    To evaluate safety and efficacy of tedizolid phosphate in the management of several skin infections

    Tedizolid Phosphate is an oxazolidinone-class antibiotic and is used for the treatment of acute bacterial skin and skin structure infections. it is a prodrug activated by plasma or intestinal phosphatases to tedizolid following administration of the drug either orally or intravenously. Once activated, tedizolid exerts its bacteriostatic microbial activity through inhibition of protein synthesis by binding to the 50S ribosomal subunit of the bacteria. The purpose of the study was to evaluate safety and efficacy of Tedizolid phosphate and compare it with that of Lenizolid Phosphate another oxazolidinone class of drugs. The study was conducted at OMNI hospital located at dilsukhnagar, Hyderabad. 126 subjects with skin infections, who satisfied the eligibility criteria, were accrued during the study period. These patients were randomized into 2 groups, and were then evaluated according to the treatment protocol. Investigational product was then administered to evaluate safety and efficacy parameters. Subjects received treatment according to the study arm/group. Subjects were asked to take drug for 7 days daily once orally till the clinical symptoms disappear/ as per PIs discretion. Samples for microbiological evaluation were done at screening, end of the therapy. Among both the formulations the group the received Tedizolid phosphate was considered safer and more efficacious as Clinical success rate was 89.9% and the group that received Lenizolid phosphate had the clinical success rate of 81%. It can be concluded that Tedizolid phosphate could be promising drug in the treatment of various skin infections

Echocardiography showed large mobile vegetations on TricuspidValve (TV). Tricuspid Valve Endocarditis (TVE) is generally responsive to medical treatment; however, about 25% of TVE patients require surgical intervention. Long-term survival of a patient is possible without a prosthetic TV replacement, particularly if the pulmonary artery pressure is normal

    The objective of this study is to design and evaluate a new EOP called swellable elementary osmotic pump (SEOP) of the freely water soluble drug, amitriptyline hydrochloride (1 g /mL) by adding water swellable polymers in the core. The hydrophilic polymers included in the core retard the highly water soluble drug by producing hydrogel within the core, which may restrict and delay the solvent contact with drug molecules and may increase the diffusional length of the solvent to achieve a constant release rate.     Carglumic Acid Treatment of a Patient with Recurrent Valproic Acid-induced Hyperammonemia: A Rare Case Report

    Valproic acid, first manufactured as an anticonvulsant, is commonly used to treat both neurological and psychiatric conditions. A rare and deadly side effect of this medication is hyperammonemia, presenting as lethargy, confusion, seizure, and, ultimately, coma. In rare circumstances, hyperammonemia can be recurrent and devastating, especially in patients with an underlying N-acetyl glutamate synthase (NAGS) deficiency, as the valproic acid can enhance this enzyme deficiency and inhibit the conversion of ammonia into urea in the liver. For these subtypes of patients, the United States Food and Drug Administration (US FDA) has recently approved carglumic acid, a medication that can act as a scavenger by effectively increasing the levels of NAGS, ultimately enhancing the conversion of ammonia to urea. In our case report, we have mentioned a patient with treatment-resistant bipolar disorder, who presented with elevated ammonia levels secondary to valproic acid treatment. Valproic acid was the only drug that was effective in his case, so we initiated therapy to reduce his elevated ammonia levels. After a thorough evaluation, we found the patient had a genetic NAGS deficiency. Carglumic acid was initiated and proved efficacious in our patient

    Formulation and invitro evaluation of oral extended release microspheres of aceclofenac using various natural polymers

    In the present work, bioadhesive microspheres of Aceclofenac using Sodium alginate along with Carbopol 934, Carbopol 971, HPMC K4M as copolymers were formulated to deliver Aceclofenac via oral route. The results of this investigation indicate that ionic cross-linking technique Ionotropic gelation method can be successfully employed to fabricate Aceclofenac microspheres. The technique provides characteristic advantage over conventional microsphere method, which involves an “all-aqueous” system, avoids residual solvents in microspheres. FT-IR spectra of the physical mixture revealed that the drug is compatible with the polymers and copolymers used. Micromeritic studies revealed that the mean particle size of the prepared microspheres was in the size range of 512-903µm and are suitable for bioadhesive microspheres for oral administration. The in-vitro mucoadhesive study demonstrated that microspheres of Aceclofenac using sodium alginate along with Carbopol934 as copolymer adhered to the mucus to a greater extent than the microspheres of Aceclofenac using sodium alginate along with Carbopol 971 and HPMC K4M as copolymers. The invitro drug release decreased with increase in the polymer and copolymer concentration. Analysis of drug release mechanism showed that the drug release from the formulations followed non-Fickian diffusion and the best fit model was found to be Korsmeyer-Peppas. Based on the results of evaluation tests formulation coded T4 was concluded as best formulation

Improved fault detection will reduce energy consumption and increase energy efficiency, which will reduce greenhouse gases and other air emissions that contribute to air pollution. The method employed techniques from signal processing and time series analysis to evaluate the correlation among measuring parameters and to assess the presence or absence of faults in the system.     Pathophysiology of Dyslipidemia and its Management by PCSK9 Inhibitors: A Literature Review

    Dyslipidemia is a disease of abnormal lipid levels in the blood that contributes to the atherosclerotic process. This atherogenic process leads to the formation of plaque and also leads to thromboembolic events and other vascular accidents. It is known that high-density lipoprotein cholesterol serves as a protective effect on the vessel wall and causes the reduction in the progression of atherosclerosis. And multiple interventions are directed in maintaining a higher level of the aforementioned lipoprotein cholesterol. While the low-density lipoprotein stays controversial but lowering its levels through various therapeutic agents is the main goal in the management of dyslipidemia. A newer group of drugs, PCSK9 inhibitors lowers the levels of low-density lipoprotein through modulating PCKS9 gene involved in cholesterol metabolism and affects the levels of the lipoproteins by controlling the receptors. The inhibitors of this gene decrease PCSK9-induced low-density lipoprotein receptor degradation in the lysosomes of hepatocytes increasing its recycling and expression on the cell surface, causing increased clearance of low-density lipoprotein from the circulation. These drugs Alicuromab, Evolocumab and along with other agents can be a novel approach in controlling dyslipidemic state. This review revisits the literature in understanding the pathophysiology of dyslipidemia along with its management by PCKS9 inhibitors, its mechanism of action, its pharmacokinetics, the results of the clinical trials and the limitations in its application
