    Folliculogenesis, Fertility and Biotechnology in Dairy Cattle

    The ovarian follicle population is formed by thousands of follicles, preantral and antral, where oocytes are included. During fetal life, the first follicles produced are preantral, and, as they undergo the development process, they reach the final stage of antral follicles, where a cavity/or antrum is developed. All this growth phase is called folliculogenesis, and this chapter will abord the most important aspects of this process. Moreover, not all follicles reach the preovulatory phase and can be fertilized, so we will discuss how reproductive biotechniques can positively influence the fertility of bovine females. We will also discuss the possibility of antral follicle count to influence reproductive performance and the correlation to biotechniques. Finally, we present alternatives on how to improve fertility and productive efficiency in dairy herds

    Risk factors for bovine herpesvirus 1 infection in cattle herds in the West region of Parana State

    O objetivo deste estudo foi determinar a prevalência de rebanhos positivos (focos) e identificar os fatores de risco que possam estar associados com a infecção pelo herpesvírus bovino 1 (BoHV-1) em rebanhos bovinos com atividade reprodutiva, na região Oeste do Estado do Paraná. O delineamento estatístico, amostras de soro e informações referentes às propriedades foram as empregadas para o estudo da brucelose bovina no Estado do Paraná dentro do contexto do Programa Nacional de Controle e Erradicação da Brucelose e Tuberculose. Foram avaliadas 1930 fêmeas com idade igual ou superior a 24 meses, provenientes de 295 rebanhos não vacinados contra o BoHV-1. Para o diagnóstico sorológico da infecção pelo BoHV-1, foi utilizado um ensaio imunoenzimático (ELISA) indireto. Em cada propriedade foi aplicado um questionário epidemiológico, afim de obter informações epidemiológicas e práticas de manejo empregadas. Dos 295 rebanhos analisados, 190 foram considerados positivos para o BoHV-1, com a prevalência de rebanhos de 64,41% (I.C.95% = 58,65-69,87%). As variáveis consideradas fatores de risco para a infecção pelo BoHV-1 na análise de regressão logística multivariada foram: i) número ('> OU ='23) fêmeas com idade '> OU ='24 meses (OR=2,22; IC: 1,09-4,51); ii) compra de reprodutores (OR=2,68; IC: 1,48-4,82); iii) uso de pastagens comuns (OR=5,93; IC: 1,31-26,82); iv) histórico de abortamento nos últimos 12 meses (OR=2,37; IC: 1,09-5,16); v) presença de animais silvestres (OR=8,86; IC: 1,11-70,73). Estes resultados indicam que a infecção pelo BoHV-1 está amplamente distribuída na região estudada e que fatores relacionados às características das propriedades e ao manejo estão associados à infecçãoThe aim of this study was to determine the prevalence of positive herds and to identify the risk factors for BoHV-1 infection in breeding cattle herds in the Western region of Parana State. The statistic delineation, the serum samples and information regarding the selected farms, were the same employed in the study of bovine brucellosis for Parana State in the context of National Program for Control and Eradication of Brucellosis and Tuberculosis. The sampling was performed in two stages. Blood samples were collected from 1930 females aged '> OU ='24 months from 295 non vaccinated herds. Serum samples were tested for antibodies against BoHV-1 using an indirect ELISA. The epidemiological questionnaire was applied on all the selected farms and aimed to obtain epidemiological data. Hundred ninety of the 295 herds were positive for BoHV-1, presenting prevalence of positive herds of 64.41% [58.65-69.87%]. The number ('> OU ='23) of females aged 24 months (OR=2.22; IC: 1.09-4.51), purchased cattle (OR=2.68; IC: 1.48-4.82), use of common grass (OR=5.93; IC: 1.31-26.82), history of abortion in the last 12 months (OR=2.37; IC: 1.09-5.16) and presence of wildlife animals (OR=8.86; IC: 1.11-70.73) were identified as risk factors for BoHV-1 infection in the multivariate logistic regression.     Diagnóstico molecular de herpesvírus ovino tipo 2 em surto de febre catarral malígna em bovinos do Rio Grande do Norte

    Molecular findings that confirmed the participation of ovine herpesvirus 2 (OVH-2) in the lesions that were consistent with those observed in malignant catarrhal fever of cattle are described. Three mixed-breed cattle from Rio Grande do Norte state demonstrated clinical manifestations that included mucopurulent nasal discharge, corneal opacity and motor incoordination. Routine necropsy examination demonstrated ulcerations and hemorrhage of the oral cavity, corneal opacity, and lymph node enlargement. Significant histopathological findings included widespread necrotizing vasculitis, non-suppurative meningoencephalitis, lymphocytic interstitial nephritis and hepatitis, and thrombosis. PCR assay performed on DNA extracted from kidney and mesenteric lymph node of one animal amplified a product of 423 base pairs corresponding to a target sequence within the ovine herpesvirus 2 (OVH-2) tegument protein gene. Direct sequencing of the PCR products, from extracted DNA of the kidney and mesenteric lymph node of one cow, amplified the partial nucleotide sequences (423 base pairs) of OVH-2 tegument protein gene. Blast analysis confirmed that these sequences have 98-100% identity with similar OVH-2 sequences deposited in GenBank. Phylogenetic analyses, based on the deduced amino acid sequences, demonstrated that the strain of OVH-2 circulating in ruminants from the Brazilian states of Rio Grande do Norte and Minas Gerais are similar to that identified in other geographical locations. These findings confirmed the active participation of OVH-2 in the classical manifestations of sheep associated malignant catarrhal fever.Os achados moleculares confirmaram a participação do herpesvírus ovino tipo 2 (OVH-2) nas lesões observadas em um surto de febre catarral malígna em bovinos. Três bovinos oriundos de propriedade rural de Mossoró, Rio Grande do Norte apresentaram manifestações clínicas, que incluíram secreção nasal mucopurulenta, opacidade da córnea e incoordenação motora. A necropsia revelou ulcerações e hemorragias da cavidade oral, opacidade da córnea e linfonodomegalia. Os achados histopatológicos significativos incluíam vasculite necrosante generalizada, meningoencefalite não supurativa, nefrite intersticial linfocítica, hepatite linfocítica e trombose. A PCR, realizada a partir de DNA extraído do rim e do linfonodo mesentérico de um dos animais, amplificou um produto com 423 pares de base do gene da proteína do tegumento do herpesvírus ovino 2 (OVH-2). O sequenciamento direto dos produtos da PCR e a análise pelo Blast demonstraram que o produto amplificado apresentava 98-100% de identidade com sequências do OVH-2 depositadas no GenBank. As análises filogenéticas, baseadas nas sequências deduzidas de aminoácidos demonstraram que a cepa de OVH-2 circulando em ruminantes nos estados de Rio Grande do Norte e Minas Gerais são semelhantes àquelas identificadas em outras regiões geográficas.     Phylogenetic position of an uncharacterized Brazilian strain of bovine papillomavirus in the genus Xipapillomavirus based on sequencing of the L1 open reading frame

    The use of PCR assays with degenerate primers has suggested the existence of numerous as yet uncharacterized bovine papillomaviruses (BPV). Despite the endemic nature of BPV infections, the identification of BPV types in Brazilian cattle is still only sporadic. However, in a recent analysis of a partial segment of the L1 gene, we observed notable diversity among the BPV types detected. The aim of this study was to determine the phylogenetic position of the previously identified wild strain BPV/BR-UEL2 detected in the state of Paraná in Brazil. Since previous analysis of the partial L1 sequence had shown that this strain was most closely related to BPV type 4, genus-specific primers were designed. Phylogenetic analysis using complete L1 ORF sequences revealed that BPV/BR-UEL2 was related to BPV types classified in the genus Xipapillomavirus and shared the highest L1 nucleotide sequence similarity with BPV type 4 (78%). This finding suggests that BPV/BR-UEL2 should be classified as a potential new type of BPV in the genus Xipapillomavirus

    Severe diarrhea outbreak in beef calves (Bos indicus) caused by G6P[11], an emergent genotype of bovine rotavirus group A

    The episodes of diarrhea caused by neonatal bovine rotavirus group A (BoRVA) constitute one of the major health problems in the calf rearing worldwide. The main G (VP7) and P (VP4) genotypes of BoRVA strains involved in the etiology of diarrhea in calves are G6P[1], G10P[11], G6P[5], and G8P[1]. However, less frequently, other G and P genotypes have been described in BoRVA strains identified in diarrheic fecal samples of calves. This study describes the identification and molecular characterization of an emerging genotype (G6P[11]) in BoRVA strains involved in the etiology of a diarrhea outbreak in beef calves in a cattle herd of high production in extensive management system. The diarrhea outbreak, which showed high morbidity (60%) and lethality (7%) rates, occurred in calves (n= 384) Nelore (Bos indicus) up to 30-day-old from the State of Mato Grosso do Sul, Brazil. BoRVA was identified in 80% (16/20) of the fecal samples analyzed by polyacrylamide gel electrophoresis (PAGE) technique. In all PAGE-positive fecal samples were amplified products with 1,062-bp and 876-bp in the RT-PCR assays for VP7 (G type) and VP4 (VP8*) (P type) of BoRVA, respectively. The nucleotide sequence analysis of VP7 and VP4 genes of four wild-type BoRVA strains showed G6-III P[11]-III genotype/lineage. The G6P[11] genotype has been described in RVA strains of human and animal hosts, however, in calves this genotype was only identified in some cross-sectional studies and not as a single cause of diarrhea outbreaks in calves with high morbidity and lethality rates as described in this study. The monitoring of the G and P genotypes of BoRVA strains involved in diarrhea outbreaks in calves is important for both animal and public health by allowing the identification of the most frequent genotypes, the characterization of novel genotypes and to identify reassortments with genotypes described in animal and human hosts. The results of this study show the importance of the monitoring of the genotypes of BoRVA strains involved in episodes of bovine neonatal diarrhea as for characterization of frequency of occurrence and pathogenic potential of uncommon genotypes as for monitoring of the emergency of different BoRVA genotypes not included in commercial vaccines

    Identification of canine papillomavirus type 1 (CPV1) DNA in dogs with cutaneous papillomatosis

    Canine oral papillomavirus (COPV), also known as Canine Papillomavirus type 1 (CPV1), induces papillomas at the mucous membranes of the oral cavity and at the haired skin of dogs. The classification of Papillomavirus (PV) types is based on the L1 capsid protein and nucleotide sequence; so far, 14 CPV types have been described in several countries, but the molecular characterization of CPV in Brazil is lacking. This study investigated the presence of the PV in seven papillomas from four mixed breed dogs from Londrina/PR, Southern Brazil, by partial sequencing of the L1 gene. Seven exophytic cutaneous lesions were surgically removed and processed for histopathological and molecular characterization. Histopathology confirmed the lesions as viral papillomas due to typical histological features. Polymerase Chain Reaction (PCR) assay using the FAP59 and FAP64 primers targeted the L1 gene followed by sequence analysis of the amplicons identified CPV1 in all evaluated papilloma samples. This study represents the first description of CPV1 DNA associated with canine papillomatosis in Brazil
