were the most prevalent disorders. The group composed of subjects with any disorder was represented by young and married women, with lower schooling level and family history for psychiatric disorders. CONCLUSION: In our study, there was a high prevalence of psychiatric disorders in outpatient women receiving care in a menopause clinic, in relation to women receiving care in other outpatient clinics, as described in the literature. There was also a high prevalence of comorbid diseases (55.5% of patients with any disorder) complicating the primary disorder, which may compromise the prognosis due to lack of early specific treatment
