    Low Dopamine D2/D3 Receptor Availability is Associated with Steep Discounting of Delayed Rewards in Methamphetamine Dependence.

    BackgroundIndividuals with substance use disorders typically exhibit a predilection toward instant gratification with apparent disregard for the future consequences of their actions. Indirect evidence suggests that low dopamine D2-type receptor availability in the striatum contributes to the propensity of these individuals to sacrifice long-term goals for short-term gain; however, this possibility has not been tested directly. We investigated whether striatal D2/D3 receptor availability is negatively correlated with the preference for smaller, more immediate rewards over larger, delayed alternatives among research participants who met DSM-IV criteria for methamphetamine (MA) dependence.MethodsFifty-four adults (n = 27 each: MA-dependent, non-user controls) completed the Kirby Monetary Choice Questionnaire, and underwent positron emission tomography scanning with [(18)F]fallypride.ResultsMA users displayed steeper temporal discounting (p = 0.030) and lower striatal D2/D3 receptor availability (p < 0.0005) than controls. Discount rate was negatively correlated with striatal D2/D3 receptor availability, with the relationship reaching statistical significance in the combined sample (r = -0.291, p = 0.016) and among MA users alone (r = -0.342, p = 0.041), but not among controls alone (r = -0.179, p = 0.185); the slopes did not differ significantly between MA users and controls (p = 0.5).ConclusionsThese results provide the first direct evidence of a link between deficient D2/D3 receptor availability and steep temporal discounting. This finding fits with reports that low striatal D2/D3 receptor availability is associated with a higher risk of relapse among stimulant users, and may help to explain why some individuals choose to continue using drugs despite knowledge of their eventual negative consequences. Future research directions and therapeutic implications are discussed

    Surfactant status and respiratory outcome in premature infants receiving late surfactant treatment.

    BACKGROUND:Many premature infants with respiratory failure are deficient in surfactant, but the relationship to occurrence of bronchopulmonary dysplasia (BPD) is uncertain. METHODS:Tracheal aspirates were collected from 209 treated and control infants enrolled at 7-14 days in the Trial of Late Surfactant. The content of phospholipid, surfactant protein B, and total protein were determined in large aggregate (active) surfactant. RESULTS:At 24 h, surfactant treatment transiently increased surfactant protein B content (70%, p < 0.01), but did not affect recovered airway surfactant or total protein/phospholipid. The level of recovered surfactant during dosing was directly associated with content of surfactant protein B (r = 0.50, p < 0.00001) and inversely related to total protein (r = 0.39, p < 0.0001). For all infants, occurrence of BPD was associated with lower levels of recovered large aggregate surfactant, higher protein content, and lower SP-B levels. Tracheal aspirates with lower amounts of recovered surfactant had an increased proportion of small vesicle (inactive) surfactant. CONCLUSIONS:We conclude that many intubated premature infants are deficient in active surfactant, in part due to increased intra-alveolar metabolism, low SP-B content, and protein inhibition, and that the severity of this deficit is predictive of BPD.     Predation is a major cause of mortality and nest failure for the northern bobwhite (Colinus virginianus) across its range. Coyotes (Canis latrans) are a potential predator of bobwhites and typically the most common mesocarnivore on bobwhite range in Texas. Few data exist regarding the importance of bobwhites in the coyote’s diet in the Rolling Plains of Texas. We describe the seasonal and annual diets of coyotes on the Rolling Plains Quail Research Ranch (RPQRR), Fisher County, Texas. The RPQRR encompasses 1,902 ha of rolling terrain consisting of ridges and mesquite (Prosopis glandulosa) dominated rangeland; other common shrubs include netleaf hackberry (Celtis laevigata), littleleaf sumac (Rhus microphylla), lotebush (Ziziphus obtusifolia), wolfberry (Lycium berlandieri), chittam (Bumrlia lanuginosa), catclaws (Acacia spp., Mimosa spp.), and agarito (Mahonia trifoliolata). Prickly pear (Opuntia spp.) is abundant on most sites. The study area received 58 and 64 cm of rainfall, respectively during 2009 and 2010, below the 30-year average of 76 cm for Fisher County, Texas. We collected, and examined 720 coyote scats from December 2008 to December 2010. Each scat was prepared for analysis by placing it inside a nylon mesh bag and washing it for 2 cycles in an automatic washing machine. We analyzed scat contents macroscopically and any guard hairs were analyzed microscopically to identify prey to genus. We also collected estimates of abundance for a range of potential food sources including bobwhites, small mammals, and insects. Preliminary analyses suggest coyotes were minor predators of quail and their diets mainly consisted of seasonally-available mast (e.g., tunas of prickly pear) and rodents. Variation in timing and amount of rainfall during our study allowed us to document how coyotes adjusted their diets to the resulting fluctuations in food availability (especially mast)

    Increasing Student Success through a Cocktail of Cognitive Interventions

    We extended a series of interventions developed in modern cognitive psychology to a group of students who had been academically dismissed and were at high risk to not complete college. Students learned how to respond adaptively to academic failure, how to embrace challenge, how to set realistic goals, and how to persist until their goals are achieved. The interventions were delivered within a sophomore seminar course. Within the class, students learned about, considered and practiced aspects of growth mindset, goal orientation, grit, stereotype threat, and belongingness. Before beginning the class, the 68 students had a mean cumulative GPA of 1.45, a course completion rate of 60%, and it was expected that over half would drop out of college within the next year. Following the intervention, students earned a mean semester GPA of 2.39, a course completion rate of 73%, 72% were retained for the next semester, and 58% were still enrolled one year later. These findings provide support for the benefits of these techniques used together to afford student success in a population of students that have previously struggled academically.

    Immunization of Mice with Anthrax Protective Antigen Limits Cardiotoxicity but Not Hepatotoxicity Following Lethal Toxin Challenge

    Protective immunity against anthrax is inferred from measurement of vaccine antigen-specific neutralizing antibody titers in serum samples. In animal models, in vivo challenges with toxin and/or spores can also be performed. However, neither of these approaches considers toxin-induced damage to specific organ systems. It is therefore important to determine to what extent anthrax vaccines and existing or candidate adjuvants can provide organ-specific protection against intoxication. We therefore compared the ability of Alum, CpG DNA and the CD1d ligand alpha-galactosylceramide (alphaGC) to enhance protective antigen-specific antibody titers, to protect mice against challenge with lethal toxin, and to block cardiotoxicity and hepatotoxicity. By measurement of serum cardiac Troponin I (cTnI), and hepatic alanine aminotransferase (ALT), and aspartate aminotransferase (AST), it was apparent that neither vaccine modality prevented hepatic intoxication, despite high Ab titers and ultimate survival of the subject. In contrast, cardiotoxicity was greatly diminished by prior immunization. This shows that a vaccine that confers survival following toxin exposure may still have an associated morbidity. We propose that organ-specific intoxication should be monitored routinely during research into new vaccine modalities
