    Promoter methylation analysis of WNT/β-catenin pathway regulators and its association with expression of DNMT1 enzyme in colorectal cancer

    Background: Aberrant DNA methylation as the most important reason making epigenetic silencing of genes is a main mechanism of gene inactivation in patients with colorectal cancer. In this study, we decided to identify promoter methylation status of ten genes encoding WNT negative regulators, and measure the expression of DNMT1 enzyme in colorectal cancer samples. Results: Aberrant methylation of APC gene was statistically significant associated with age over 50 (p = 0.017), DDK3 with male (p < 0.0001), SFRP4, WIF1, and WNT5a with increasing tumor stage (p = 0.004, p = 0.029, and p = 0.004), SFRP4 and WIF1 with tumor differentiation (p = 0.009 and p = 0.031) and SFRP2 and SFRP5 with histological type (p = 0.001 and p = 0.025). The increasing number of methylated genes correlated with the expression levels of the DNMT1 mRNA. Conclusions: The rate of gene promoter methylation of WNT pathway regulators is high in colorectal cancer cells. Hyper-methylation is associated with increased expression of the DNMT1 enzyme. © 2014 Mansour Samaei et al.; licensee BioMed Central

    Background: CD1d presents glycolipid antigens to invariant natural killer T (iNKT) cells. The role of CD1d in the development of peptic ulcer and gastric cancer has not been revealed, yet. Objective: To clarify the expression of alternatively spliced variants of CD1d in peptic ulcer and gastric cancer. Methods: Patients with dyspepsia were selected and divided into three groups of non-ulcer dyspepsia (NUD), peptic ulcer disease (PUD), and gastric cancer (GC), according to their endoscopic and histopathological examinations. H. pylori infection was diagnosed by rapid urease test and histopathology. The expression levels of V2, V4, and V5 spliced variants of CD1d molecule were determined by quantitative Reverse Transcriptase PCR. Results: Relative gene expression levels of V4 were higher in GC patients (n=37) than those in NUD (n=49) and PUD (n=51) groups (p<0.05 and p<0.01, respectively). Moreover, GC patients showed higher expression levels of V5 compared to NUD and PUD groups (p<0.001 and p<0.001, respectively). Positive correlation coefficients were attained between V4 and V5 expression in patients with PUD (r=0.734, p<0.0001) and GC (r=0.423, p<0.01), but not in patients with NUD. Among NUD patients, the expression levels of V4, but not V5, were higher in H. pylori-positive patients than in H. pylori-negative ones (p<0.01). Conclusion: Collectively, both membrane-bound (V4) and soluble (V5) isoforms of CD1d were over-expressed in gastric tumor tissues, suggesting that they are involved in anti-tumor immune responses. © 2015, Shiraz University of Medical Sciences.     Hormonal sex reversal of rainbow trout (Oncorhynchus mykiss) by ethynylestradiol-17α (EE2)

    Rainbow trout (Oncorhynchus mykiss) females grow larger and mature later than males, making all-female stocks economically advantageous. The objective of this research was to develop methods for the production of monosex populations of trout through the use of sex steroids. The synthetic estrogen ethynylestradiol-17α (EE2) was administered in single period-immersion treatment of 400 µg/l for 1, 2, 4 and 8 h to groups of newly-hatched sac fries of rainbow trout and in a 30 day dietary treatment of 5, 10, 15 and 20 mg/kgf of newly swum up fries. 73.4 And 94.5% females were obtained with 1 and 2-h single-immersion of EE2 respectively (38% female in control). However, higher doses progressively reduced the survival drastically. Sex ratios of dietary treatment of fry were 60, 57.4, 78 and 94% females respectively. Treatments also resulted in a slight increase of both weight and length. This increase was related to the method of hormonal exposure (dietary > immersion), but not dosage-dependent for utilized hormone. This demonstrates that the direct feminization of rainbow trout can be achieved and resulted in sex reversal ratio effectively but not decreased the growth, as observed in hormonal sex reversed females, and it would be a noticeable approach to the direct feminization of trout

    Different mechanics of snap-trapping in the two closely related carnivorous plants Dionaea muscipula and Aldrovanda vesiculosa

    The carnivorous aquatic Waterwheel Plant (Aldrovanda vesiculosa L.) and the closely related terrestrial Venus Flytrap (Dionaea muscipula SOL. EX J. ELLIS) both feature elaborate snap-traps, which shut after reception of an external mechanical stimulus by prey animals. Traditionally, Aldrovanda is considered as a miniature, aquatic Dionaea, an assumption which was already established by Charles Darwin. However, videos of snapping traps from both species suggest completely different closure mechanisms. Indeed, the well-described snapping mechanism in Dionaea comprises abrupt curvature inversion of the two trap lobes, while the closing movement in Aldrovanda involves deformation of the trap midrib but not of the lobes, which do not change curvature. In this paper, we present the first detailed mechanical models for these plants, which are based on the theory of thin solid membranes and explain this difference by showing that the fast snapping of Aldrovanda is due to kinematic amplification of the bending deformation of the midrib, while that of Dionaea unambiguously relies on the buckling instability that affects the two lobes.Comment: accepted in Physical Review

    A study on growth parameters, blood factors and proximate composition of rainbow trout (Oncorhynchus mykiss) cultured in underground brackish and freshwater

    In this research, growth rate, blood parameters and proximate composition of rainbow trout (Oncorhynchus mykiss) cultured in brackish and freshwater were evaluated. Two treatments (brackish and freshwater) in 3 replications were investigated. Six 1.5 m³ fiberglass tanks were used for this experiment, containing 180 rainbow trout specimens with average weight of 47.2±0.1g. Feeding rate in treatments was about 3% of body weight based on water temperature and fish biomass. Total length and weight of fish were measured in 15 days intervals. After 135 days of culture period, some fish samples were transferred to the Lab. Blood were directly collected from the heart. Muscle composition was analyzed for proximate composition. The results showed that the body weight of samples in brackish water was increased significantly after 45 days of culture period. Blood factors including WBC, RBC, Hb, Hct, MCV, MCH, MCHC, lymphocyte, thrombocyte and concentration of K+ ions in fish cultured in brackish water showed a significant increase (P<0.05). Muscle composition analysis showed a significant increasing (p<0.05) in fat and dry matter in freshwater treatment, while moisture was significantly higher in brackish water (P<0.01)

    Disease classification from capillary electrophoresis: mass spectrometry

    We investigate the possibility of using pattern recognition techniques to classify various disease types using data produced by a new form of rapid Mass Spectrometry. The data format has several advantages over other high-throughput technologies and as such could become a useful diagnostic tool. We investigate the binary and multi-class performances obtained using standard classifiers as the number of features is varied and conclude that there is potential in this technique and suggest research directions that would improve performance
