Otimização e avaliação do extrato hidroalcóolico de Schinus terebinthifolius Raddi obtido por ultrasom.
Schinus terebinthifolius
Raddi is a plant family Anacardiaceae, native to Brazil. Have
been isolated from this plant several secondary metabolites: terpenes, lectins,
saponins, tannins, phenolics, flavonoi
ds and alkaloids, responsible for various
biological
activities:
antibacterial,
antiviral,
antifungal,
anti
-
inflammatory,
anticarcinogenic, antihypertensive and hypolipidemic. The objective is to evaluate and
optimize the extraction conditions of active su
bstances present in leaves of Schinus
terebinthifolius Raddi ultrasound. The leaves of Schinus terebinthifolius Raddi (100 g)
were placed in ethanol at 50, 70, 80 and 96% for extraction by ultrasound. The extracts
were analyzed by GC
-
FID and had a higher c
oncentration of
α
-
pinene (0.0225 mL /
mL) and limonene (0.1475 mL / mL) in the alcohol content of 80% and 70%
respectively. These extracts were evaluated by physical
-
chemical parameters of pH
(4.85, 4.96, 5.36 and 5.79), dry (0.0546 g, 0.0653 g, 1.2074 g a
nd 1.0501 g), density
(0.9253, 0.9185, 0.9037 and 0.8546) and refractive index at 20 ° C (1.3620, 1.3622,
1.3653 and 1.3657), respectively. Most obtaining the dry residue was extracted in 80%
ethanol. We use this alcoholic with different proportions of the
drug (10, 20 and 30%)
and obtained the highest concentration of
α
-
pinene (0.0037 mL / mL) and limonene
(0.0473 mL / mL) in 30%. To optimize the extraction conducted an extraction kinetics in
proportion and alcohol content for a period of 5 to 60 minutes. The best time to extract
the
α
-
pinene (0.02024 mL / mL) and limon
ene (0.15911 mL / ml) was 10 minutes.
Knowledge of the extraction conditions and the chemical behavior of the constituents of
a plant represents a key step in the evaluation of an extract.Schinus terebinthifolius

Avaliação do efeito da ouabaína na viabilidade e modulação de citocinas em timócitos e linfócitos expostos a radiação ultravioleta
Ouabain is a cardiac glycoside discovered in human plasma in 1991 (HAMLYN et al., 1991). This endogenous substance can inhibit Na + / K + -ATPase and has been extensively studied for its ability to interfere with various mechanisms that regulate and maintain homeostasis. Thus, the aim of this study was to evaluate in vitro and in vivo ouabain regarding cell viability and the profile of pro-inflammatory cytokines in thymus and mesenteric lymph node cells exposed to UV radiation. Therefore, Swiss albinus mice were used conducting cell culture from the thymus and the mesenteric lymph node. These cells were treated with ouabain 100, 10, 1 µM and 100 nM in a plaque of 96 wells, and also with a pre-treatment with ouabain 0.56 mg/kg during three days via intraperitoneal injection. The cells obtained from the maceration of the organs were centrifuged, resuspended and counted. The concentration was adjusted to 4x105céls/ml with PBS and plated for exposure to UVC and UVB for 2 minutes. After the stimuli, the RPMI medium supplemented with 10% of SFB is added and cells are kept in culture for 6 to 24 hours. Viability was analysed with the MTT 5 mg/mL and with the pro-inflammatory cytokines TNF-a and IL-6, as determined by the method ELISA. Our results showed that after 6h and 24h the culture in the presence of ouabain in different concentrations 100,10, 1 µM and100 nM, only in the 100 nM the cells were protected from cell death, this result was observed for the 24h culture of thymocytes and lymphocytes that were stimulated by UVC, but this was not observed for UVB stimulation. In these cells, the pre-treatment with 0.56 mg/Kg ouabain in the 6 hours culture presented a cytoprotective effect on the thymocytes and mesenteric lymph node lymphocytes exposed to UVC and UVB. Besides these effects on viability, ouabain in the concentration of 100 nM increased TNF-a and reduced IL-6. Nevertheless, the pre-treatment with 0.56 mg/Kg did not modulate these cytokines compared to the group receiving radiation. These results contribute to the understanding of the ouabain action over important parameters of the inflammatory process, viability and cytokines, triggered by ultraviolet radiation.NenhumaA ouabaína é um glicosídeo cardíaco que foi descoberto no plasma humano em 1991 (HAMLYN et al., 1991). Essa substância endógena é capaz de inibir a Na+/K+-ATPase e vem sendo amplamente estudada por sua capacidade em interferir em vários mecanismos que regulam e mantém a homeostase. Portanto o objetivo desse trabalho foi avaliar in vitro e in vivo a ação da ouabaína na viabilidade celular e no perfil de citocinas pró-inflamatórias em células do timo e linfonodo mesentérico expostas a radiação UV. Para isso foram usados camundongos Swiss albinus e realizada a cultura de células do timo e linfonodo mesentérico. Essas células foram tratadas com oubaína nas concentrações de 100,10, 1 µM e 100 nM na placa de 96 poços, como também através do pré-tratamento com ouabaína na dose de 0,56 mg/kg por três dias através da injeção intra-peritoneal dessa dose. As células obtidas da maceração dos órgãos foram centrifugadas, resuspensas e contadas. A concentração foi ajustada para 4x105céls/ml com PBS e plaqueadas para a exposição a UVC e UVB por 2 minutos. Após o estímulo, adicionou-se o meio RPMI suplementado com 10% de SFB e manteve-se essas células em cultura por 6 e 24 horas. A viabilidade foi analisada com o MTT 5 mg/mL. O sobrenadante da cultura foi utilizado para determinar os níveis das citocinas próinflamatórias TNF-a e IL-6 pelo método de ELISA. Nossos resultados mostraram que o prétratamento com ouabaína na dose de 0,56 mg/Kg na cultura de 6 horas apresentou um efeito citoprotetor nos timócitos e linfócitos linfonodo mesentérico expostos a UVC e UVB. Esse mesmo tipo de célula em cultura por 6h e 24h na presença de ouabaína em diferentes concentrações 100,10, 1 µM e 100 nM, apresentou efeito em 24 horas mediante o estímulo da UVC, esse dado não foi observado para UVB. A concentração de 100 nM protegeu os timócitos e linfócitos do linfonodo mesentérico da morte celular. No sobrenadante dessa concentração houve aumento do TNF-a e redução de IL-6 quando, e no pré-tratamento com 0,56 mg/Kg não houve alteração dessas citocinas comparada ao grupo que recebeu radiação. Esses resultados contribuem para o entendimento da sua ação ouabaína sobre parâmetros importantes do processo inflamatório, viabilidade e citocinas, desencadeado por radiação ultravioleta
Ouabain Modulates Zymosan-Induced Peritonitis in Mice
Ouabain, a potent inhibitor of the Na+, K+-ATPase, was identified as an endogenous substance. Recently, ouabain was shown to affect various immunological processes. We have previously demonstrated the ability of ouabain to modulate inflammation, but little is known about the mechanisms involved. Thus, the aim of the present work is to evaluate the immune modulatory role of ouabain on zymosan-induced peritonitis in mice. Our results show that ouabain decreased plasma exudation (33%). After induction of inflammation, OUA treatment led to a 46% reduction in the total number of cells, as a reflex of a decrease of polymorphonuclear leukocytes, which does not appear to be due to cell death. Furthermore, OUA decreased TNF-α (57%) and IL-1β (58%) levels, without interfering with IL-6 and IL-10. Also, in vitro experiments show that ouabain did not affect endocytic capacity. Moreover, electrophoretic mobility shift assay (EMSA) shows that zymosan treatment increased (85%) NF-κB binding activity and that ouabain reduced (30%) NF-κB binding activity induced by zymosan. Therefore, our data suggest that ouabain modulated acute inflammatory response, reducing the number of cells and cytokines levels in the peritoneal cavity, as well as NFκB activation, suggesting a new mode of action of this substance
