    During embryonic development and tissue homeostasis, reproducible proportions of differentiated cell types are specified from populations of multipotent precursor cells. Molecular mechanisms that enable both robust cell-type proportioning despite variable initial conditions in the precursor cells, and the re-establishment of these proportions upon perturbations in a developing tissue remain to be characterized. Here, we report that the differentiation of robust proportions of epiblast-like and primitive endoderm-like cells in mouse embryonic stem cell cultures emerges at the population level through cell-cell communication via a short-range fibroblast growth factor 4 (FGF4) signal. We characterize the molecular and dynamical properties of the communication mechanism and show how it controls both robust cell-type proportioning from a wide range of experimentally controlled initial conditions, as well as the autonomous re-establishment of these proportions following the isolation of one cell type. The generation and maintenance of reproducible proportions of discrete cell types is a new function for FGF signaling that might operate in a range of developing tissues

    Regional Sea Level Variability and Trends, 1960-2007: A Comparison of Sea Level Reconstructions and Ocean Syntheses

    Several existing statistical and dynamical reconstructions of past regional sea level variability and trends are compared with each other and with tide gauges over the 48 year period 1960-2007, partially predating the satellite altimetry era. Evaluated statistical reconstructions were built from tide-gauge data (TGR), and dynamical reconstructions from ocean data assimilation (ODA) approaches. Although most of the TGRs yield global-mean time series of sea level with trends deviating within ± 0.1 mm yr-1, the spatial anomalies of the trends deviate substantially between the reconstructions over the period predating altimetry. In contrast, TGRs match observed regional trend patterns fairly well during the satellite altimetry era. TGRs match tide-gauge data better than ODA results; however, they exhibit less variability in the open ocean compared to altimetric data. Over the prealtimetry period, all reconstructed regional sea level trend patterns deviate substantially from each other. In terms of detrended correlations in this earlier period, the reconstructions match tide gauges, and each other, much better in the Pacific than in the Atlantic. An ensemble of all TGR and ODA estimates provides some improvements in correlations and trends to both tide gauges and altimetry. Nevertheless, a lack of independent open ocean sea surface height data predating altimetry makes impossible the validation of the ensemble for prealtimetry open ocean sea level trends and variability. Estimating regional sea level changes prior to altimetry therefore remains an unsolved challenge

    High-sensitivity monitoring of micromechanical vibration using optical whispering gallery mode resonators

    The inherent coupling of optical and mechanical modes in high finesse optical microresonators provide a natural, highly sensitive transduction mechanism for micromechanical vibrations. Using homodyne and polarization spectroscopy techniques, we achieve shot-noise limited displacement sensitivities of 10^(-19) m Hz^(-1/2). In an unprecedented manner, this enables the detection and study of a variety of mechanical modes, which are identified as radial breathing, flexural and torsional modes using 3-dimensional finite element modelling. Furthermore, a broadband equivalent displacement noise is measured and found to agree well with models for thermorefractive noise in silica dielectric cavities. Implications for ground-state cooling, displacement sensing and Kerr squeezing are discussed.Comment: 25 pages, 8 figure

    This report develop a concept for evaluating alternative design solutions for urban streets: the Street Performance Assessment Scheme. It should be generally valid and applicable to any redesign task, to compare the performance of a road/street section: (1) with the goals formulated for each case study, (2) in situations before and after the implementation of a redesign solution, and (3) between different case studies in cross-site assessments. In this report, the Street Performance Assessment Scheme is applied to streets in five European cities. These streets have major movement and 'place' functions and have limited space availabilities, and are thus typical examples for the most challenging design tasks that urban street designers face

    Spin-momentum locking from topological quantum chemistry: applications to multifold fermions

    In spin-orbit coupled crystals, symmetries can protect multifold degeneracies with large Chern numbers and Brillouin zone spanning topological surface states. In this work, we explore the extent to which the nontrivial topology of chiral multifold fermions impacts the spin texture of bulk states. To do so, we formulate a definition of spin-momentum locking in terms of reduced density matrices. Using tools from the theory of topological quantum chemistry, we show how the reduced density matrix can be determined from the knowledge of the basis orbitals and band representation forming the multifold fermion. We show how on-site spin orbit coupling, crystal field splitting, and Wyckoff position multiplicity compete to determine the spin texture of states near chiral fermions. We compute the spin texture of multifold fermions in several representative examples from space groups P432P432 (207) and P213P2_13 (198). We show that the winding number of the spin around the Fermi surface can take many different integer values, from zero all the way to ±7\pm 7. Finally, we conclude by showing how to apply our theory to real materials using the example of PtGa in space group P213P2_13.Comment: 28 pages, 6 figure
