    IN VIVO EVALUATION OF WOUND HEALING AND ANTI-INFLAMMATORY ACTIVITY OF METHANOLIC EXTRACT OF ROOTS OF CENTAUREA AFRICANA (L.) IN TOPICAL FORMULATION

    Objective: The present study was to evaluate the anti-inflammatory and wound healing activities of methanolic extract of Centaurea africana roots in albino Wistar rats.Methods: Following extraction of the C. africana roots with 80% methanol, the extract was formulated as an ointment (5% and 10% w/w). The ointment was then evaluated for wound healing activity using excision and incision wound models. Parameters, including wound contraction, epithelization time, histopathologically, and hydroxyproline content, were determined using the excision model, whereas tensile strength was measured from the incision model. In parallel, edema of the ear, locally induced by croton oil, was studied for the assessment of anti-inflammatory activity.Results: Wound treated with 5% and 10% methanolic extract ointment exhibited a significant wound healing activity in both models as evidenced by increased wound contraction, shorter epithelization time, higher tissue breaking strength, and increased hydroxyproline content. The methanolic extract also produced dose-related significant reduction (p<0.001) of inflammation.Conclusion: Results of the present study revealed that C. africana displays remarkable wound healing and anti-inflammatory activities.Keywords: Anti-inflammatory activity, Centaurea africana roots, Methanolic extract, Incision and excision wound model

    Fizikalno-kemijske karakteristike i detekcija rezidua antibiotika u sirovom kravljem mlijeku isporučenom u tri industrije za preradu mlijeka u Alžiru

    The uncontrolled use of antimicrobials in veterinary medicine may result in the presence of their residues in food matrices of animal origin such as milk. The aim of this study was to examine the physicochemical quality and to detect antibiotic residues (β-lactams and tetracyclines) in milk, as their presence has resulted in the rejection of significant quantities of milk for marketing. The study was conducted on 274 milk samples delivered to three milk-processing units located in the suburbs of Algiers and surrounding cities (Blida and Boumerdes). Antibiotic residue detection was performed using a fast screening test (BetaStar® Combo), with a reading result within five minutes. The results indicated that the milk processing industry of Blida had the highest rejected milk volumes (43.4%), followed by Algiers (24%) and Boumerdes (19.1%). Non-compliance of milk acidity was the first cause for milk rejection (47.6 %), followed by the presence of antibiotic residues (26.8%), and incompliance with requirements for density (13.4%) and fat levels (12.2%). The study revealed 22 positive cases (8.03%) of antibiotic residues. Among these, 90.91% were positive for β-lactams, and only 9.09% for tetracyclines. All tetracyclines cases and half of the β-lactams cases were found in the Blida milk processing company, indicating that the good practice of milk production and collection must be strengthened in that company.Nekontrolirana uporaba antimikrobnih sredstava u veterinarskoj medicini može rezultirati prisustvom njihovih rezidua u matricama hrane životinjskog podrijetla kao što je i mlijeko. Ovaj rad ima za cilj proučavanje fizikalno-kemijske kakvoće i detekcije ostataka antibiotika (β-laktama i tetraciklina), čije je određivanje u mlijeku prouzročilo odbijanje znatnih količina za plasman na tržištu. Istraživanje je provedeno na 274 uzorka mlijeka dostavljena u tri jedinice za preradu mlijeka koje se nalaze u predgrađima Alžira i drugih gradovima (Blida i Boumerdes). Detekcija ostataka antibiotika provedena je brzim testom (BetaStar® Combo), s rezultatom očitanja unutar pet minuta. Rezultati su pokazali da je najveća količina odbijenog mlijeka u industriji prerade mlijeka iz jedinice Blida (43,4 %), a slijede Alžir (24 %) i Boumerdes (19,1 %). Nesukladnost kiselosti mlijeka bila je prvi uzrok odbacivanje mlijeka (47,6 %), a zatim slijede: prisutnost antibiotskih ostataka (26,8 %), gustoća (13,4 %) i nesukladnost razine masti (FL) (12,2 %). Od svih procijenjenih uzoraka, studija je otkrila 22 pozitivna slučaja (8,03 %) na ostatke antibiotika. Među njima je 90,91 % bilo pozitivno na β-laktame, dok je samo 9,09 % pozitivno na tetracikline. Svi slučajevi tetraciklina i polovica β-laktama pronađeni su u mljekoprerađivačkoj tvrtki Blida, što ukazuje da se u toj kompaniji mora ojačati dobra praksa uzgoja i prikupljanja mlijeka

    CHEMICAL ANALYSIS, ANTIOXIDANT, ANTI-INFLAMMATORY AND ANTINOCICEPTIVE EFFECTS OF ACETONE EXTRACT OF ALGERIAN SOLENOSTEMMA ARGEL (DELILE) HAYNE LEAVES

    Objective: To investigate the qualitative composition of the acetonic extract from leaves of S. argel (AESA) and their anti-inflammatory and analgesic properties in vivo. Methods: AESA profile was established by UHPLC/DAD/ESI-MS2. AESA was subjected to the acute oral toxicity study according to the OECD-420 method. Antioxydant activity of AESA was performed by DPPH radical scavenging assay. Anti-inflammatory effects of AESA were determined in two animal models: carrageenan-induced paw edema in rats and cotton pellet-induced granuloma formation in rats. Further, anti-nociceptives activities of AESA were assessed by hot plate test, acetic acid-induced abdominal writhing test and formalin test. Results: The in vivo AESA toxicity was low. AESA expresses a maximum radical scavenging activity with a IC50 value of 36,05 μg/ml. The AESA at 250 and 400 mg/kg significantly reduced carrageen an induced paw edema by 70.09% and 85.53% 6h after carrageenan injection, respectively. AESA produced significant dose-dependent anti-inflammatory effect against cotton pellets-induced granuloma formation in rats. In addition, AESA at 250 and 400 mg/kg significantly reduced acetic acid-induced writhing by 56.83 and 80.41%, respectively. Oral administration of 250 and 400 mg/kg of AESA caused a significant dose dependent anti-nociceptive effect in both neurogenic and inflammatory phases of formalin-induced licking. AESA also impacted the pain latency in the hot plat test. Conclusion: These data suggest that AESA possesses antioxidant, anti-inflammatory and anti-nociceptive effects. These results support the traditional use of S. argel to cure pain and inflammatory diseases in the Algerian Sahara

    Epidemiology of dog-mediated zoonotic diseases in Algeria: a One Health control approach

    Globally, human–dog interactions cause significant social, economic and human health costs. Public health problems linked with dogs include mainly zoonotic diseases. Recently the concept of a global and integrative approach to improve the health and well-being of people, animals and the environment has been strongly endorsed. This concept, often referred to as One Health, also reflects the collaboration in the field of surveillance and monitoring. Because humans and animals often suffer from the same pathogens and share the same environment, a cross-sector approach integrating human and animal disease surveillance information is required. The aims of the present study were to describe the incidence of dog-mediated zoonotic diseases in Algeria between 2010 and 2017, and to propose a One Health approach to control these diseases in Algeria. Rabies, leishmaniasis and echinococcosis are the major zoonoses in Algeria, with a reported average number of deaths per year, respectively, of 18, 7947 and 387. These zoonoses occur with the uncontrolled proliferation of household waste deposits, particularly in and around urban area which maintain the presence of stray dogs. The persistence of these diseases indicates the need for greater partnership and collaboration among multiple sectors, including medical doctors, veterinarians, ecologists, environmentalists and law-enforcement agents. Such partnerships permit the sharing of information, facilities and resources under a One Health approach; permit rapid communication among disciplines as well as interdisciplinary training/education opportunities and raising awareness among human population; and allow a combined effort towards disease surveillance/control, which will consequently improve the efficiency of the control programmes.     ANTIBACTERIAL ACTIVITY OF MENTHA PULEGIUM ESSENTIAL OIL AGAINST AVIAN ISOLATED ESBL PRODUCING BACTERIA AND ITS SYNERGISTIC POTENTIAL WITH ANTIBIOTICS

    Objective: The aim of this study was to evaluate the antibacterial effect of Algerian Mentha pulegium essential oil against extended-spectrum β-lactamases (ESBL) producing bacteria isolated from avian livestock. The synergistic interactions between conventional antibiotics and Mentha pulegium essential oil were assessed.Methods: Essential oil composition was determined by gas chromatograph-flame ionization detector (GC/FID), gas chromatograph-mass spectrometer (GC/MS) analysis.Antibacterial activity of Mentha pulegium essential oil against ESBL producing bacteria was investigated by disc diffusion assay. Minimum inhibitory concentration (MIC) of essential oil and it synergistic interaction with conventional antibiotics were determined by micro-broth dilution method and checkerboard test, respectively.Results: The results indicate that Mentha pulegium essential oil with a high amount of pulegone (88.78%) had high inhibitory activity against the tested strains and particularly displays a satisfactory action against the studied ESBL producing bacteria from animal origin, with a diameter ranging from 13 to 26 mm. Out of 51 combinations tested between essential oil and antibiotics 60.78 % showed total synergy, 13.72 % had presented a partial synergy. The best antibacterial activities were obtained with the combination of Mentha pulegium essential oil and cefazolin, cefotaxime and gentamycin.Conclusion: This study allowed concluding that Mentha pulegium essential oil showed not only satisfactory antibacterial properties, but also acts synergistically combined with conventional antibiotics, which make it a promising alternative to antimicrobial drugs; beside that, it might reduce the minimum effective dose of the drugs which minimizes their possible side effects

    Occurrence and seasonal variation of aflatoxin M1 in raw cow milk collected from different regions of Algeria

    Background and Aim: Aflatoxins are metabolites of molds that exert potentially toxic effect on animals and humans. This study aimed to investigate the occurrence of aflatoxin M1 (AFM1) in raw cow milk collected during 1 year (2016-2017) from different regions of Algeria and risk factors associated with the contamination. Materials and Methods: During the survey period, 84 samples of raw milk were collected in three regions of Algeria (northeast, north center, and northwest) during four seasons. AFM1 levels were analyzed by competitive enzyme-linked immunosorbent assay. Results: AFM1 was detected in 39 (46.43%) samples (total mean concentration, 71.92 ng/L; range, 95.59-557.22 ng/L). However, the AFM1 levels exceeded the maximum tolerance limit set by the Food and Drug Administration in the USA (500 ng/L) in only 1 sample (1.19%). Statistical analysis revealed significant differences (p<0.005) between AFM1 levels in milk samples collected in the spring and autumn. The mean AFM1 levels in samples collected in the spring were significantly higher than those in samples collected in autumn. Conclusion: The survey indicates that farmers involved in milk production should be made aware of the adverse effects of aflatoxin contamination in animal feed. A systematic control program of supplementary feedstuff for lactating cows should be introduced by the public health authorities

    Characterization of quinolone-resistant Enterobacteriaceae strains isolated from poultry in Western Algeria: First report of qnrS in an Enterobacter cloacae

    Aim: Multidrug-resistant (MDR) Enterobacteriaceae have frequently been reported, in both human and veterinary medicine, from different parts of the world as a consequence of antibiotic usage. However, there is a lack of published data regarding antimicrobial resistance in non-Escherichia coli (E. coli) Enterobacteriaceae from animals in Algeria. This study aimed to evaluate the frequency of resistance to antibiotics with a focus on quinolones and to investigate the presence of qnr genes in Enterobacteriaceae of poultry origin. Materials and Methods: A total of 310 samples of poultry origin were collected from 2010 to 2014 from broiler and layer farms and hatcheries located in different geographic areas of Western Algeria (including Mostaganem, Oran, Mascara, Relizane, Chlef, Tiaret, and Tissemsilt). Antimicrobial susceptibility testing was performed using disc diffusion assay. Polymerase chain reaction and sequencing accomplished the characterization of qnr genes (qnrA, qnrB, and qnrS). Results: A total of 253 Enterobacteriaceae strains were isolated in this study. These isolates exhibited high levels of resistance to quinolones and other families of antibiotics. All the strains isolated in this study were resistant to at least one antibiotic. Among them, 233 (92.09%) were considered MDR. Among the 18 randomly selected nalidixic acid (NA)- resistant Enterobacteriaceae isolates, one E. coli and one Enterobacter cloacae were carrying qnrS1. By contrast, qnrA and qnrB were not detected in this study. Conclusion: This is the first report on the identification of the qnrS gene in E. cloacae isolated from animal source in Algeria. Further studies have to be conducted to determine the real prevalence of qnr genes

    Seroprevalence of West Nile virus antibodies in equids in the North-East of Algeria and detection of virus circulation in 2014

    International audienceWest Nile fever (WNF) is a viral disease of wild birds transmitted by mosquitoes. Humans and equids can also be affected and suffer from meningoencephalitis. In Algeria, since the 1994 epidemic, no data on WNV circulation was available until 2012. In September 2012, a fatal human case of WNV neuro-invasive infection occurred in Jijel province. This study describes the first seroprevalence study of West Nile virus (WNV) antibodies conducted in the equine population in Algeria. During 2014, serum samples were collected from 293 equids (222 donkeys and 71 horses) asymptomatic and unvaccinated for WNV in three localities in Northeastern wetlands of Algeria. Antibodies against WNV were found in 51 samples (seroprevalence 17.4%) of sampled equids, distributed as follows: 19 (seroprevalence 26.8%) horses and 32 (seroprevalence 14.4%) donkeys. Moreover 7 horses coming from Blida, in the center of Algeria, were tested before and after an 8-months stay in North-East Algeria. We observe a seroconversion in 2 horses, showing WNV circulation in 2014 in this specific region of Algeria. (C) 2016 Published by Elsevier Ltd
