What derives less readable 10-K reports: Investor vs Firm-complexity
School of Business Administration (Management Engineering)In this study, I suggest a new channel to explain the negative association between the readability of 10-K reports and firm???s stock price volatility. I find that a high level of firm-complexity influences a decrease in readability of 10-K reports as a firm???s future uncertainty increases. Firms with higher complexity, such as growth firms, release more hard-to-read 10-K reports. The new suggested channel, firm-complexity channel, better explains poor readability of 10-K reports.ope
Gaussian Centered L-moments
As various types of media currently generate so-called big data, data visualization faces the challenge of selecting a few representative variables that best summarize important structure inherent in data. The conventional moments-based summary statistics can be useful for the purpose of variable screening. In particular, they can find important distributional features such as bimodality and skewness. However, their sensitivity to outliers can lead to selection based on a few extreme outliers rather than distributional shape. To address this type of non-robustness, we consider the L-moments. But, describing a marginal distribution with the L-moments has an intuitive limitation in practice because these moments take zero values at the uniform distribution; the interest usually lies in the shape of the marginal distribution compared to the Gaussian, but the sign and magnitude of the L-moments are not as useful as expected for this purpose. As a remedy, we propose the Gaussian Centered L-moments with zeros at the Gaussian distribution while sharing robustness of the L-moments. The Gaussian Centered L-moments can be especially useful for gene expression data in which variable screening corresponds to finding biologically meaningful genes. The mixtures of Gaussian distributions seems to be underlying mechanism generating gene expression profiles, and this suggests moments that are sensitive to departure from Gaussianity. This dissertation deeply investigates theoretical properties of the Gaussian Centered L-moments in various ways. First, by the means of Oja's criteria, the first four terms of the Gaussian Centered L-moments are shown to describe the shape of a distribution in a physically meaningful fashion. Second, comparison between robustness of the conventional, L- and Gaussian Centered L-moments is made based on asymptotic behavior of their influence functions on Tukey's h distributions. Third, the efficiencies of these moments in capturing departure from Gaussianity are compared by developing Jarque-Bera type goodness-of-fit test statistics for Gaussianity. While developing such test statistics, a method for obtaining optimal balance between skewness and kurtosis estimators is introduced. Finally, comprehensive performances including both the robustness and efficiency of the different moments on high dimensional gene expression data are analyzed by the Gene Set Enrichment Analysis.Doctor of Philosoph
Kaposi's Sarcoma-Associated Herpesvirus Latency Locus Compensates for Interleukin-6 in Initial B Cell Activation
Interleukin 6 (IL-6) is considered a proliferation and survival factor for B cells. To assess the role of IL-6 in Kaposi sarcoma-associated herpesvirus (KSHV) latency, KSHV latency locus-transgenic mice (referred to as latency mice) lacking IL-6 were evaluated. IL-6 −/− latency mice had the same phenotypes as the latency mice, i.e., increased frequency of marginal zone B cells, hyperplasia, and hyperglobulinemia, indicating that the KSHV latency locus, which includes all viral microRNAs (miRNAs), can compensate for lack of IL-6 in premalignant B cell activation
Current Studies on Bakanae Disease in Rice: Host Range, Molecular Identification, and Disease Management
AbstractThe seed borne disease such as bakanae is difficult to control. Crop yield loss caused by bakanae depending on the regions and varieties grown, ranging from 3.0% to 95.4%. Bakanae is an important disease of rice worldwide and the pathogen was identified as Fusarium fujikuroi Nirenberg (teleomorph: Gibberella fujikuroi Sawada). Currently, four Fusaria (F. fujikuroi, F. proliferatum, F. verticillioides and F. andiyazi) belonging to F. fujikuroi species complex are generally known as the pathogens of bakanae. The infection occurs through both seed and soil-borne transmission. When infection occurs during the heading stage, rice seeds become contaminated. Molecular detection of pathogens of bakanae is important because identification based on morphological and biological characters could lead to incorrect species designation and time-consuming. Seed disinfection has been studied for a long time in Korea for the management of the bakanae disease of rice. As seed disinfectants have been studied to control bakanae, resistance studies to chemicals have been also conducted. Presently biological control and resistant varieties are not widely used. The detection of this pathogen is critical for seed certification and for preventing field infections. In South Korea, bakanae is designated as a regulated pathogen. To provide highly qualified rice seeds to farms, Korea Seed & Variety Service (KSVS) has been producing and distributing certified rice seeds for producing healthy rice in fields. Adenovirus-induced Reactive Astrogliosis Exacerbates the Pathology of Parkinson's Disease
Parkinson's disease (PD) is the most prevalent neurodegenerative motor disorder. While PD has been attributed to dopaminergic neuronal death in substantia nigra pars compacta (SNpc), accumulating lines of evidence have suggested that reactive astrogliosis is critically involved in PD pathology. These pathological changes are associated with alpha-synuclein aggregation, which is more prone to be induced by an A53T mutation. Therefore, the overexpression of A53T-mutated alpha-synuclein (A53T-alpha-syn) has been utilized as a popular animal model of PD. However, this animal model only shows marginal-to-moderate extents of reactive astrogliosis and astrocytic alpha-synuclein accumulation, while these phenomena are prominent in human PD brains. Here we show that Adeno-GFAP-GFP virus injection into SNpc causes severe reactive astrogliosis and exacerbates the A53T alpha-syn-mediated PD pathology. In particular, we demonstrate that AAV-CMV-A53T-alpha-syn injection, when combined with Adeno-GFAP-GFP, causes more significant loss of dopaminergic neuronal tyrosine hydroxylase level and gain of astrocytic GFAP and GABA levels. Moreover, the combination of AAV-CMV-A53T-alpha-syn and Adeno-GFAP-GFP causes an extensive astrocytic alpha-syn expression, just as in human PD brains. These results are in marked contrast to previous reports that AAV-CMV-A53T-alpha-syn alone causes alpha-syn expression mostly in neurons but rarely in astrocytes. Furthermore, the combination causes a severe PD-like motor dysfunction as assessed by rotarod and cylinder tests within three weeks from the virus injection, whereas Adeno-GFAP-GFP alone or AAV-CMV-A53T-alpha-syn alone does not. Our findings implicate that inducing reactive astrogliosis exacerbates PD-like pathologies and propose the virus combination as an advanced strategy for developing a new animal model of PD.11Nsciescopu
Evaluation of Seasonal Groundwater Quality Changes Associated with Groundwater Pumping and Level Fluctuations in an Agricultural Area, Korea
This study was conducted to evaluate seasonal groundwater quality due to groundwater pumping and hydrochemical characteristics with groundwater level fluctuations in an agricultural area in Korea. Groundwater levels were observed for about one year using automatic monitoring sensors, and groundwater uses were estimated based on the monitoring data. Groundwater use in the area is closely related to irrigation for rice farming, and rising groundwater levels occur during the pumping, which may be caused by the irrigation water of rice paddies. Hydrochemical analysis results for two separate times (17 July and 1 October 2019) show that the dissolved components in groundwater decreased overall due to dilution, especially at wells in the alluvial aquifer and shallow depth. More than 50% of the samples were classified as CaHCO3 water type, and changes in water type occurred depending on the well location. Water quality changes were small at most wells, but changes at some wells were evident. In addition, the groundwater quality was confirmed to have the effect of saltwater supplied during the 2018 drought by comparison with seawater. According to principal component analysis (PCA), the water quality from July to October was confirmed to have changed due to dilution, and the effect was strong at shallow wells. In the study areas where rice paddy farming is active in summer, irrigation water may be one of the important factors changing the groundwater quality. These results provide a qualitative and quantitative basis for groundwater quality change in agricultural areas, particularly rice paddies areas, along with groundwater level and usage
Epstein-Barr virus enhances genome maintenance of Kaposi sarcoma-associated herpesvirus
Primary effusion lymphoma (PEL) is a B cell lymphoma that is always associated with Kaposi's sarcoma-associated herpesvirus (KSHV) and in many cases also with Epstein-Barr virus (EBV); however, the requirement for EBV coinfection is not clear. Here, we demonstrate that adding exogenous EBV to KSHV+ single-positive PEL leads to increased KSHV genome maintenance and KSHV latency-associated nuclear antigen (LANA) expression. To show that EBV was necessary for naturally coinfected PEL, we nucleofected KSHV+/EBV+ PEL cell lines with an EBV-specific CRISPR/Cas9 plasmid to delete EBV and observed a dramatic decrease in cell viability, KSHV genome copy number, and LANA expression. This phenotype was reversed by expressing Epstein-Barr nuclear antigen 1 (EBNA-1) in trans, even though EBNA-1 and LANA do not colocalize in infected cells. This work reveals that EBV EBNA-1 plays an essential role in the pathogenesis of PEL by increasing KSHV viral load and LANA expression
Kaposi's Sarcoma-Associated Herpesvirus Latency Locus Renders B Cells Hyperresponsive to Secondary Infections
Kaposi's sarcoma-associated herpesvirus (KSHV) induces B cell hyperplasia and neoplasia, such as multicentric Castleman's disease (MCD) and primary effusion lymphoma (PEL). To explore KSHV-induced B cell reprogramming in vivo, we expressed the KSHV latency locus, inclusive of all viral microRNAs (miRNAs), in B cells of transgenic mice in the absence of the inhibitory Fc gamma RIIB receptor. The BALB/c strain was chosen as this is the preferred model to study B cell differentiation. The mice developed hyperglobulinemia, plasmacytosis, and B lymphoid hyperplasia. This phenotype was ameliorated by everolimus, which is a rapamycin derivative used for the treatment of mantle cell lymphoma. KSHV latency mice exhibited hyperresponsiveness to the T-dependent (TD) antigen mimic anti-CD40 and increased incidence of pristane-induced inflammation. Lastly, the adaptive immunity against a secondary infection with Zika virus (ZIKV) was markedly enhanced. These phenotypes are consistent with KSHV lowering the activation threshold of latently infected B cells, which may be beneficial in areas of endemicity, where KSHV is acquired in childhood and infections are common.IMPORTANCE Kaposi's sarcoma-associated herpesvirus (KSHV) establishes latency in B cells and is stringently linked to primary effusion lymphoma (PEL) and the premalignant B cell hyperplasia multicentric Castleman's disease (MCD). To investigate potential genetic background effects, we expressed the KSHV miRNAs in BALB/c transgenic mice. BALB/c mice are the preferred strain for B cell hybridoma development because of their propensity to develop predictable B cell responses to antigen. The BALB/c latency mice exhibited a higher incidence of B cell hyperplasia as well as sustained hyperglobulinemia. The development of neutralizing antibodies against ZIKV was augmented in BALB/c latency mice. Hyperglobulinemia was dampened by everolimus, a derivative of rapamycin, suggesting a role for mTOR inhibitors in managing immune activation, which is hallmark of KSHV infection as well as HIV infection
