    Comparison of regulatory regions in the mitochondrial genomes of grasses

The recent sequencing of the mitochondrial genomes of 10 closely related grasses allowed a comparative analysis of regulatory regions. To look for conserved regions and potential “swapped” regulatory regions, we have performed a comparative analysis of the upstream and downstream regions of all of the protein-coding genes in the mitochondrial genomes of eleven grasses: five mitochondrial types of maize (two fertile and three cytoplasmic male sterile), three other taxa within the genus (Zea mays ssp. parviglumis, Z. luxurians, Z. perennis), two close relatives (Tripsacum dactyloides, Sorghum bicolor), and an outgroup, rice. These genomes contain an average of 35 protein-coding genes, composed of 40 transcriptional units. Our analyses examined 1000 base pairs (bp) upstream of the first exon of each transcriptional unit and 1000 bp downstream of its last exon. The reference genome was NB, the most common fertile maize mitochondrial genotype. Compared with the genes from NB, more than half of the mitochondrial genes in the other genomes contain sequences that flank different genes in NB; we refer to these as “swapped” regions. More than 25% of the translocated sequences are longer than 100 bp, and 21 are greater than 500 bp. The longer sequences are more likely to have a regulatory function. In addition, some of these regions were found multiple times: 12 of the translocated gene-flanking regions were found flanking five or more other genes; four had sequences that were flanking ten or more. Furthermore, in Z. luxurians, Z. perennis and T. dactyloides, the co-transcribed 18S and 5S ribosomal RNA genes have been translocated immediately upstream of the start of cox1, with the 5S rRNA 3' end only 80 bp from the start of cox1 exon 1.     Spitzer, Near-Infrared, and Submillimeter Imaging of the Relatively Sparse Young Cluster, Lynds 988e

    We present {\it Spitzer} images of the relatively sparse, low luminosity young cluster L988e, as well as complementary near-infrared (NIR) and submillimeter images of the region. The cluster is asymmetric, with the western region of the cluster embedded within the molecular cloud, and the slightly less dense eastern region to the east of, and on the edge of, the molecular cloud. With these data, as well as with extant Hα\alpha data of stars primarily found in the eastern region of the cluster, and a molecular 13^{13}CO gas emission map of the entire region, we investigate the distribution of forming young stars with respect to the cloud material, concentrating particularly on the differences and similarities between the exposed and embedded regions of the cluster. We also compare star formation in this region to that in denser, more luminous and more massive clusters already investigated in our comprehensive multi-wavelength study of young clusters within 1 kpc of the Sun.Comment: 21 pages, 6 tables, 13 figures.     A Field-Based Approach for Determining ATOFMS Instrument Sensitivities to Ammonium and Nitrate

    Aerosol time-of-flight mass spectrometry (ATOFMS) instruments measure the size and chemical composition of individual particles in real-time. ATOFMS chemical composition measurements are difficult to quantify, largely because the instrument sensitivities to different chemical species in mixed ambient aerosols are unknown. In this paper, we develop a field-based approach for determining ATOFMS instrument sensitivities to ammonium and nitrate in size-segregated atmospheric aerosols, using tandem ATOFMS-impactor sampling. ATOFMS measurements are compared with collocated impactor measurements taken at Riverside, CA, in September 1996, August 1997, and October 1997. This is the first comparison of ion signal intensities from a single-particle instrument with quantitative measurements of atmospheric aerosol chemical composition. The comparison reveals that ATOFMS instrument sensitivities to both NH_4^+ and NO_3^- decline with increasing particle aerodynamic diameter over a 0.32−1.8 ÎŒm calibration range. The stability of this particle size dependence is tested over the broad range of fine particle concentrations (PM_(1.8) = 17.6 ± 2.0−127.8 ± 1.8 ÎŒg m^(-3)), ambient temperatures (23−35 °C), and relative humidity conditions (21−69%), encountered during the field experiments.     Hygroscopic Behavior and Liquid-Layer Composition of Aerosol Particles Generated from Natural and Artificial Seawater

    Sea-salt aerosol (SSA) particles affect the Earth\u27s radiative balance and moderate heterogeneous chemistry in the marine boundary layer. Using conventional and environmental transmission electron microscopes (ETEM), we investigated the hygroscopic growth and liquid-layer compositions of particles generated from three types of aqueous salt solutions: sodium chloride, laboratory-synthesized seawater (S-SSA particles), and natural seawater (N-SSA particles). Three levels of morphological change were observed with the ETEM as the laboratory-generated particles were exposed to increasing relative humidity (RH). The first level, onset of observable morphological changes, occurred on average at 70, 48, and 35% RH for the NaCl, S-SSA, and N-SSA particles, respectively. The second level, rounding, occurred at 74, 66, and 57% RH for NaCl, S-SSA, and N-SSA particles, respectively. The third level, complete deliquescence, occurred at 75% RH for all particles. Collected ambient SSA particles were also examined. With the exception of deliquescence, they did not exhibit the same hygroscopic characteristics as the NaCl particles. The ambient particles, however, behaved most similarly to the synthesized and natural SSA particles, although the onset of morphological change was slightly higher for the S-SSA particles. We used energy-dispersive X-ray spectrometry to study the composition of the liquid layer formed on the S-SSA and N-SSA particles. The layer was enriched in Mg, S, and O relative to the solid particle core. An important implication of these results is that MgSO4-enriched solutions on the surface of SSA particles may be the solvents of many heterogeneous reactions

    Group Report: What is the Role of Heuristics in Litigation

    This chapter examines the role of heuristics in the Anglo-American and Continental litigation systems by considering two broad areas: heuristics that appear in legal rules and procedures, as well as heuristics used by various legal actors (e.g., judges, juries, lawyers). It begins with theoretical accounts of heuristics in psychology and law. Next, it explores the role that heuristics play in the litigation process from the selection and construction of cases to the appellate process. Although procedural rules are in place to ensure that legal decision processes are deliberative, the complexities and uncertainties inherent in legal judgments promote the use of simplifying heuristic strategies. Accordingly, numerous possible instances of heuristics are identified both in legal rules and in the judgment processes of legal actors. The prescriptive utility of heuristics is considered with reference to competing legal ideals. If legal decision makers are to come closer to legal ideals, then the law must strive for perfection through complexity. If legal ideals take account of psychological reality, then the law should design an environment that recognizes human constraints and thereby facilitates heuristic decision strategies that are adaptive. Considerably more scientific work is needed to specify the conditions under which various heuristics are used in the legal domain and under which conditions these heuristics are used successfully to achieve legal objectives

    Adhesion Molecule Expression in Polymorphic Light Eruption

    Endothelial leukocyte adhesion molecule-1 (ELAM-1), vascular cell adhesion molecule-1 (VCAM-1), and intercellular adhesion molecule-1 (ICAM-1) are cytokine-regulated cell-surface Ieukocyte adhesion molecules. We have investigated the in vivo kinetics and pattern of expression of these adhesion molecules in relation to tissue accumulation of leukocytes in the photodermatosis, polymorphic light eruption (PMLE), which is characterized by dense perivascular leukocytic infiltration. Immunohistology was performed on biopsies taken at varying time points from PMLE lesions induced in 11 subjects by suberythemal solar simulated irradiation. Vascular endothelial ELAM-1 expression was first observed at 5 h, maximal at 24 to 72 h, and remained elevated at 6 d. VCAM-1, minimally expressed in control skin, was induced above background levels on endothelium and some perivascular cells after 24h and maintained at 6 d. Endothelial cell ICAM-1 expression was increased above control levels at 72h and 6 d. Keratinocyte ICAM-1 expression, most marked overlying areas of dermal leukocytic infiltration, began at 5h and was strong at 72h and 6 d. In addition to lymphocytes, significant number of neutrophils of but not eosinophils were detected in the dermal leukocytic infiltrate that appeared at 5h and persisted at 6 d. The pattern of adhesion molecule expression that we have observed is similar to that seen in normal skin during a delayed hypersensitivity reaction: These observations support an immunologic basis for PMLE

    Acute effects of nitrate-rich beetroot juice on blood pressure, hemostasis and vascular inflammation markers in healthy older adults: A randomized, placebo-controlled crossover study

    Aging is associated with a vasoconstrictive, pro-coagulant, and pro-inflammatory profile of arteries and a decline in the bioavailability of the endothelium-derived molecule nitric oxide. Dietary nitrate elicits vasodilatory, anti-coagulant and anti-inflammatory effects in younger individuals, but little is known about whether these benefits are evident in older adults. We investigated the effects of 140 mL of nitrate-rich (HI-NI; containing 12.9 mmol nitrate) versus nitrate-depleted beetroot juice (LO-NI; containing ≀0.04 mmol nitrate) on blood pressure, blood coagulation, vascular inflammation markers, plasma nitrate and nitrite before, and 3 h and 6 h after ingestion in healthy older adults (five males, seven females, mean age: 64 years, age range: 57–71 years) in a randomized, placebo-controlled, crossover study. Plasma nitrate and nitrite increased 3 and 6 h after HI-NI ingestion (p < 0.05). Systolic, diastolic and mean arterial blood pressure decreased 3 h relative to baseline after HI-NI ingestion only (p < 0.05). The number of blood monocyte-platelet aggregates decreased 3 h after HI-NI intake (p < 0.05), indicating reduced platelet activation. The number of blood CD11b-expressing granulocytes decreased 3 h following HI-NI beetroot juice intake (p < 0.05), suggesting a shift toward an anti-adhesive granulocyte phenotype. Numbers of blood CD14++CD16+ intermediate monocyte subtypes slightly increased 6 h after HI-NI beetroot juice ingestion (p < 0.05), but the clinical implications of this response are currently unclear. These findings provide new evidence for the acute effects of nitrate-rich beetroot juice on circulating immune cells and platelets. Further long-term research is warranted to determine if these effects reduce the risk of developing hypertension and vascular inflammation with aging

    Measurement of Oxygenated Polycyclic Aromatic Hydrocarbons Associated with a Size-Segregated Urban Aerosol

    Size-segregated atmospheric particles were collected in Boston, MA, using a micro-orifice impactor. The samples were analyzed for oxygenated polycyclic aromatic hydrocarbons (OPAH) using gas chromatography/mass spectrometry. Seven PAH ketones (1-acenaphthenone, 9-fluorenone, 11H-benzo[a]fluoren-11-one, 7H-benzo[c]fluoren-7-one, 11H-benzo[b]fluoren-11-one, benzanthrone, and 6H-benzo[cd]pyrene-6-one), four PAH diones (1,4-naphthoquinone, phenanthrenequinone, 5,12-naphthacenequinone, and benzo[a]pyrene-6,12-dione), and one PAH dicarboxylic acid anhydride (naphthalic anhydride) were identified. Seven additional compounds with mass spectra typical of OPAH were tentatively identified. OPAH were generally distributed among aerosol size fractions based on molecular weight. Compounds with molecular weights between 168 and 208 were ap proximately evenly distributed between the fine (aerodynamic diameter, D_p, 2 ÎŒm) particles. OPAH with molecular weights of 248 and greater were associated primarily with the fine aerosol fraction. Most OPAH were distributed with particle size in a broad, unimodal hump similar to the the distributions observed for PAH in the same samples. These results suggest that OPAH are initially associated with fine particles after formation by either combustion or gas phase photooxidation and then partition to larger particles by vaporization and sorption. Two OPAH were distributed in bimodal distributions with peaks at D_p ≈ 2 ÎŒm and D_p ≈ 2 ÎŒm. These bimodal distributions may be indicative of sorption behavior different from PAH and other OPAH
