    Granule Cell Dispersion in Human Temporal Lobe Epilepsy: Proteomics investigation of neurodevelopmental migratory pathways

    Granule cell dispersion (GCD) is a common pathological feature observed in the hippocampus of patients with Mesial Temporal Lobe Epilepsy (MTLE). Pathomechanisms underlying GCD remain to be elucidated, but one hypothesis proposes aberrant reactivation of neurodevelopmental migratory pathways, possibly triggered by febrile seizures. This study aims to compare the proteomes of basal and dispersed granule cells in the hippocampus of eight MTLE patients with GCD to identify proteins that may mediate GCD in MTLE. Quantitative proteomics identified 1882 proteins, of which 29% were found in basal granule cells only, 17% in dispersed only and 54% in both samples. Bioinformatics analyses revealed upregulated proteins in dispersed samples were involved in developmental cellular migratory processes, including cytoskeletal remodelling, axon guidance and signalling by Ras homologous (Rho) family of GTPases (P<0.01). The expression of two Rho GTPases, RhoA and Rac1, was subsequently explored in immunohistochemical and in situ hybridisation studies involving eighteen MTLE cases with or without GCD, and three normal post mortem cases. In cases with GCD, most dispersed granule cells in the outer-granular and molecular layers have an elongated soma and bipolar processes, with intense RhoA immunolabelling at opposite poles of the cell soma, while most granule cells in the basal granule cell layer were devoid of RhoA. A higher density and percentage of cells expressing RhoA was observed in cases with GCD than without GCD (P<0.004). In GCD cases, the density and percentage of cells expressing RhoA was significantly higher in the inner molecular layer than granule cell layer (P<0.026), supporting proteomic findings. In situ hybridisation studies using probes against RHOA and RAC1 mRNAs revealed fine peri- and nuclear puncta in granule cells of all cases. The density of cells expressing RHOA mRNAs were significantly higher in the inner molecular layer of cases with GCD than without GCD(P=0.05). In summary, our study has found limited evidence for ongoing adult neurogenesis in the hippocampus of patients with MTLE, but evidence of differential dysmaturation between dispersed and basal granule cells has been demonstrated, and elevated expression of Rho GTPases in dispersed granule cells may contribute to the pathomechanisms underpinning GCD in MTLE

    Many-Spin Effects and Tunneling Properties of Magnetic Molecules

    Spin tunneling in molecular magnets has attracted much attention, however theoretical considerations of this phenomenon up to now have not taken into account the many-spin nature of molecular magnets. We present, to our knowledge, the first successful attempt of a realistic calculation of tunneling splittings for Mn12_{12} molecules, thus achieving a quantitatively accurate many-spin description of a real molecular magnet in the energy interval ranging from about 100 K down to 10−12^{-12} K. Comparison with the results of the standard single-spin model shows that many-spin effects affect the tunneling splittings considerably. The values of ground state splitting given by single-spin and many-spin models differ from each other by a factor of five.Comment: 3REVTeX pages, 2 figure

    Experimental investigation on CFRP-steel bond properties using ionic liquid

    Solving the problem of pre mature debonding of CFRP retrofitted structure is a main concern for most of structural engineers nowadays. Reducing the brittleness of the bonding agent at the CFRP/concrete interface is a major factor to avoid this behaviour. In this research, the effect of modifying the bonding agent using different percentages of ionic liquid (IL) is investigated. This paper reports on an experimental investigation on the behaviour of modified epoxy resin with IL. Steel plates were used as hosting surface of the CFRP laminates, the laminates were attached to the steel surface using the IL modified epoxy. The shear mechanism at the interface of CFRP laminates to steel plates is discussed considering the relationship between the shear and the slip at the interface. The shear stress- displacement are traced for all specimens, the results are compared with control test prepared using unmodified epoxy. A 20% IL modified epoxy shows improved Behaviour. The improvement is with respect to ductility enhancement of the overall behaviour

    Atrial high-rate episodes: prevalence, stroke risk, implications for management, and clinical gaps in evidence

    Self-terminating atrial arrhythmias are commonly detected on continuous rhythm monitoring, e.g. by pacemakers or defibrillators. It is unclear whether the presence of these arrhythmias has therapeutic consequences. We sought to summarize evidence on the prevalence of atrial high-rate episodes (AHREs) and their impact on risk of stroke. We performed a comprehensive, tabulated review of published literature on the prevalence of AHRE. In patients with AHRE, but without atrial fibrillation (AF), we reviewed the stroke risk and the potential risk/benefit of oral anticoagulation. Atrial high-rate episodes are found in 10-30% of AF-free patients. Presence of AHRE slightly increases stroke risk (0.8% to 1%/year) compared with patients without AHRE. Atrial high-rate episode of longer duration (e.g. those >24 h) could be associated with a higher stroke risk. Oral anticoagulation has the potential to reduce stroke risk in patients with AHRE but is associated with a rate of major bleeding of 2%/year. Oral anticoagulation is not effective in patients with heart failure or survivors of a stroke without AF. It remains unclear whether anticoagulation is effective and safe in patients with AHRE. Atrial high-rate episodes are common and confer a slight increase in stroke risk. There is true equipoise on the best way to reduce stroke risk in patients with AHRE. Two ongoing trials (NOAH-AFNET 6 and ARTESiA) will provide much-needed information on the effectiveness and safety of oral anticoagulation using non-vitamin K antagonist oral anticoagulants in patients with AHRE.info:eu-repo/semantics/publishedVersio

    Openness to Experience, Divergent Thinking, and Gender Differences: Domain and Facet Traits

    Recent creativity studies have provided strong evidence supporting the role of the personality trait of Openness to Experience (Openness) in predicting creative behaviors. These studies have been largely conducted within the framework of the broad Big Five personality traits. In contrast, the validity of narrow traits of which compositing the broad traits has not been adequately addressed. We expected that considering narrow traits might help us deeper understand the role of Openness in creativity. We examined the extent to which narrow personality traits contribute to the prediction of divergent thinking beyond the Big Five factors. To this end, 144 Arab undergraduate students provided data on the domain and facets of Openness. Creativity was measured by Alternative Uses tests. This study has yielded two important results. First, Openness was, at the domain as well as at the facet level, a significant and positive predictor of creativity. Second, the Openness to Feelings is a main predictor for creative female students whereas Openness to Ideas is a significant predictor for creative men students. The limitations and implications of these findings are discussed in the light of previous research

    Markov modeling of moving target defense games

    We introduce a Markov-model-based framework for Moving Target Defense (MTD) analysis. The framework allows modeling of broad range of MTD strategies, provides general theorems about how the probability of a successful adversary defeating an MTD strategy is related to the amount of time/cost spent by the adversary, and shows how a multi-level composition of MTD strategies can be analyzed by a straightforward combination of the analysis for each one of these strategies. Within the proposed framework we define the concept of security capacity which measures the strength or effectiveness of an MTD strategy: the security capacity depends on MTD specific parameters and more general system parameters. We apply our framework to two concrete MTD strategies

    The channeling of the recoiling nucleus in crystalline detectors after a WIMP collision would produce a larger scintillation or ionization signal in direct detection experiments than otherwise expected.
