Impact of stressing a pen mate on physiological responses of growing pigs
Crossbred barrows and gilts (n = 36), weighing 16.59 ± 2.1 kg, were used to test the effects of stressing a pen mate on the physiological responses of growing pigs. Pigs were randomly allotted to 6 groups after stratifying according to gender, litter origin, and body weight. Dominance order was determined within each group, and 1 to 3 d prior to the stress treatment the most- and leastdominant pigs within a group were fitted with indwelling catheters in their vena cavas. Over 3 d, groups were either: 1) isolated from audile and visual contact with stressed pigs in a separate room (non-stressed control); 2) separated by a curtain from visual contact with stressed pigs; or 3) allowed to maintain audile and visual contact with stressed pigs. Blood samples were collected 30, 15, and 0 min before exposure to the stressor (snout-snare) treatment and again at 1, 2, 3, 4, 5, 7.5, 10, 15, 20, 25, and 30 min after stressor application. Serum cortisol and plasma glucose, lactate, and nonesterified fatty acids (NEFA) concentrations were measured. There were no treatment à sampling-time interactions (P \u3e 0.17) for concentrations of cortisol, glucose, lactate or NEFA, nor were these metabolites affected by stressor treatment (P \u3e 0.42). Humoral measures of the stress response were not affected by visual and/or audile contact with pen mates undergoing a stressful event
Producing the docile body: analysing Local Area Under-performance Inspection (LAUI)
Sir Michael Wilshaw, the head of the Office for Standards in Education (OfSTED), declared a 'new wave' of Local Area Under-performance Inspections (LAUI) of schools 'denying children the standard of education they deserve'. This paper examines how the threat of LAUI played out over three mathematics lessons taught by a teacher in her first year in the profession. A Foucauldian approach is mobilised with regard to disciplinary power and 'docile bodies'. The paper argues that, in the case in point, LAUI was a tool mediating performative conditions and, ultimately, the docile body. The paper will be of concern to policy sociologists, teachers, school leaders, and those interested in school inspection
Transcriptome response of pubertal male Clarias macrocephalus to dietary 17α-methyltestosterone
We studied the mechanism of the effects of 17α-methyltestosterone (MT) on previously reported enhanced growth and maturation of the Asian catfish Clarias macrocephalus. A total of 18 pubertal male (3 fish·tank-1) catfish (ABW=125.97 g) were stocked into six experimental circular tanks and fed with a basal diet (control) or a basal diet containing 60 mg MT kg-1 diet for 90 days. At the termination of the experiment, livers were excised, and total RNA was extracted, evaluated, and reverse transcribed to cDNA. Six libraries of cDNA (3 for the control and 3 for the MT-treated group) were subjected to bioinformatics. Differentially expressed genes (DEGs) were identified using similarity scores of the de novo assembled transcript with those in the seven public databases. There were 141 up- and 72 down-regulated DEGs, mostly belonging to uncharacterized/not yet identified proteins. Thus, DEGs in the top enriched KEGG pathways were used to characterize the effects of MT. Upregulated DEGs included tyrosine hydroxylase (TH), Cytoplasmic Polyadenylation Element Binding (CPEB), Fas-associated death domain (FADD), Major Histocompatibility Complex class 1 (MHC-1), other immune-related genes, among others. Down-regulated DEGs included Cytochrome P450 family one subfamily A member 1 (CYP1A1), Alternative oxidase (AOX), UDP-glycosyltransferase (UGT), Acyl-CoA Synthase Bubblegum Family Member 1 (ACSBG), Stearoyl-CoA Desaturase (SCD), among others. In conclusion, MT-treated male Clarias macrocephalus exhibited up-regulation of pathways protecting cellular conditions like cell proliferation, survival, development and homeostasis processes, for development and homeostasis processes. MT also affected changes in bile acid production and the inhibition of the production/conversion of testosterone. MT resulted in the down-regulation of a serotonergic system that possibly affected gonadal development, inhibition of the retinoid enzyme that would otherwise diminish the full effects of MT or its metabolite, and inhibition of the formation of estrogen, leading to down-regulation of several estrogen-related KEGG pathways. The results provide valuable information about the key genes for use as biomarkers of maturation and reproduction for the Asian catfish and contribute to our understanding of the molecular mechanisms and regulative pathways behind these two processes in fish
Impact of Scottish vocational qualifications on residential child care : have they fulfilled the promise?
This article will present findings from a doctoral study exploring the impact of 'SVQ Care: Promoting Independence (level III)' within children's homes. The study focuses on the extent to which SVQs enhance practice and their function within a 'learning society'. A total of 30 staff were selected from seven children's homes in two different local authority social work departments in Scotland. Each member of staff was interviewed on four separate occasions over a period of 9 months. Interviews were structured using a combination of repertory grids and questions. Particular focus was given to the assessment process, the extent to which SVQs enhance practice and the learning experiences of staff. The findings suggest that there are considerable deficiencies both in terms of the SVQ format and the way in which children's homes are structured for the assessment of competence. Rather than address the history of failure within residential care, it appears that SVQs have enabled the status quo to be maintained whilst creating an 'illusion' of change within a learning society
Critical Race Theory and Education: racism and anti-racism in educational theory and praxis
What is Critical Race Theory (CRT) and what does it offer educational researchers and practitioners outside the US? This paper addresses these questions by examining the recent history of antiracist research and policy in the UK. In particular, the paper argues that conventional forms of antiracism have proven unable to keep pace with the development of increasingly racist and exclusionary education polices that operate beneath a veneer of professed tolerance and diversity. In particular, contemporary antiracism lacks clear statements of principle and theory that risk reinventing the wheel with each new study; it is increasingly reduced to a meaningless slogan; and it risks appropriation within a reformist âcan doâ perspective dominated by the de-politicized and managerialist language of school effectiveness and improvement. In contrast, CRT offers a genuinely radical and coherent set of approaches that could revitalize critical research in education across a range of inquiries, not only in self-consciously "multicultural" studies. The paper reviews the developing terrain of CRT in education, identifying its key defining elements and the conceptual tools that characterise the work. CRT in education is a fast changing and incomplete project but it can no longer be ignored by the academy beyond North America
N-P co-limitation of primary production and response of arthropods to N and P in early primary succession on Mount St. Helens Volcano
Background: The effect of low nutrient availability on plant-consumer interactions during early succession is poorly understood. The low productivity and complexity of primary successional communities are expected to limit diversity and abundance of arthropods, but few studies have examined arthropod responses to enhanced nutrient supply in this context. We investigated the effects of nitrogen (N) and phosphorus (P) addition on plant productivity and arthropod abundance on 24-yr-old soils at Mount St. Helens volcano. Methodology/Principal Findings:We measured the relative abundance of eight arthropod orders and five families in plots that received N, P, or no nutrients for 3-5 years. We also measured plant % cover, leaf %N, and plant diversity. Vegetation responded rapidly to N addition but showed a lagged response to P that, combined with evidence of increased N fixation, suggested P-limitation to N availability. After 3 yrs of fertilization, orthopterans (primarily Anabrus simplex (Tettigoniidae) and Melanoplus spp (Acrididae)) showed a striking attraction to P addition plots, while no other taxa responded to fertilization. After 5 yrs of fertilization, orthopteran density in the same plots increased 80%-130% with P addition and 40% with N. Using structural equation modeling, we show that in year 3 orthopteran abundance was associated with a P-mediated increase in plant cover (or correlated increases in resource quality), whereas in year 5 orthopteran density was not related to cover, diversity or plant %N, but rather to unmeasured effects of P, such as its influence on other aspects of resource quality. Conclusions/Significance:The marked surprising response to P by orthopterans, combined with a previous observation of P-limitation in lepidopteran herbivores at these sites, suggests that P-mediated effects of food quantity or quality are critical to insect herbivores in this N-P co-limited primary successional system. Our results also support a previous suggestion that the availability of N in these soils is P-limited. © 2010 Bishop et al
