Variability and differentiation in Melitaea species (Lepidoptera: Nyphalidae) in the Carpathian Basin
A Melitaea genus Mellicta subgenusába tarozĂł lepkefajok populáciĂłi EurĂłpa egyes rĂ©szein hanyatlĂłak vagy alacsony egyedszámĂşak, Ăgy konzerváciĂłbiolĂłgiai Ă©s termĂ©szetvĂ©delmi jelentĹ‘sĂ©gĂĽk igen nagy. A subgenus taxonĂłmiailag bonyolult, mivel kĂĽlsĹ‘ jellegeik igen variábilisak, valamint rejtett fajok, alfajok lehetnek jelen. Az Ă©lĹ‘helyeik nagyon hasonlĂłak, bizonyos Ă©lĹ‘helyeken egymás mellett fordulnak elĹ‘. Életmenet stratĂ©giájukbĂłl adĂłdĂłan a Melitaea-fajok populáciĂłiban nagymĂ©rtĂ©kű lehet a genetikai sodrĂłdás a vĂ©letlenszerű esemĂ©nyek (sztochasztikus hatások) bekövetkezĂ©se miatt. Ugyanis egy nĹ‘stĂ©ny egy csomĂłba viszonylag nagy mennyisĂ©gű petĂ©t rak le, Ă©s a fiatal hernyĂłk is egy darabig társasan Ă©lnek, ezĂ©rt egy nĹ‘stĂ©ny összes utĂłda egyszerre van kitĂ©ve a kĂĽlönbözĹ‘ környezeti faktorok hatásainak.
Az általam vizsgált három tarkalepkefaj hasonlĂł fejlĹ‘dĂ©smenetű, de Ă©letciklusukban egy igen lĂ©nyeges eltĂ©rĂ©st lehet megfigyelni, ez pedig a voltinizmus. MĂg a M. britomartis Ă©s a M. aurelia Ă©vente egy nemzedĂ©kes, addig a M. athalia esetĂ©ben kĂ©t tĂpust lehet elkĂĽlönĂteni. Az egyik fenolĂłgiai tĂpusnak egy generáciĂłja van egy Ă©vben (monovoltin), mĂg a másik tĂpusnak kettĹ‘ (bivoltin). Továbbá a M. athalia lárvális fejlĹ‘dĂ©se kis mĂ©rtĂ©kben eltĂ©r a másik kĂ©t fajĂ©tĂłl, ugyanis hernyĂłik már a diapauza elĹ‘tt szĂ©tszĂ©lednek a hernyĂłfĂ©szekbĹ‘l, egyedileg táplálkoznak Ă©s egyedileg esztiválnak illetve hibernálnak. Ezek a lárvák a másik kĂ©t faj hernyĂłinál jĂłval szĂ©lesebb tápnövĂ©nyspektrumot kĂ©pesek hasznosĂtani.
Vizsgálataim során a Mellicta subgenus három fajának Kárpát-medencei populáciĂłiban tanulmányoztam a genetikai variabilitás szerkezetĂ©t, továbbá a differenciálĂłdás mĂ©rtĂ©kĂ©t. Ennek a vizsgálatnak az eredmĂ©nyeire alapozva kĂĽlönös figyelmet fordĂtottam a M. athalia kĂ©t fenolĂłgiai tĂpusának vizsgálatára, valamint a fajon belĂĽli differenciálĂłdás mintázatának feltárására. EzĂ©rt ennek a fajnak kĂĽlön vizsgáltam a genetikai variabilitását Ă©s fajon belĂĽli differenciálĂłdásának mĂ©rtĂ©kĂ©t is, továbbá párhuzamos elemzĂ©st vĂ©geztem a fenotĂpusos variabilitás Ă©s differenciálĂłdás vonatkozásában. Az eredmĂ©nyeket felhasználva a genetikai Ă©s a fenotĂpusos variabilitás szintje illetve struktĂşrája közötti korreláciĂłt is megvizsgáltam a M. athalia populáciĂłkban.
Dolgozatom főbb eredményeit az alábbiakban foglalom össze:
A vizsgált három Melitaea-faj enzimpolimorfizmus vizsgálata során három EvolĂşciĂłsan Szignifikáns EgysĂ©get (ESU-t) kĂĽlönĂtettem el a Mellicta subgenuson belĂĽl a három fajnak megfelelĹ‘en. Az eredmĂ©nyeim alapján a M. britomartis Ă©s a M. aurelia közelebbi, mĂg a M. athalia távolabbi rokonnak bizonyultak. Ez ellentmond Wahlberg Ă©s mtsai. eredmĂ©nyeinek, mivel a vizsgálataik alapján a M. athalia Ă©s a M. britomartis bizonyult közeli rokonnak. TermĂ©szetesen azt meg kell jegyezni, hogy Ĺ‘k más mĂłdszerrel, a Melitaeini tribusz összes fajának egy-egy egyedĂ©t vizsgálták. A M. athalia kĂ©t fenolĂłgiai tĂpusa között egyik analĂzis során sem mutatkozott kĂĽlönbsĂ©g, azaz nem tekinthetĹ‘k genetikailag kĂ©t kĂĽlön csoportnak. Mindezek alapján valĂłszĂnűsĂthetĹ‘, hogy az egy illetve kĂ©t generáciĂłs jelleg egyfajta fenotĂpusos plaszticitás, vagyis az elkĂĽlönĂĽlĂ©s Ă©lĹ‘helybeli sajátosságokon alapszik. Az analĂzisek eredmĂ©nyei azonban további kĂ©rdĂ©seket vetettek fel, ugyanis a M. athalia populáciĂłk genetikailag nagymĂ©rtĂ©kű differenciálĂłdást mutattak egymástĂłl.
A M. athalia genetikai valamint morfometriai adatainak párhuzamos elemzĂ©se során fĂ©nyt derĂtettem a populáciĂłk differenciálĂłdási struktĂşrájának mintázatára. Minden genetikai analĂzisben kimutattam a populáciĂłk regionális differenciálĂłdását, ráadásul ebben az esetben a rĂ©giĂłk kĂ©t jĂłl elkĂĽlönĂthetĹ‘ csoportot alkottak a keleti-, ill. nyugati Kárpát-medencĂ©nek megfelelĹ‘en. A M. athalia kĂ©t fenolĂłgiai tĂpusa ebben a vizsgálatban sem kĂĽlönĂĽlt el egymástĂłl egyik analĂzisben sem, tehát ebben a tĂpusĂş vizsgálatban is azt tapasztaltam, hogy nincs a kĂ©t csoport között genetikai kĂĽlönbsĂ©g. A fenotĂpusos differenciálĂłdást megvizsgálva kĂĽlön-kĂĽlön a szárny valamint a hĂmivarszerv jellegeire vonatkoztatva szintĂ©n a populáciĂłk regionális differenciálĂłdását tapasztaltam. A rĂ©giĂłk a szárnyjellegek vonatkozásában rĂ©szlegesen kĂĽlönĂĽltek el a keleti-, ill. nyugati Kárpát-medencĂ©nek megfelelĹ‘en, azonban az ivarszerv jellegei esetĂ©ben ilyen mintázat nem volt megfigyelhetĹ‘. EredmĂ©nyeim azt tĂĽkrözik tehát, hogy a genetikai valamint a morfometriai vizsgálatok eredmĂ©nyei hasonlĂłak, ugyanis mindkĂ©t esetben a populáciĂłk regionális differenciálĂłdása volt kimutathatĂł. Ez a mintázat azonban az enzimpolimorfizmus vizsgálatok során markánsabbnak bizonyult, mivel ebben az esetben a rĂ©giĂłkra ráépĂĽl egy magasabb hierarchia szint is, amely rĂ©vĂ©n a földrajzi rĂ©giĂłk a keleti-, ill nyugati Kárpát-medencĂ©nek megfelelĹ‘en csoportosulnak. Bár a mintavĂ©teli stratĂ©giábĂłl adĂłdĂłan nem zárhatĂł ki a genetikai klin meglĂ©te, azonban az eredmĂ©nyek sokkal erĹ‘sebben támasztják alá a keleti-, ill. nyugati Kárpát-medence populáciĂłinak elkĂĽlönĂĽlĂ©sĂ©t.
A genetikai variabilitást megvizsgálva a Mellicta subgenuson belĂĽl mindhárom faj tekintetĂ©ben magas szintű enzimpolimorfizmust tapasztaltam. A vizsgált 15 enzimlokusz közĂĽl mind rendelkezett alternatĂv allĂ©lokkal, sĹ‘t a legtöbb igen polimorfnak bizonyult. A polimorfizmus mutatĂłi mindhárom esetben magasnak mutatkoztak, de a három faj közĂĽl M. athalia volt a legvariábilisabb.
Mivel a Melitaea-fajok az r-K kontinuum r-hez közelebbi vĂ©gĂ©n helyezkednek el, a populáciĂłikat jelentĹ‘s mĂ©rtĂ©kben Ă©rintik a sztochasztikus hatások, Ăgy intenzĂv drift várhatĂł melynek következmĂ©nyekĂ©nt általánosságban a populáciĂłknak alacsony szintű lesz a variabilitása.
A dolgozatom eredmĂ©nyei ezzel ellentĂ©tben magas szintű variabilitást tĂĽkröztek, tehát nem a drift tűnt a variabilitás szintjĂ©t befolyásolĂł elsĹ‘dleges tĂ©nyezĹ‘nek. KĂ©t további lehetsĂ©ges tĂ©nyezĹ‘ hatását elemezve (általános ökolĂłgiai jellemzĹ‘k eltĂ©rĂ©sei, kĂĽlönös tekintettel a tápnövĂ©ny hasznosĂtás spektrumának nagyságára, vizsgált populáciĂłk helyzete a faj areáján belĂĽl) oly mĂłdon, hogy a 3 Mellicta faj variabilitását a Melitaeini tribusz 4 további fajának (Euphydryas maturna, E. aurinia, Melitaea phoebe Ă©s M. ornata) enzimpolimorfizmu adataival hasonlĂtottam össze. Az eredmĂ©nyek azt mutatták, hogy a vizsgált fajok variabilitásának mintázatát a centrális–marginális hipotĂ©zis magyarázza meg a legteljesebben.
A morfolĂłgiai jellegek változatossága sok vizsgálatban tĂĽkrözte a genetikai variabilitás szintjĂ©t Ă©s szerkezetĂ©t. MeglepĹ‘ mĂłdon a Levene-változĂłk százalĂ©kos Ă©rtĂ©kei azt sugallták, hogy az ivarszerveken mĂ©rt jellegek variabilitása magasabb, mint a szárnyjellegekĂ©. Ez az eredmĂ©ny váratlan, hiszen a szárnyak köztudottan ki vannak tĂ©ve a környezeti tĂ©nyezĹ‘k hatásainak, mĂg az ivarszervek morfolĂłgiája feltehetĹ‘en sokkal jobban behatárolt.
A M. athalia fenotĂpusos variabilitásának vizsgálata során a kĂĽlönbözĹ‘ analĂzisek arra világĂtottak rá, hogy sem a szárny, sem pedig az ivarszerv jellegek variabilitásában nincs egyĂ©rtelmű, szignifikáns kĂĽlönbsĂ©g a fenotĂpusos variancia mĂ©rtĂ©kĂ©ben a hierarchia felsĹ‘ szintjĂ©n. Ugyanakkor a kĂ©t jellegcsoport esetĂ©ben ellentĂ©tes tendenciát tapasztaltam a morfometriai variabilitás mĂ©rtĂ©kĂ©ben a Kárpát-medence keleti, illetve nyugati terĂĽletĂ©nek populáciĂłi között. MĂg a szárnyakon mĂ©rt jellegek varianciája párhuzamos tendenciákat mutat a genetikai varianciával, addig az ivarszervek jellegeinĂ©l ellentĂ©tes trend volt megfigyelhetĹ‘.
A genetikai Ă©s a fenotĂpusos variabilitás szintje közötti korreláciĂłs vizsgálat eredmĂ©nyei elsĹ‘ megközelĂtĂ©sben azt mutatták, hogy a genetikai Ă©s morfolĂłgiai variabilitás között nincs szoros kapcsolat sem a szárnyak, sem pedig az ivarszerv jellegeit tekintve. A rĂ©szletesebb elemzĂ©s azonban arra világĂtott rá, hogy a szárnyjellegek kĂ©t tĂpusának (mĂ©ret+alak Ă©s mintázat) variabilitása között nincs korreláciĂł. ĂŤgy ezt a kĂ©t jellegcsoportot kĂĽlön kezelve a vizsgálati eredmĂ©nyek azt tĂĽkröztĂ©k, hogy a kevĂ©sbĂ© variábilis szárnymĂ©ret Ă©s alak jobban jelzi a genetikai variabilitás szintjĂ©t, mint a szárny mintázata, melyrĹ‘l számottevĹ‘ mĂ©rtĂ©kű plaszticitást tĂ©telezhetĂĽnk fel. Az a tĂ©ny, hogy a mĂ©retet Ă©s alakot meghatározĂł jellegek fenotĂpusos varianciája alacsonyabb szintű, mint a mintázatot jellemzĹ‘kĂ©, valamint hogy a szárnymĂ©ret varianciája szignifikáns korreláciĂłban van az enzimpolimorfizmus mĂ©rtĂ©kĂ©vel azt sugallja, hogy ezeknek a jellegeknek a fenotĂpusos varianciáját nagyrĂ©szt genetikai faktorok határozzák meg.
A genetikai Ă©s a fenotĂpusos differenciálĂłdás mĂ©rtĂ©kĂ©nek összehasonlĂtása során vĂ©gzett Mantel teszt eredmĂ©nyei azt mutatták, hogy a szárny jellegeit tekintve szoros korreláciĂł van a differenciálĂłdás ezen kĂ©t szintje között. Ugyanakkor Ă©rdekes, hogy a variabilitás szintjĂ©hez hasonlĂłan a differenciálĂłdás mĂ©rtĂ©kĂ©ben sem tapasztaltam korreláciĂłt a genitália jellegei Ă©s az enzimpolimorfizmus között. Ráadásul a kĂ©t morfometriai jellegcsoport (szárny-, Ă©s genitáliajellegek) differenciálĂłdása sem korrelált egymással. A korreláciĂł hiánya arra utal, hogy eltĂ©rĹ‘ környezeti tĂ©nyezĹ‘k hatnak a kĂ©t morfolĂłgiai jellegcsoport fenotĂpusos varianciájára.
Species of the Mellicta subgenus are listed as vulnerable or declining in several countries of Europe. Therefore, these species are of obviously great conservation concern. The subgenus is taxonomically complicated due to the great variety of the species in their external characters. Even the existence of cryptic species has been suggested. The habitat requirements and food plants of Melitaea species partly overlap, they often co-occur within the same habitat. Melitaea species have fairly similar life cycle: females lay a great number of eggs in a few batches and their caterpillars remain together feeding gregariously. As a consequence, all offspring of a female might be affected by various environmental factors. Due to this special reproductive strategy populations of the Melitaea species are greatly exposed to stochastic events and genetic drift is likely to have a large effect on their genetic composition.
The studied three Melitaea species differ in voltinism. Hungarian populations of M. britomartis and M. aurelia have a single brood per year, while M. athalia has two ecotypes: one has one brood per year (monovoltine) and the other usually has two (bivoltine). Moreover larval development of M. athalia is slightly different compared to the other two. Caterpillars of this species spread before the diapause, feed and aestivate/hibernate solitarily. Furthermore, larvae of M. athalia can exploit a wider range of food plants than those of the other two Melitaea species.
In my surveys, three Melitaea species were studied and I have analysed the structure of genetic variation and the level of differentiation of these species in the Carpathian Basin. I was particularly interested in the level of differentiation among M. athalia populations with special regard to the two ecotypes of this species. Accordingly, I have analysed the pattern of genetic differentiation among M. athalia populations in the Carpathian Basin. In addition to the study of enzyme polymorphism I also surveyed the structure of morphometric variation using selected characteristics of wings and external genitalia. Thus, the structure of genetic and phenotypic variation was investigated in parallel. This approach allowed me to analyse the association between genetic and phenotypic variation in this species.
The results can be summarized in the following points:
Three distinct Evolutionary Significant Units (ESUs) were detected within the Mellicta subgenus in the Carpathian Basin corresponding to the three species. The results revealed that M. britomartis and M. aurelia seem to be closely related, while M. athalia appears to be a further relative. This result contradicts to those of Wahlberg and his coworkers’ surveys. In their tree, M. athalia and M. britomartis were closely related, while M. aurelia occurred in a neighbouring branch with other related species. It has to be mentioned, however, that their approach was entirely different: they used other markers to study the phylogeny of all species of the Melitaeini tribe using 1-3 individuals from each. None of my results supported the differentiation between the two ecotypes of M. athalia, that is, they cannot be considered as genetically separated groups. The apparent difference in the life cycle between the mono- and bivoltine ecotypes is probably the consequence of the high phenotypic plasticity of the species and it is probably the consequence of habitat quality. The results also indicated that M. athalia populations were highly differentiated from each other.
In parallel analyses of genetic and morphometric data I was able to describe the pattern of differentiation in M. athalia. All genetic analyses suggested a clear regional differentiation. Moreover, the regions could be separated to the East and West geographic zones of the Carpathian Basin. The two ecotypes of M. athalia were not separated in these analyses either implying that there are no genetic differences between them. Findings of the morphometric studies of wings and genital organs showed a regional pattern of differentiation of the phenotypic variation as well. Considering the characters measured on the wings, the separation between the regions of the East and West geographic zones of the Carpathian Basin was less clear than in the study of enzyme polymorphism. This West-East separation, however, was not detected in the characters measured on the genital structures. Accordingly, the results of my work indicated that the structure of genetic and phenotypic variation exhibited certain similarities in M. athalia, as regional pattern of differentiation was detected for both types of variation. The geographic pattern, however, proved to be more complex in the genetic analyses as the regions composed two clear groups corresponding to the East and West geographic zones of the Carpathian Basin. Considering our sampling strategy the presence of a genetic cline cannot be completely ruled out. Nevertheless, the results support the West-East separation of the populations more strongly.
Analysing the level of genetic variability in the Mellicta subgenus, high enzyme polymorphism was found in all three species. All studied 15 enzyme loci had alternative alleles; moreover most of them were highly polymorphic. The classidcal parameters of genetic variation were high in all three species, but the M. athalia seemed to be the most divers.
Since Melitaea species take place at the r side of the r-K continuum, stochastic events affect their populations to a great extent. Thus, genetic drift was expected resulting in low level of variability within the populations. As opposed to my expectations the results indicated an extremely high level of enzyme polymorphism. Looking for the explanation of this high variation the effect of two further possible factors was considered: (i) general ecology of the species especially the range of host plant utilization; (ii) the site of the studied populations relative to the distribution area of the species. Moreover, the genetic data of the three Mellicta species were compared to those of four other species of the Melitaeini tribus (Euphydryas maturna, E. aurinia, Melitaea phoebe and M. ornata). The results showed that the pattern of genetic variation in the seven species was best explained by the central-marginal hypothesis.
In the analysis of phenotypic variation first we compared the different morphometric characters. Surprisingly, the Levene-variables expressed as percentages of trait averages suggested that the variation of characters of external genitalia were higher than those of the wing traits. This result was unexpected, as it is generally known that wing traits are more affected by environmental factors, while the morphology of external genitalia is more determined by genetic factors.
In many surveys, the genetic structure of populations was reflected in the variability of morphological characters. Thus, we contrasted the phenotypic and genetic variation in the M. athalia populations studied. Both types of variation exhibited a geographic pattern; that is a more or less expressed regional differentiation was observed in the analysis of enzyme polymorphism and morphometric traits of the wings and external genitalia as well. Few significant differences were found in the phenotypic variation of the characters of the wings and genitalia between either the two ecotypes or the two zones of the Carpathian Basin. Nevertheless a tendency was found for several traits of the wings for higher variation in the populations of the East zone of the Carpathian Basin. This tendency was in accordance with that observed for enzyme polymorphism. In contrast, the traits of the genital organs showed an opposite tendency; i.e. the populations of the West zone of the Carpathian Basin seemed to be more variable than those of the East one.
Testing the correlation between the level of phenotypic and genetic variation at first sight I did not detect significant relationship between them neither for the traits of external genitalia nor for the wing characters. In a more detailed analysis, however, I found that the two types of wing traits (size+shape and pattern) are not correlated with each other. Accordingly, I handled the two types of wing characters separately in all further analyses, and found that variation in the size and shape of the wings which are less variable and less influenced by environmental factors is significantly correlated to the level of enzyme polymorphism. That is, phenotypic variation of these characters is a better indicator of genetic variation than that of wing pattern which is assumed to be more plastic. These results suggested that the phenotypic variance of the size and shape of the wings was mainly determined by genetic factors. Nevertheless, the level of morphometric variation of the traits of genitalia was not correlated to either the amount of enzyme polymorphism or to that of phenotypic variation in the size and shape of the wings.
I have used a series of Mantel tests to compare the level of genetic and morphometric differentiation among the M. athalia populations. The results indicated a relatively strong correlation between phenotypic differentiation estimated on the basis of the size and shape characters of the wings and genetic differentiation characterised by enzyme polymorphism. No correlation was found, however, between the level of genetic and phenotypic differentiation when the traits of external genitalia were used in the analyses. Moreover, phenotypic differentiation was not correlated either between the two types of morfometric traits. The lack of correlation between wing traits and those of the genitalia implies that different environmental factors influence them
