Foi detectado maior concentração dessas substâncias nas células cultivadas in vitro.AMEIRA, O. A. [i.e. LAMEIRA, O. A.]. Disponível também on-line

Sites of Biosynthesis of Outer and Inner Membrane Proteins of Neurospora crassa Mitochondria
Outer and inner membranes of Neurospora crassa mitochondria were separated by the combined swelling, shrinking, sonication procedure. Membranes were characterized by electron microscopy and by marker enzyme activities. A red carotenoid pigment was found to be concentrated in the outer membrane. The inner mitochondrial membrane was resolved into about 20 protein bands on polyacrylamide gel electrophoresis, whereas the outer membrane shows essentially one single protein band. Only negligible incorporation of radioactive amino acids occurs into outer membrane when isolated mitochondria are synthesizing polypeptide chains. In agreement with this observation labeling of outer membrane protein is almost entirely blocked, when whole Neurospora cells are incubated with radioactive amino acids in the presence of cycloheximide, an inhibitor of cytoplasmic protein synthesis. Finally, the essential electrophoretic protein band from outer membrane does not become labeled when mitochondria are incubated with radioactive amino acids either in vitro or in vivo in the presence of cycloheximide. It is concluded that the vast majority, if not all, of the outer membrane protein is synthesized by the cytoplasmic system and that polypeptide chains formed by the mitochondrial ribosomes are integrated into the inner mitochondrial membrane
Theory of the density fluctuation spectrum of strongly correlated electrons
The density response function of the two-dimensional
model is studied starting from a mixed gauge formulation of the slave boson
approach. Our results for are in remarkable agreement with exact
diagonalization studies, and provide a natural explanation of the anomalous
features in the density response in terms of the spin polaron nature of the
charge carriers. In particular we have identified unexplained low energy
structures in the diagonalization data as arising from the coherent polaron
motion of holes in a spin liquid.Comment: 4 pages with 4 figures, to be published in Physical Review B (RC
Prune cAMP phosphodiesterase binds nm23-H1 and promotes cancer metastasis
We identify a new enzymatic activity underlying metastasis in breast cancer and describe its susceptibility to therapeutic inhibition. We show that human prune (h-prune), a phosphoesterase DHH family appertaining protein, has a hitherto unrecognized cyclic nucleotide phosphodiesterase activity effectively suppressed by dipyridamole, a phosphodiesterase inhibitor. h-prune physically interacts with nm23-H1, a metastasis suppressor gene. The h-prune PDE activity, suppressed by dipyridamole and enhanced by the interaction with nm23-H1, stimulates cellular motility and metastasis processes. Out of 59 metastatic breast cancer cases analyzed, 22 (37%) were found to overexpress h-prune, evidence that this novel enzymatic activity is involved in promoting cancer metastasis
Schwinger-boson approach to quantum spin systems: Gaussian fluctuactions in the "natural" gauge
We compute the Gaussian-fluctuation corrections to the saddle-point
Schwinger-boson results using collective coordinate methods. Concrete
application to investigate the frustrated J1-J2 antiferromagnet on the square
lattice shows that, unlike the saddle-point predictions, there is a quantum
nonmagnetic phase for 0.53 < J2/J1 < 0.64. This result is obtained by
considering the corrections to the spin stiffness on large lattices and
extrapolating to the thermodynamic limit, which avoids the infinite-lattice
infrared divergencies associated to Bose condensation. The very good agreement
of our results with exact numerical values on finite clusters lends support to
the calculational scheme employed.Comment: 4 pages, Latex, 3 figures included as eps files, minor correction
Compressibility of the Two-Dimensional infinite-U Hubbard Model
We study the interactions between the coherent quasiparticles and the
incoherent Mott-Hubbard excitations and their effects on the low energy
properties in the Hubbard model. Within the framework of a
systematic large-N expansion, these effects first occur in the next to leading
order in 1/N. We calculate the scattering phase shift and the free energy, and
determine the quasiparticle weight Z, mass renormalization, and the
compressibility. It is found that the compressibility is strongly renormalized
and diverges at a critical doping . We discuss the nature
of this zero-temperature phase transition and its connection to phase
separation and superconductivity.Comment: 4 pages, 3 eps figures, final version to appear in Phys. Rev. Let
Fermented soy-derived bioactive peptides selected by a molecular docking approach show antioxidant properties involving the keap1/nrf2 pathway
Bioactive peptides are a group of molecules with health beneficial properties, deriving from food matrices. They are protein fragments consisting of 2–20 amino acids that can be released by microbial fermentation, food processing and gastrointestinal digestion. Once hydrolyzed from their native proteins, they can have different functions including antioxidant activity, which is important for cell protection by oxidant agents. In this work, fermented soy products were digested in vitro in order to improve the release of bioactive peptides. These were extracted, purified and analyzed in vitro and in a cellular model to assess their antioxidant activity. Peptide sequences were identified by LC-MS/MS analysis and a molecular docking approach was used to predict their ability to interact with Keap1, one of the key proteins of the Keap1/Nrf2 pathway, the major system involved in redox regulation. Peptides showing a high score of interaction were selected and tested for their antioxidant properties in a cellular environment using the Caco-2 cell line and examined for their capability to defend cells against oxidative stress. Our results indicate that several of the selected peptides were indeed able to activate the Keap1/Nrf2 pathway with the consequent overexpression of antioxidant and phase II enzymes
