    The Role of Self-Focused Cognition in Emotion Regulation

    The present dissertation reports a set of three studies that sought to characterize the effects of self-focused cognition on emotion regulation, specifically, cognitive reappraisal. Across the three studies, I investigated the effects of self-distancing, disengagement of self-focused thought, and changing the content of self-focused thought on multiple measures of emotion regulation success and emotion regulation difficulty. Results broadly suggested that disengaging self-focused cognition provides distinct advantages for emotion regulation, which are independent of effects on emotional reactivity. Specifically, I observed that other-focused cognition resulted in equally successful, but less difficult emotion regulation, the ability to more quickly disengage from self-focused thought was associated with greater emotion regulation success, and a greater tendency towards engaging in self-focused thought was associated with increased emotion regulation difficulty. I discuss the possible mechanisms explaining these effects, their specific implications for the study of emotion regulation, as well as their broader implications for the study of self-regulation

    Assessment of textural differentiations in forest resources in Romania using fractal analysis

    Deforestation and forest degradation have several negative effects on the environment including a loss of species habitats, disturbance of the water cycle and reduced ability to retain CO2, with consequences for global warming. We investigated the evolution of forest resources from development regions in Romania affected by both deforestation and reforestation using a non-Euclidean method based on fractal analysis. We calculated four fractal dimensions of forest areas: the fractal box-counting dimension of the forest areas, the fractal box-counting dimension of the dilated forest areas, the fractal dilation dimension and the box-counting dimension of the border of the dilated forest areas. Fractal analysis revealed morpho-structural and textural differentiations of forested, deforested and reforested areas in development regions with dominant mountain relief and high hills (more forested and compact organization) in comparison to the development regions dominated by plains or low hills (less forested, more fragmented with small and isolated clusters). Our analysis used the fractal analysis that has the advantage of analyzing the entire image, rather than studying local information, thereby enabling quantification of the uniformity, fragmentation, heterogeneity and homogeneity of forests

    Recent Deforestation Pattern Changes (2000-2017) in the Central Carpathians:A Gray-Level Co-Occurrence Matrix and Fractal Analysis Approach

    The paper explores the distribution of tree cover and deforested areas in the Central Carpathians in the central-east part of Romania, in the context of the anthropogenic forest disturbances and sustainable forest management. The study aims to evaluate the spatiotemporal changes in deforested areas due to human pressure in the Carpathian Mountains, a sensitive biodiverse European ecosystem. We used an analysis of satellite imagery with Landsat-7 Enhanced Thematic Mapper Plus (Landsat-7 ETM+) from the University of Maryland (UMD) Global Forest Change (GFC) dataset. The workflow started with the determination of tree cover and deforested areas from 2000–2017, with an overall accuracy of 97%. For the monitoring of forest dynamics, a Gray-Level Co-occurrence Matrix analysis (Entropy) and fractal analysis (Fractal Fragmentation-Compaction Index and Tug-of-War Lacunarity) were utilized. The increased fragmentation of tree cover (annually 2000–2017) was demonstrated by the highest values of the Fractal Fragmentation-Compaction Index, a measure of the degree of disorder (Entropy) and heterogeneity (Lacunarity). The principal outcome of the research reveals the dynamics of disturbance of tree cover and deforested areas expressed by the textural and fractal analysis. The results obtained can be used in the future development and adaptation of forestry management policies to ensure sustainable management of exploited forest areas

    REQUITE: A prospective multicentre cohort study of patients undergoing radiotherapy for breast, lung or prostate cancer

    Purpose: REQUITE aimed to establish a resource for multi-national validation of models and biomarkers that predict risk of late toxicity following radiotherapy. The purpose of this article is to provide summary descriptive data. Methods: An international, prospective cohort study recruited cancer patients in 26 hospitals in eight countries between April 2014 and March 2017. Target recruitment was 5300 patients. Eligible patients had breast, prostate or lung cancer and planned potentially curable radiotherapy. Radiotherapy was prescribed according to local regimens, but centres used standardised data collection forms. Pre-treatment blood samples were collected. Patients were followed for a minimum of 12 (lung) or 24 (breast/prostate) months and summary descriptive statistics were generated. Results: The study recruited 2069 breast (99% of target), 1808 prostate (86%) and 561 lung (51%) cancer patients. The centralised, accessible database includes: physician-(47,025 forms) and patient-(54,901) reported outcomes; 11,563 breast photos; 17,107 DICOMs and 12,684 DVHs. Imputed genotype data are available for 4223 patients with European ancestry (1948 breast, 1728 prostate, 547 lung). Radiation-induced lymphocyte apoptosis (RILA) assay data are available for 1319 patients. DNA (n = 4409) and PAXgene tubes (n = 3039) are stored in the centralised biobank. Example prevalences of 2-year (1-year for lung) grade >= 2 CTCAE toxicities are 13% atrophy (breast), 3% rectal bleeding (prostate) and 27% dyspnoea (lung). Conclusion: The comprehensive centralised database and linked biobank is a valuable resource for the radiotherapy community for validating predictive models and biomarkers. Patient summary: Up to half of cancer patients undergo radiation therapy and irradiation of surrounding healthy tissue is unavoidable. Damage to healthy tissue can affect short-and long-term quality-of-life. Not all patients are equally sensitive to radiation "damage" but it is not possible at the moment to identify those who are. REQUITE was established with the aim of trying to understand more about how we could predict radiation sensitivity. The purpose of this paper is to provide an overview and summary of the data and material available. In the REQUITE study 4400 breast, prostate and lung cancer patients filled out questionnaires and donated blood. A large amount of data was collected in the same way. With all these data and samples a database and biobank were created that showed it is possible to collect this kind of information in a standardised way across countries. In the future, our database and linked biobank will be a resource for research and validation of clinical predictors and models of radiation sensitivity. REQUITE will also enable a better understanding of how many people suffer with radiotherapy toxicity

    The Impact of Antibiotic Use on Mortality in Patients Hospitalized in a COVID-19 Centre from Romania: A Retrospective Study

    Background and Objectives: Considering the significant number of patients worldwide that received empirical antibiotic therapy for COVID-19 infection due to their critical condition and the lack of therapeutical guidelines, we wanted to find out the consequences of antibiotic use in our study population. Materials and Methods: We conducted a retrospective cohort study including symptomatic patients older than 18 years, hospitalized for SARS-CoV-2 between March and December 2020 in the Internal Medicine and Pneumology Departments of Colentina Clinical Hospital. The elected outcome was death, while independent variables were antibiotic therapy and literature-cited parameters associated with mortality in this disease. Results: Out of 198 included patients, 96 (48.48%) patients received antibiotic therapy during hospitalization. Female gender (OR = 2.61, p = 0.04), history of neoplasm (OR = 7.147, p = 0.01), heart failure (OR = 8.62, p = 0.002), and diabetes mellitus (OR = 3.05, p = 0.02) were significantly associated with death in multivariate analysis. Antibiotic treatment showed a higher probability of death both in bivariate (OR = 5.333, p p = 0.01). Conclusions: After adjusting for confounders, in-hospital antibiotic administration did not improve survival in COVID-19 patients

    The objective of the study is to identify the urban development pressure exerted upon the limits imposed by the depth of the phreatic level. The territorial systems from the vicinity of Bucharest registered a momentous dynamics and it enabled the expanding of urbanized areas beyond the limits imposed by the natural conditions. The present research involved the creation of a database concerning the depth of the phreatic level, as well as the identification of the expansion stages undertaken by the urbanized areas. There were applied questionnaires concerning the degree of knowledge regarding the development restrictions for the beneficiaries of built spaces. The spatial layout of the indicators which were used in the research permitted the discovery of some areas where the decision to build something had been taken without analyzing the low depth of piezometric level

    In Silico and In Vitro Experimental Studies of New Dibenz[b,e]oxepin-11(6H)one O-(arylcarbamoyl)-oximes Designed as Potential Antimicrobial Agents

    In a drug-repurposing-driven approach for speeding up the development of novel antimicrobial agents, this paper presents for the first time in the scientific literature the synthesis, physico-chemical characterization, in silico analysis, antimicrobial activity against bacterial and fungal strains in planktonic and biofilm growth state, as well as the in vitro cytotoxicity of some new 6,11-dihydrodibenz[b,e]oxepin-11(6H)one O-(arylcarbamoyl)oximes. The structures of intermediary and final substances (compounds 7a–j) were confirmed by 1H-NMR, 13C-NMR and IR spectra, as well as by elemental analysis. The in silico bioinformatic and cheminformatic studies evidenced an optimal pharmacokinetic profile for the synthesized compounds 7a–j, characterized by an average lipophilic character predicting good cell membrane permeability and intestinal absorption; low maximum tolerated dose for humans; potassium channels encoded by the hERG I and II genes as potential targets and no carcinogenic effects. The obtained compounds exhibited a higher antimicrobial activity against the planktonic Gram-positive Staphylococcus aureus and Bacillus subtilis strains and the Candida albicans fungal strain. The obtained compounds also inhibited the ability of S. aureus, B. subtilis, Escherichia coli and C. albicans strains to colonize the inert substratum, accounting for their possible use as antibiofilm agents. All the active compounds exhibited low or acceptable cytotoxicity levels on the HCT8 cells, ensuring the potential use of these compounds for the development of new antimicrobial drugs with minimal side effects on the human cells and tissues

    Development of a method for generating SNP interaction-aware polygenic risk scores for radiotherapy toxicity

    Aim: To identify the effect of single nucleotide polymorphism (SNP) interactions on the risk of toxicity following radiotherapy (RT) for prostate cancer (PCa) and propose a new method for polygenic risk score incorporating SNP-SNP interactions (PRSi). Materials and methods: Analysis included the REQUITE PCa cohort that received external beam RT and was followed for 2 years. Late toxicity endpoints were: rectal bleeding, urinary frequency, haematuria, nocturia, decreased urinary stream. Among 43 literature-identified SNPs, the 30% most strongly associated with each toxicity were tested. SNP-SNP combinations (named SNP-allele sets) seen in >= 10% of the cohort were condensed into risk (RS) and protection (PS) scores, respectively indicating increased or decreased toxicity risk. Performance of RS and PS was evaluated by logistic regression. RS and PS were then combined into a single PRSi evaluated by area under the receiver operating characteristic curve (AUC). Results: Among 1,387 analysed patients, toxicity rates were 11.7% (rectal bleeding), 4.0% (urinary frequency), 5.5% (haematuria), 7.8% (nocturia) and 17.1% (decreased urinary stream). RS and PS combined 8 to 15 different SNP-allele sets, depending on the toxicity endpoint. Distributions of PRSi differed significantly in patients with/without toxicity with AUCs ranging from 0.61 to 0.78. PRSi was better than the classical summed PRS, particularly for the urinary frequency, haematuria and decreased urinary stream endpoints. Conclusions: Our method incorporates SNP-SNP interactions when calculating PRS for radiotherapy toxicity. Our approach is better than classical summation in discriminating patients with toxicity and should enable incorporating genetic information to improve normal tissue complication probability models.
