    Seed Production of \u3cem\u3eBrachiaria ruziziensis\u3c/em\u3e in India--Seed Collection Methods and Feed Opportunities

    Brachiaria ruziziensis is an important fodder crop suitable for high rainfall areas and soils with low nutrient supply. Though the area under B. ruziziensis cultivation in India is not properly documented, it is widely grown in Kerala (Stür et al. 1996) and in parts of Karnataka and Goa. By virtue of its shade tolerance and adaptability, B. ruziziensis have wide scope for adoption in other parts of India. In India, Brachiaria is mainly planted through root slips as availability of good quality filled seeds is very less. It is known that the proportion of filled to unfilled seeds depends on method of harvest and seed collection method (Hare et al., 2007). Appropriate method of harvest depends on growth habit, synchrony of crop development, standing seeds, fallen seeds, availability of labour and on previous experience. Hence a study was conducted for first time in India to standardise harvesting and seed collection method in B.ruziziensis

    Mediator Subunits MED16, MED14, and MED2 Are Required for Activation of ABRE-Dependent Transcription in Arabidopsis

    The Mediator complex controls transcription of most eukaryotic genes with individual subunits required for the control of particular gene regulons in response to various perturbations. In this study, we reveal the roles of the plant Mediator subunits MED16, MED14, and MED2 in regulating transcription in response to the phytohormone abscisic acid (ABA) and we determine which cis elements are under their control. Using synthetic promoter reporters we established an effective system for testing relationships between subunits and specific cis-acting motifs in protoplasts. Our results demonstrate that MED16, MED14, and MED2 are required for the full transcriptional activation by ABA of promoters containing both the ABRE (ABA-responsive element) and DRE (drought-responsive element). Using synthetic promoter motif concatamers, we showed that ABA-responsive activation of the ABRE but not the DRE motif was dependent on these three Mediator subunits. Furthermore, the three subunits were required for the control of water loss from leaves but played no role in ABA-dependent growth inhibition, highlighting specificity in their functions. Our results identify new roles for three Mediator subunits, provide a direct demonstration of their function and highlight that our experimental approach can be utilized to identify the function of subunits of plant transcriptional regulators

    Salt Tolerance Mechanisms in Perennial Fodder Grasses

    Salinity stress is one of the most damaging stresses in crop plants. It reduces the productivity of the soil and makes it unsuitable for crop cultivation. Fodder crops are considered the best alternative in such uncultivable land. Using salinity-affected land for pasture development is the best alternative to utilize such lands. However, the extent of salinity tolerance varies among different grass species. In this study, Pearl millet Napier hybrids (PMN hybrid) and guinea grass varieties were studied for salinity tolerance in artificially created saline soils in the ratio of 13:7:1:2 (NaCl: Na2SO4: MgCl: CaSO4, respectively) to understand the salinity tolerance mechanisms existing in perennial fodder grasses. Morphologically, the plant height increased in saline-tolerant PMN hybrid varieties, creating more space in nonphotosynthetic tissues to store accumulated salts away from photosynthetic tissues. Whereas in guinea grass tolerant varieties, tiller number increased under salinity. The fresh weight was highest under salinity in the PMN hybrids. In contrast, dry weight was high in control (no salt) plants, implying more water accumulation in PMN hybrids under salinity to dilute the concentration of salts absorbed by the plant. In Guinea grass, varieties like DGG1 had lower leaf succulence than control and high salt excretion through leaf hairs. Tissue tolerance in PMN hybrids was less compared to guinea grass. Membrane stability was maintained in saline-tolerant varieties. The double bond index increased in tolerant PMN hybrid varieties under salinity compared to control, implying fatty acid remodelling for maintaining the stability of membranes under salinity.     Consumer willingness to pay for a hypothetical Chagas disease vaccine in Brazil : a cross-sectional study and the implications

    Aim: Chagas disease is a serious public health problem, endemic in 21 countries in Latin America. A future vaccine can contribute to decreasing the number of cases and its complications. Methods: A cross-sectional study was conducted with residents of the northern region of Brazil, on the willingness to pay for a hypothetical vaccine against Chagas disease (effective protection of 80%). Results: We interviewed 619 individuals and seven were excluded from the analysis and the value of willingness to pay was US$23.77 (100.00 BRL). Conclusion: The Northern region of Brazil is one of the largest markets for this vaccine, due to its epidemiological relevance, so economic studies with this vaccine will be important to assist in the assessment of technologies

    Independent and combined effects of improved water, sanitation, and hygiene, and improved complementary feeding, on child stunting and anaemia in rural Zimbabwe: a cluster-randomised trial.

    BACKGROUND: Child stunting reduces survival and impairs neurodevelopment. We tested the independent and combined effects of improved water, sanitation, and hygiene (WASH), and improved infant and young child feeding (IYCF) on stunting and anaemia in in Zimbabwe. METHODS: We did a cluster-randomised, community-based, 2 × 2 factorial trial in two rural districts in Zimbabwe. Clusters were defined as the catchment area of between one and four village health workers employed by the Zimbabwe Ministry of Health and Child Care. Women were eligible for inclusion if they permanently lived in clusters and were confirmed pregnant. Clusters were randomly assigned (1:1:1:1) to standard of care (52 clusters), IYCF (20 g of a small-quantity lipid-based nutrient supplement per day from age 6 to 18 months plus complementary feeding counselling; 53 clusters), WASH (construction of a ventilated improved pit latrine, provision of two handwashing stations, liquid soap, chlorine, and play space plus hygiene counselling; 53 clusters), or IYCF plus WASH (53 clusters). A constrained randomisation technique was used to achieve balance across the groups for 14 variables related to geography, demography, water access, and community-level sanitation coverage. Masking of participants and fieldworkers was not possible. The primary outcomes were infant length-for-age Z score and haemoglobin concentrations at 18 months of age among children born to mothers who were HIV negative during pregnancy. These outcomes were analysed in the intention-to-treat population. We estimated the effects of the interventions by comparing the two IYCF groups with the two non-IYCF groups and the two WASH groups with the two non-WASH groups, except for outcomes that had an important statistical interaction between the interventions. This trial is registered with ClinicalTrials.gov, number NCT01824940. FINDINGS: Between Nov 22, 2012, and March 27, 2015, 5280 pregnant women were enrolled from 211 clusters. 3686 children born to HIV-negative mothers were assessed at age 18 months (884 in the standard of care group from 52 clusters, 893 in the IYCF group from 53 clusters, 918 in the WASH group from 53 clusters, and 991 in the IYCF plus WASH group from 51 clusters). In the IYCF intervention groups, the mean length-for-age Z score was 0·16 (95% CI 0·08-0·23) higher and the mean haemoglobin concentration was 2·03 g/L (1·28-2·79) higher than those in the non-IYCF intervention groups. The IYCF intervention reduced the number of stunted children from 620 (35%) of 1792 to 514 (27%) of 1879, and the number of children with anaemia from 245 (13·9%) of 1759 to 193 (10·5%) of 1845. The WASH intervention had no effect on either primary outcome. Neither intervention reduced the prevalence of diarrhoea at 12 or 18 months. No trial-related serious adverse events, and only three trial-related adverse events, were reported. INTERPRETATION: Household-level elementary WASH interventions implemented in rural areas in low-income countries are unlikely to reduce stunting or anaemia and might not reduce diarrhoea. Implementation of these WASH interventions in combination with IYCF interventions is unlikely to reduce stunting or anaemia more than implementation of IYCF alone.     Genomic epidemiology of SARS-CoV-2 in a UK university identifies dynamics of transmission

    AbstractUnderstanding SARS-CoV-2 transmission in higher education settings is important to limit spread between students, and into at-risk populations. In this study, we sequenced 482 SARS-CoV-2 isolates from the University of Cambridge from 5 October to 6 December 2020. We perform a detailed phylogenetic comparison with 972 isolates from the surrounding community, complemented with epidemiological and contact tracing data, to determine transmission dynamics. We observe limited viral introductions into the university; the majority of student cases were linked to a single genetic cluster, likely following social gatherings at a venue outside the university. We identify considerable onward transmission associated with student accommodation and courses; this was effectively contained using local infection control measures and following a national lockdown. Transmission clusters were largely segregated within the university or the community. Our study highlights key determinants of SARS-CoV-2 transmission and effective interventions in a higher education setting that will inform public health policy during pandemics.</jats:p

    Effect of angiotensin-converting enzyme inhibitor and angiotensin receptor blocker initiation on organ support-free days in patients hospitalized with COVID-19

    IMPORTANCE Overactivation of the renin-angiotensin system (RAS) may contribute to poor clinical outcomes in patients with COVID-19. Objective To determine whether angiotensin-converting enzyme (ACE) inhibitor or angiotensin receptor blocker (ARB) initiation improves outcomes in patients hospitalized for COVID-19. DESIGN, SETTING, AND PARTICIPANTS In an ongoing, adaptive platform randomized clinical trial, 721 critically ill and 58 non–critically ill hospitalized adults were randomized to receive an RAS inhibitor or control between March 16, 2021, and February 25, 2022, at 69 sites in 7 countries (final follow-up on June 1, 2022). INTERVENTIONS Patients were randomized to receive open-label initiation of an ACE inhibitor (n = 257), ARB (n = 248), ARB in combination with DMX-200 (a chemokine receptor-2 inhibitor; n = 10), or no RAS inhibitor (control; n = 264) for up to 10 days. MAIN OUTCOMES AND MEASURES The primary outcome was organ support–free days, a composite of hospital survival and days alive without cardiovascular or respiratory organ support through 21 days. The primary analysis was a bayesian cumulative logistic model. Odds ratios (ORs) greater than 1 represent improved outcomes. RESULTS On February 25, 2022, enrollment was discontinued due to safety concerns. Among 679 critically ill patients with available primary outcome data, the median age was 56 years and 239 participants (35.2%) were women. Median (IQR) organ support–free days among critically ill patients was 10 (–1 to 16) in the ACE inhibitor group (n = 231), 8 (–1 to 17) in the ARB group (n = 217), and 12 (0 to 17) in the control group (n = 231) (median adjusted odds ratios of 0.77 [95% bayesian credible interval, 0.58-1.06] for improvement for ACE inhibitor and 0.76 [95% credible interval, 0.56-1.05] for ARB compared with control). The posterior probabilities that ACE inhibitors and ARBs worsened organ support–free days compared with control were 94.9% and 95.4%, respectively. Hospital survival occurred in 166 of 231 critically ill participants (71.9%) in the ACE inhibitor group, 152 of 217 (70.0%) in the ARB group, and 182 of 231 (78.8%) in the control group (posterior probabilities that ACE inhibitor and ARB worsened hospital survival compared with control were 95.3% and 98.1%, respectively). CONCLUSIONS AND RELEVANCE In this trial, among critically ill adults with COVID-19, initiation of an ACE inhibitor or ARB did not improve, and likely worsened, clinical outcomes.     Not AvailableAn evaluation was conducted in eastern India over three years, 2005–2007, to compare the performance of certain System of Rice Intensification (SRI) practices: transplanting single, young (10-day-old) seedlings in a square pattern; no continuous flooding; and use of a mechanical weeder – with those currently endorsed by the Central Rice Research Institute of India, referred to here as recommended management practices (RMP). All plots received the same fertilization, a combination of organic and inorganic nutrients, and the SRI spacing used was 20% less than usually recommended. Accordingly, the results reported here are designated as a modification of SRI recommendations (SRIm). The objective of this research was to understand the benefits in terms of yield and other physiological parameters, if any, from using most if not all recommended SRI practices compared to RMP. These selected SRI practices out-yielded RMP by 42%, with the higher yield associated with various phenotypical alterations, which are reported here. Significant measurable changes were observed in physiological processes and plant characteristics, such as longer panicles, more grains panicle−1 and higher % of grain-filling. The decreased plant density with SRIm management was compensated for by increased per-plant productivity. SRIm hills with single plants were found to have deeper and better-distributed root systems, higher xylem exudation rates, more open plant architecture with more erect and larger leaves, and more tillers than did RMP hills having multiple plants. Due to the reduction in number of hills m−2 in SRIm plots compared to RMP, no significant difference was found in root dry weight or leaf number, tillers or panicle number on an area basis. Nevertheless, in spite of SRIm having fewer hills and fewer tillers per unit area, the leaf area index (LAI) with SRIm practice was greater due to larger leaves. These together with altered plant architecture, contributed to more light interception by SRIm plants. The higher leaf chlorophyll content at ripening stage reflected delayed senescence and the greater fluorescence efficiency (Fv/Fm and PS II) associated with SRIm practices contributed to more efficient utilization of light and a higher rate of photosynthesis, which was probably responsible for the observed increase in grain filling and heavier grains compared to RMP plants. The higher photosynthesis rate coupled with lower transpiration in SRIm plants indicated that they were using water more efficiently than did RMP plants. The latter produced 1.6 μ mol CO2 fixed per m mol water transpired, compared to 3.6 μ mol CO2 in SRIm plants.Not Availabl
