    Peptide Substrates for Rho-Associated Kinase 2 (Rho-Kinase 2/ROCK2)

    Peptide substrates sensitive for a certain protein kinase could be important for new-drug development and to understand the mechanism of diseases. Rho-associated kinase (Rho-kinase/ROCK) is a serine/threonine kinase, and plays an important part in cardiovascular disease, migration and invasion of tumor cells, and in neurological disorders. The purpose of this study was to find substrates with high affinity and sensitivity for ROCK2. We synthesized 136 peptide substrates from protein substrates for ROCK2 with different lengths and charged peptides. Incorporation of 32P [counts per minute (CPM)] for each peptide substrate was determined by the radiolabel assay using [γ-32P]ATP. When the top five peptide substrates showing high CPMs (R4, R22, R133, R134, and R135) were phosphorylated by other enzymes (PKA, PKCα, and ERK1), R22, R133, and R135 displayed the highest CPM level for ROCK2 compared with other enzymes, whereas R4 and R134 showed similar CPM levels for ROCK2 and PKCα.     How do surgeons experience and cope with the death and dying of their patients? A qualitative study in the context of life-limiting illnesses

    BackgroundThe present qualitative study explores surgeons' experiences and coping mechanisms with the death of their patients in the context of life-limiting illnesses.MethodsNine Australian surgeons participated in in-depth interviews. Thematic analysis of the interview transcriptions allowed for identification of themes and subthemes. Methodological trustworthiness and rigor were preserved at all stages of the research process.ResultsThe essence of surgical practice, comprising participants' descriptions of their identity as surgeons, influenced their experiences and coping mechanisms when dealing with death and dying. The following subthemes were identified: distancing from patients, wanting to fix problems, doing in terms of operating on a patient, prominence of the surgeon's personal ability, and high personal responsibility for patient outcomes. Giving hope and having a mixed surgical practice were some of the coping mechanisms described by participants.DiscussionDeath and dying are a part of surgical practice, and surgeons face challenges as they care for dying patients. Although participants stated that they felt less affected by dealing with patients dying of life-limiting illnesses than by unexpected deaths, their descriptions of their encounters, reactions, and behaviors, as well as their use of language, suggest that these deaths have a lasting impact. Potential areas for improvement of surgical practice and approaches to teaching about death and dying in surgery are addressed.     Interpreting realized pollen flow in terms of pollinator travel paths and land-use resistance in heterogeneous landscapes

    Widespread ecosystem change has led to declines in species world-wide. The loss of pollinators in particular constitutes a problem for ecosystem function and crop production. Understanding how landscape change affects pollinator movement, effective pollen flow, and plant and pollinator survival is therefore a global priority. In this study we investigated patterns of effective pollen flow, using wild cherry tree (Prunus avium) progeny arrays, to address two questions in three case studies: Do land-use types present different resistances to pollinator movement? Which pollinator travel path best explains the pollination data (straight lines, weighted straight lines, least cost paths or pair-wise resistance)? Trees and progeny arrays were genotyped and effective pollen flow and pollinator movement were estimated using the spatially explicit mating model. We found that pollinators did modify their travel paths in response to land-use type and arrangement, but the travel path that best described pollinator movement and the resistance rank of the land uses depended on the type and size of land-use patches and the landscape context. We propose a novel theoretical framework rooted in behavioural ecology, the resource model, for interpreting pollinator behaviour in heterogeneous landscapes. We conclude by discussing the importance and practicality of conservation and management strategies in which native and non-native land-use types together provide functional habitat and support ecosystem services across economic landscapes
