    Equine-Assisted Interventions (EAIs) for Children with Autism Spectrum Disorders (ASD): Behavioural and Physiological Indices of Stress in Domestic Horses (Equus caballus) during Riding Sessions

    Equine-assisted interventions (EAIs) are well-known complementary practices combining physical activity with emotional/cognitive stimulation. They are especially suited for children with autism spectrum disorders (ASD) who need a high degree of physical and psychological enrichment. Even though EAIs have become a common practice, stress responses in horses interacting with individuals that can manifest inappropriate behaviours, such as ASD children, have not been thoroughly investigated. Our multicentre study aimed to investigate behavioural and physiological indices of stress in horses involved in EAI standardised sessions with children with ASD compared to typically developing (TD) children. A controlled within-subject design with repeated measurements involving 19 horses and 38 children was adopted. Stress-related behaviours, heart rate, heart rate variability, and eye temperature were recorded during the riding sessions. Moreover, blood samples were collected from horses before and after each session to monitor changes in blood adrenocorticotropic hormone (ACTH), cortisol, and catecholamines. Results indicate that, in general, stress responses in horses involved in EAIs did not differ as a function of the horse being ridden by children with ASD or TD. A lower sympathetic tone in horses involved in ASD sessions was found, while in the mounting and dismounting phases, horses displayed behavioural signs of stress, independently from children’s behaviour. We conclude that professionals working in EAI should increase their awareness of animal welfare and refine riding practices, taking into account horse’s needs

    Oxidative stability of food is one of the most important parameters affecting integrity and consequently nutritional properties of dietary constituents. Antioxidants are widely used to avoid deterioration during transformation, packaging, and storage of food. In this paper, novel poly (vinyl alcohol) (PVA)-based films were prepared by solvent casting method adding an hydroxytyrosol-enriched extract (HTyrE) or an oleuropein-enriched extract (OleE) in different percentages (5, 10 and 20% w/w) and a combination of both at 5% w/w. Both extracts were obtained from olive oil wastes and by-products using a sustainable process based on membrane technologies. Qualitative and quantitative analysis of each sample carried out by high performance liquid chromatography (HPLC) and nuclear resonance magnetic spectroscopy (NMR) proved that the main components were hydroxytyrosol (HTyr) and oleuropein (Ole), respectively, two well-known antioxidant bioactive compounds found in Olea europaea L. All novel formulations were characterized investigating their morphological, optical and antioxidant properties. The promising performances suggest a potential use in active food packaging to preserve oxidative-sensitive food products. Moreover, this research represents a valuable example of reuse and valorization of agro-industrial wastes and by-products according to the circular economy model

    ESTABLISHMENT AND CHARACTERIZATION OF A CANCER STEM CELL LINE FROM A RARE BONE SARCOMA

    Objective: To establish of a bone cancer stem cell (CSC) line from a rare human bone disease, the high grade osteoblastic osteosarcoma (OS). Material and Methods: Central high grade osteoblastic OS samples were collected at the “Unit Ortopedia Oncologica e Ricostruttiva”, AOUC Careggi, Florence, with informed consent approved by the local Ethical Committee. First, primary human cancer cell cultures of osteoblastic OS have been established. After that, the subpopulations of CSCs have been isolated from these, with the sphere-formation assay. Hence, the cancer stem cell phenotype has been evaluated by several cellular assays/stainings, by flow cytometry analysis and by analyzing the pro-files expressions of a few genes related to CSCs. Results: We have established primary cell cultures of high grade osteoblastic OS from each samples collected. Consequently, from these we have started to isolate CSCs. Nowadays we have established a OS-CSC line, called as OSA5-CSC. The cancer stem cell phenotype from OSA5-CSC line was confirmed by observing the capacity of the OSA5-CSCs to differentiate into osteoblasts and into adipocytes, by showing the positive presence of the MSCs and ofthe ESCs markers into the cell line, and by evaluating a good rate (14 %) as clonogenic capacity. The presence of the ESC markers was confirmed also by their gene expression together with the expression of CD133. On the other hand, the malignant phenotype was verify by demonstrating the presence of the expression of genes involved in invasion/migration process (i.e., AXL and EZR) and in the pluripotency of CSC (i.e., MYC), by the agar soft and bythe ALDH1A1 assays. Conclusions: In conclusion, we have collected a few biopsies of this rare bone disease. We have established primary cell cultures from these. We have completely characterized and studied a OS-CSC line at cellular and molecular level. Nowadays we are concluding the characterization of the OS-CSCs isolated from the relative cell cultures and we are starting the study of micro-RNAs related to their phenotype

    Intubation Practices and Adverse Peri-intubation Events in Critically Ill Patients from 29 Countries

    Importance: Tracheal intubation is one of the most commonly performed and high-risk interventions in critically ill patients. Limited information is available on adverse peri-intubation events. Objective: To evaluate the incidence and nature of adverse peri-intubation events and to assess current practice of intubation in critically ill patients. Design, Setting, and Participants: The International Observational Study to Understand the Impact and Best Practices of Airway Management in Critically Ill Patients (INTUBE) study was an international, multicenter, prospective cohort study involving consecutive critically ill patients undergoing tracheal intubation in the intensive care units (ICUs), emergency departments, and wards, from October 1, 2018, to July 31, 2019 (August 28, 2019, was the final follow-up) in a convenience sample of 197 sites from 29 countries across 5 continents. Exposures: Tracheal intubation. Main Outcomes and Measures: The primary outcome was the incidence of major adverse peri-intubation events defined as at least 1 of the following events occurring within 30 minutes from the start of the intubation procedure: cardiovascular instability (either: systolic pressure <65 mm Hg at least once, <90 mm Hg for >30 minutes, new or increase need of vasopressors or fluid bolus >15 mL/kg), severe hypoxemia (peripheral oxygen saturation <80%) or cardiac arrest. The secondary outcomes included intensive care unit mortality. Results: Of 3659 patients screened, 2964 (median age, 63 years; interquartile range [IQR], 49-74 years; 62.6% men) from 197 sites across 5 continents were included. The main reason for intubation was respiratory failure in 52.3% of patients, followed by neurological impairment in 30.5%, and cardiovascular instability in 9.4%. Primary outcome data were available for all patients. Among the study patients, 45.2% experienced at least 1 major adverse peri-intubation event. The predominant event was cardiovascular instability, observed in 42.6% of all patients undergoing emergency intubation, followed by severe hypoxemia (9.3%) and cardiac arrest (3.1%). Overall ICU mortality was 32.8%. Conclusions and Relevance: In this observational study of intubation practices in critically ill patients from a convenience sample of 197 sites across 29 countries, major adverse peri-intubation events - in particular cardiovascular instability - were observed frequently
