    Use of Large Lysimeters to Monitor Unsaturated Hydraulic Properties of Amended Soils

    The design and construction of large 1.21.2~mm diameter lysimeters has been implemented to monitor the soil water retention behaviour and permeability characteristics of contaminated soils under remediation. The work was carried out as part of a larger project focussing on the sustainable remediation of low value brownfield land. Three lysimeters have been filled with lead contaminated soil: one control; one with a \ac{WTR} amendment; and one with a \ac{WTR} and compost amendment. A new software system was built to control the \ac{TDR} point water content measurement and irrigation system, which could log data to an online unified data repository; provided an interface for connectivity to any serial port device; deal with templating for simplified setup; and realtime feedback for the end user. High capacity tensiometers were used in conjunction with the \ac{TDR} point water content measurement system to read volumetric water contents and suctions in the large control lysimeter over a series of wetting and drying cycles, each lasting several months. The results demonstrate that there was a difference between small scale laboratory tests and the data obtained from the lysimeters, particularly in the near surface soil due to cracking. Where cracking was not present, the agreement was stronger, but differences suggested that the drying curves in the lysimeter was predominantly scanning behaviour whereas the element tests were likely more representative of primary drying behaviour

    Longer-Run Effects of Antipoverty Policies on Disadvantaged Neighborhoods

    We estimate the longer-run effects of minimum wages, the Earned Income Tax Credit, and welfare on key economic indicators of economic self-sufficiency in disadvantaged neighborhoods. We find that the longer-run effects of the EITC are to increase employment and to reduce poverty and public assistance. We also find some evidence that higher welfare benefits had longer-run adverse effects, and quite robust evidence that tighter welfare time limits reduce poverty and public assistance in the longer run. The evidence on the long-run effects of the minimum wage on poverty and public assistance is not robust, with some evidence pointing to reductions and some to increases

    Residual stresses in condition monitoring and repair of thermal power generation components

    Residual stresses have a significant impact on fatigue and fracture of engineering components and structures, with an effect that is largely dependent on the sign of the residual stress relative to that of the applied stress, i.e. on whether they add to, or subtract from, the applied stress. The present paper will emphasise the importance of detailed knowledge of residual stresses to applications in thermal power generation. The context of the examples is condition monitoring and repair procedures where assessment of the influence of residual stress fields is important to both fatigue and fracture performance, and to certification of the repair procedure itself. The main conclusion in the paper is that the innovative use of solid-state friction taper hydro-pillar processes can offer additional capability in condition monitoring of through-thickness creep damage in thermal power plant, as well as provide cost-effective local repair of creep or fatigue damage in, for example, thick-walled steam pipe and blade-disc attachment holes

    Social Capital Determinants and Labor Market Networks

    We explore the links between determinants of social capital and labor market networks at the neighborhood level. We harness rich data taken from multiple sources, including matched employer-employee data with which we measure the strength of labor market networks, data on neighborhood homogeneity that has previously been tied to social capital, and new data – not previously used in the study of social capital – on the number and location of non-profit sector establishments at the neighborhood level. We use a machine learning algorithm to identify the potential determinants of social capital that best predict neighborhood-level variation in labor market networks. We find evidence suggesting that smaller and less centralized schools, and schools with fewer poor students, foster social capital that builds local labor market networks, as does a larger Republican vote share. The presence of establishments in a number of non-profit-oriented industries are identified as predictive of strong labor market networks, likely because they either provide public goods or facilitate social contacts. These industries include, for example, churches and other religious institutions, fire and rescue services including volunteer fire departments, country clubs and golf courses, labor unions, chamber music groups, hobby clubs, and schools

    During acute infectious mononucleosis (AIM), large clones of Epstein-Barr virus-specific T lymphocytes are produced. To investigate the dynamics of clonal expansion, we measured cell proliferation during AIM using deuterated glucose to label DNA of dividing cells in vivo, analyzing cells according to CD4, CD8 and CD45 phenotype. The proportion of labeled CD8(+)CD45R0(+) T lymphocytes was dramatically increased in AIM subjects compared to controls (mean 17.5 versus 2.8%/day; p<0.005), indicating very rapid proliferation. Labeling was also increased in CD4(+)CD45R0(+) cells (7.1 versus 2.1%/day; p<0.01), but less so in CD45RA(+) cells. Mathematical modeling, accounting for death of labeled cells and changing pool sizes, gave estimated proliferation rates in CD8(+)CD45R0(+) cells of 11-130% of cells proliferating per day (mean 47%/day), equivalent to a doubling time of 1.5 days and an appearance rate in blood of about 5 x 10(9) cells/day (versus 7 x 10(7) cells/day in controls). Very rapid death rates were also observed amongst labeled cells (range 28-124, mean 57%/day),indicating very short survival times in the circulation. Thus, we have shown direct evidence for massive proliferation of CD8(+)CD45R0(+) T lymphocytes in AIM and demonstrated that rapid cell division continues concurrently with greatly accelerated rates of cell disappearance

    1,2,6-thiadiazinones as novel narrow spectrum calcium/calmodulin-dependent protein kinase kinase 2 (CaMKK2) inhibitors

    We demonstrate for the first time that 4H-1,2,6-thiadiazin-4-one (TDZ) can function as a chemotype for the design of ATP-competitive kinase inhibitors. Using insights from a co-crystal structure of a 3,5-bis(arylamino)-4H-1,2,6-thiadiazin-4-one bound to calcium/calmodulin-dependent protein kinase kinase 2 (CaMKK2), several analogues were identified with micromolar activity through targeted displacement of bound water molecules in the active site. Since the TDZ analogues showed reduced promiscuity compared to their 2,4-dianilinopyrimidine counter parts, they represent starting points for development of highly selective kinase inhibitors
