    Methods of isolation and identification of pathogenic and potential pathogenic bacteria from skins and tannery effluents

    Currently there is no standard protocol available within the leather industry to isolate and identify pathogenic bacteria from hides, skins or tannery effluent. This study was therefore carried out to identify simple but effective methods for isolation and identification of bacterial pathogens from the effluent and skins during leather processing. Identification methods based on both phenotypic and genotypic characteristics were investigated. Bacillus cereus and Pseudomonas aeruginosa were used as indicator bacteria to evaluate the isolation and identification methods. Decontaminated calfskins were inoculated with a pure culture of the above mentioned bacterial species followed by a pre-tanning and chromium tanning processes. Effluent samples were collected and skins were swabbed at the end of each processing stage. Bacterial identification was carried out based on the phenotypic characteristics; such as colony appearance on selective solid media, cell morphology following a standard Gram-staining and spore staining techniques, and biochemical reactions, e.g., the ability of a bacterial species to ferment particular sugars and ability to produce certain enzymes. Additionally, an identification system based on bacterial phenotypic characteristics, known as Biolog® system was applied. A pulsed-filed gel electrophoresis (PFGE) method for bacterial DNA fingerprinting was also evaluated and used for the identification of the inoculated bacteria.     Aneurysmal degeneration of blalock-taussig shunts: Identification and surgical treatment options

    Many Blalock-Taussig shunts (subclavian to pulmonary artery anastomoses) have been created and a significant number are still being done. Two cases of aneurysmal degeneration of a Blalock-Taussig shunt and their management are described. Development of this rare complication may be related to large shunt flow and long duration. Large, symptomatic or enlarging aneurysms should be repaired and smaller ones studied by serial computed axial tomography. A simple and safe approach to correct this lesion is division and oversewing of the proximal subclavian artery through an anterior approach, assuming adequate pulmonary blood flow is already present or can be established concomitantly

    Tidally-Triggered Star Formation in Close Pairs of Galaxies

    We analyze new optical spectra of a sample of 502 galaxies in close pairs and n-tuples, separated by <= 50/h kpc. We extracted the sample objectively from the CfA2 redshift survey, without regard to the surroundings of the tight systems. We probe the relationship between star formation and the dynamics of the systems of galaxies. The equivalent widths of H\alpha (EW(H\alpha) and other emission lines anti-correlate strongly with pair spatial separation (\Delta D) and velocity separation. We use the measured EW(H\alpha) and the starburst models of Leitherer et al. to estimate the time since the most recent burst of star for- mation began for each galaxy. In the absence of a large contribution from an old stellar population to the continuum around H\alpha, the observed \Delta D -- EW(H\alpha) correlation signifies that starbursts with larger separations on the sky are, on average, older. By matching the dynamical timescale to the burst timescale, we show that the data support a simple picture in which a close pass initiates a starburst; EW(H\alpha) decreases with time as the pair separation increases, accounting for the anti-correlation. This picture leads to a method for measuring the duration and the initial mass function of interaction-induced starbursts: our data are compatible with the starburst and orbit models in many respects, as long as the starburst lasts longer than \sim10^8 years and the delay between the close pass and the initiation of the starburst is less than a few \times 10^7 years. If there is no large contribution from an old stellar population to the continuum around H\alpha the Miller-Scalo and cutoff (M <= 30 M_\sun) Salpeter initial mass functions fit the data much better than a standard Salpeter IMF.     Immunotherapy for Infarcts: In Vivo Postinfarction Macrophage Modulation Using Intramyocardial Microparticle Delivery of Map4k4 Small Interfering RNA

    The myeloid cells infiltrating the heart early after acute myocardial infarction elaborate a secretome that largely orchestrates subsequent ventricular wall repair. Regulating this innate immune response could be a means to improve infarct healing. To pilot this concept, we utilized (beta1,3-d-) glucan-encapsulated small interfering RNA (siRNA)-containing particles (GeRPs), targeting mononuclear phagocytes, delivered to mice as a one-time intramyocardial injection immediately after acute infarction. Findings demonstrated that cardiac macrophages phagocytosed GeRPs in vivo and had little systemic dissemination, thus providing a means to deliver local therapeutics. Acute infarcts were then injected in vivo with phosphate-buffered saline (PBS; vehicle) or GeRPs loaded with siRNA to Map4k4, and excised hearts were examined at 3 and 7 days by quantitative polymerase chain reaction, flow cytometry, and histology. Compared with infarcted PBS-treated hearts, hearts with intrainfarct injections of siRNA-loaded GeRPs exhibited 69-89% reductions in transcripts for Map4k4 (mitogen-activated protein kinase kinase kinase kinase 4), interleukin (IL)-1beta, and tumor necrosis factor alpha at 3 days. Expression of other factors relevant to matrix remodeling-monocyte chemoattractant protein-1 (MCP-1), matrix metalloproteinases, hyaluronan synthases, matricellular proteins, and profibrotic factors transforming growth factor beta (TGF-beta), and connective tissue growth factor (CTGF)-were also decreased. Most effects peaked at 3 days, but, in some instances (Map4k4, IL-1beta, TGF-beta, CTGF, versican, and periostin), suppression persisted to 7 days. Thus, direct intramyocardial GeRP injection could serve as a novel and clinically translatable platform for in vivo RNA delivery to intracardiac macrophages for local and selective immunomodulation of the infarct microenvironment

    Photon wave mechanics and position eigenvectors

    One and two photon wave functions are derived by projecting the quantum state vector onto simultaneous eigenvectors of the number operator and a recently constructed photon position operator [Phys. Rev A 59, 954 (1999)] that couples spin and orbital angular momentum. While only the Landau-Peierls wave function defines a positive definite photon density, a similarity transformation to a biorthogonal field-potential pair of positive frequency solutions of Maxwell's equations preserves eigenvalues and expectation values. We show that this real space description of photons is compatible with all of the usual rules of quantum mechanics and provides a framework for understanding the relationships amongst different forms of the photon wave function in the literature. It also gives a quantum picture of the optical angular momentum of beams that applies to both one photon and coherent states. According to the rules of qunatum mechanics, this wave function gives the probability to count a photon at any position in space.Comment: 14 pages, to be published in Phys. Rev.

    There is a good reason why evidence scholars continue to be fascinated and perplexed, and some courts continue at least to be perplexed, by the types of evidence that tend to be lumped together misleadingly under the headings nonassertive conduct or implied assertions. Evidence of this sort highlights a paradox of the prevailing law of hearsay. I believe that this paradox cannot be resolved without fundamentally transforming the structure of that law. Thus, while I agree - within the current framework - with many of the insights so ably stated in this Symposium, I think evidence scholars must devote their efforts to construction of a better structure

    Isolation of quiescent and nonquiescent cells from yeast stationary-phase cultures

    Quiescence is the most common and, arguably, most poorly understood cell cycle state. This is in part because pure populations of quiescent cells are typically difficult to isolate. We report the isolation and characterization of quiescent and nonquiescent cells from stationary-phase (SP) yeast cultures by density-gradient centrifugation. Quiescent cells are dense, unbudded daughter cells formed after glucose exhaustion. They synchronously reenter the mitotic cell cycle, suggesting that they are in a G0 state. Nonquiescent cells are less dense, heterogeneous, and composed of replicatively older, asynchronous cells that rapidly lose the ability to reproduce. Microscopic and flow cytometric analysis revealed that nonquiescent cells accumulate more reactive oxygen species than quiescent cells, and over 21 d, about half exhibit signs of apoptosis and necrosis. The ability to isolate both quiescent and nonquiescent yeast cells from SP cultures provides a novel, tractable experimental system for studies of quiescence, chronological and replicative aging, apoptosis, and the cell cycle
