Effects of levonorgestrel on the ovarian cycle of Göttingen minipigs
Um das Göttinger Minischwein als Versuchstier fĂŒr endokrin-pharmakologische
Fragestellungen einsetzen zu können, mĂŒssen die pharmakologischen Wirkungen
der Substanzen auf den Zyklus der Tiere beurteilt werden können. In einem
Vorversuch wurden zunÀchst Methoden auf ihre Anwendbarkeit und Aussagekraft
geprĂŒft, das Zyklusgeschehen an sechs weiblichen Minischweinen zu
charakterisieren. Als geeignet erwiesen sich der natĂŒrliche Duldungsreflex,
die Rötung/Schwellung der Vulva, die Beurteilung von Vaginalschleim, die
Ultraschalluntersuchung der Ovarien sowie die Bestimmung der
Plasmakonzentrationen von LH, FSH, 17-Ă-Estradiol und Progesteron. In dem
nachfolgenden Hauptversuch wurden 12 weibliche Tiere (1 Kontrollgruppe, 3
Dosisgruppen) ĂŒber einen Zeitraum von 24 Tagen intramuskulĂ€r mit LNG, einem
synthetischen Gestagen, behandelt. Ziel der Behandlung war es, die Ovulation
der Tiere zu hemmen und die Schwellendosis fĂŒr eine Ovulationshemmung zu
ermitteln. Als niedrigste pharmakologisch wirksame Dosis fĂŒr eine
Ovulationshemmung wurde 0,02 mg/kg/Tag LNG ermittelt. In der 0,002 mg/kg/Tag
Dosisgruppe waren durch das Verhalten und die VerĂ€nderungen am Ă€uĂeren Genital
keine eindeutigen Brunstsymptome zu erkennen. Eine Ovulation konnte jedoch
durch die Ultraschalluntersuchung, die Histologie der Ovarien sowie die
Messung der Hormonkonzentrationen nach Ende des Versuchs bestÀtigt werden. In
der 0,02 mg/kg/Tag und der 0,1 mg/kg/Tag LNG Dosisgruppe zeigten sowohl das
Verhalten der Tiere wie auch alle ĂŒbrigen Parameter eine eindeutige
Unterbrechung des Sexualzyklus an, dass keine Ovulation stattfand. Dabei
traten in der mittleren und hohen Dosisgruppe auĂer am Ovar auch
substanzbedingte Effekte an Vagina und Uterus sowie bei den Organgewichten
auf. Diese Ergebnisse korrelierten mit bereits bekannten Wirkungen bei anderen
Spezies, wie z. B. der Ratte. Das Tiermodell des Göttinger Minischweins erwies
sich daher als sehr geeignet, pharmakologische Hormonwirkungen auf den
Sexualzyklus des Göttinger Minischweins zu erfassen. Dies traf ebenfalls auf
die Auswahl der Methoden zu, die eine zusÀtzliche Möglichkeit boten, gestagene
Wirkungen von Hormonen und anderen Substanzen in toxikologischen Studien zu
untersuchen.To be able to use the Göttingen minipig as an experimental animal model for
endocrinological and pharmacological objectives, it must be possible to
evaluate the pharmacological effect of a substance on the ovarian cycle of the
animals. In a preliminary study, methods were tested for their practicability
and significance to characterize the cycle by using six female minipigs.
Suitable proved to be the natural toleration reflex, the redness/swelling of
the vulva, the evaluation of the vaginal mucus, the ultrasound examination of
the ovaries as well as the determination of the plasma concentrations of
luteinizing hormone, follicle stimulating hormone, 17-Ă-estradiol and
progesterone. In a main study, 12 female animals (3 treatment groups, 1
control group) were treated intramuscularly for 24 days with levonorgestrel
(LNG), a synthetic gestagen. The aim was to inhibit the ovulation of the
animals and to determine the threshold dose for the inhibition of ovulation.
The lowest pharmacologically effective dose for the inhibition of ovulation
was determined to be 0.02 mg/kg/day LNG. In the 0.002 mg/kg/day dose group, no
clear estrus symptoms were evident on the outer genital. However, ovulation
was confirmed by an ultrasound examination as well as the hormone
concentrations at the end of the study. In the 0.02 mg/kg/day and 0.1
mg/kg/day dose groups, both behavior and all other measured parameters showed
a clear interruption of the sexual cycle, and no ovulation occurred. Animals
in both the mid- and high dose groups showed substance related effects, not
only on the ovary but also on the vagina, uterus, and the organ weights. These
results correlated with known effects observed in other species, e.g. the rat.
Overall, the Göttinger minipig proved to be a very suitable animal model to
test pharmacological hormone effects on the sexual cycle of the minipig. This
was also applicable to the choice of methods, which offered an additional
possibility to evaluate the gestagenic effects of hormones and other
substances in toxicology studies
