    EFSO at different geographical locations verified with observing-system experiments

    ひとつひとつの観測データが気象予測に与える影響を簡易に評価する手法を確認 --北極の観測データは7日先の北米気象予測の改善に貢献することも明らかに--. 京都大学プレスリリース. 2021-04-30.An ensemble-based forecast sensitivity to observations (EFSO) diagnosis has been implemented in an atmospheric general circulation model–ensemble Kalman filter data assimilation system to estimate the impacts of specific observations from the quasi-operational global observing system on weekly short-range forecasts. It was examined whether EFSO reasonably approximates the impacts of a subset of observations from specific geographical locations for 6-hour forecasts, and how long the 6-hour observation impacts can be retained during the 7-day forecast period. The reference for these forecasts was obtained from 12 data denial experiments in each of which a subset of three radiosonde observations launched from a geographical location was excluded. The 12 locations were selected from three latitudinal bands comprising (i) four Arctic regions, (ii) four midlatitude regions in the Northern Hemisphere, and (iii) four tropical regions during the Northern Hemisphere winter of 2015/16. The estimated winter-averaged EFSO-derived observation impacts well corresponded to the 6-hour observation impacts obtained by the data denials and EFSO could reasonably estimate the observation impacts by the data denials on short-range (6-hour to 2-day) forecasts. Furthermore, during the medium-range (4-day to 7-day) forecasts, it was found that the Arctic observations tend to seed the broadest impacts and their short-range observation impacts could be projected to beneficial impacts in Arctic and midlatitude North American areas. The midlatitude area located just downstream of dynamical propagation from the Arctic toward the midlatitudes. Results obtained by repeated Arctic data-denial experiments were found to be generally common to those from the non-repeated experiments

    High-Throughput Identification and Screening of Novel Methylobacterium Species Using Whole-Cell MALDI-TOF/MS Analysis

    Methylobacterium species are ubiquitous α-proteobacteria that reside in the phyllosphere and are fed by methanol that is emitted from plants. In this study, we applied whole-cell matrix-assisted laser desorption/ionization time-of-flight mass spectrometry analysis (WC-MS) to evaluate the diversity of Methylobacterium species collected from a variety of plants. The WC-MS spectrum was reproducible through two weeks of cultivation on different media. WC-MS spectrum peaks of M. extorquens strain AM1 cells were attributed to ribosomal proteins, but those were not were also found. We developed a simple method for rapid identification based on spectra similarity. Using all available type strains of Methylobacterium species, the method provided a certain threshold similarity value for species-level discrimination, although the genus contains some type strains that could not be easily discriminated solely by 16S rRNA gene sequence similarity. Next, we evaluated the WC-MS data of approximately 200 methylotrophs isolated from various plants with MALDI Biotyper software (Bruker Daltonics). Isolates representing each cluster were further identified by 16S rRNA gene sequencing. In most cases, the identification by WC-MS matched that by sequencing, and isolates with unique spectra represented possible novel species. The strains belonging to M. extorquens, M. adhaesivum, M. marchantiae, M. komagatae, M. brachiatum, M. radiotolerans, and novel lineages close to M. adhaesivum, many of which were isolated from bryophytes, were found to be the most frequent phyllospheric colonizers. The WC-MS technique provides emerging high-throughputness in the identification of known/novel species of bacteria, enabling the selection of novel species in a library and identification without 16S rRNA gene sequencing

    Outcomes and Prognosis of Neurological Decompression and Stabilization for Spinal Metastasis: Is Assessment with the Spinal Instability Neoplastic Score Useful for Predicting Surgical Results?

    Study Design Retrospective study. Purpose To evaluate the efficacy of the Spinal Instability Neoplastic Score (SINS) in predicting surgical outcomes and survival. Patients were categorized into two groups according to the SINS, and their surgical outcomes and survival following decompression and stabilization were assessed. Overview of Literature Palliative surgery in patients with a life expectancy ≥3 months may effectively improve their overall condition in the long term. Currently, the effectiveness of the SINS for predicting surgical results and survival remains controversial. Methods This study included 44 patients who underwent decompression and stabilization for spinal metastases at Yokosuka Kyosai Hospital between 2008 and 2017. The patients were divided into two groups: stable (SINS ≤12) and unstable (SINS ≥13). Changes in the Frankel score and Eastern Cooperative Oncology Group Performance Status (ECOG-PS) were compared between the two groups, and patient survival was evaluated according to the SINS, Tokuhashi score, and Katagiri score. Results The stable group (SINS range, 7–12) included 24 patients while the unstable group (SINS range, 13–16) included 20 patients. The Frankel score significantly improved from 2.8 to 3.6 in the stable group (p <0.001) and from 2.7 to 3.9 in the unstable group (p <0.001). The ECOG-PS significantly improved from 3.2 to 2.1 in the stable group (p <0.001) and from 3.0 to 1.8 in the unstable group (p <0.001). There was a statistically significant difference in median survival between the two groups. Conclusions All patients treated with palliative surgery showed favorable outcomes, as indicated by improved the Frankel score and ECOG-PS following surgery. However, median survival was significantly better in the stable group. The results of this study indicate that the SINS is appropriate for surgical decision making and may be used to predict survival

    Predictability of Explosive Cyclogenesis over the Northwestern Pacific Region Using Ensemble Reanalysis

    The predictability of explosive cyclones over the northwestern Pacific region is investigated using an ensemble reanalysis dataset. Explosive cyclones are categorized into two types according to whether the region of the most rapid development is in the Sea of Okhotsk or Sea of Japan (OJ) or in the northwestern Pacific Ocean (PO). Cyclone-relative composite analyses are performed for analysis increments (the differences between the analysis and the 6-h forecast) and ensemble spreads (the standard deviations of ensemble members of the analysis or first guess) at the time of the maximum deepening rate. The increment composite shows that the OJ explosive cyclone center is forecast too far north compared to the analyzed center, whereas the PO explosive cyclone is forecast shallower than the analyzed center. To understand the cause of these biases, a diagnosis of the increment using the Zwack-Okossi (Z-O) development equation is conducted. The results suggest that the increment characteristics of both the OJ and PO explosive cyclones are associated with the most important cyclone development mechanisms. The OJ explosive cyclone forecast error is related to a deeper upper trough, whereas the PO explosive cyclone error is related to weaker latent heat release in the model. A diagnosis of the spread utilizing the Z-O development equation clarifies the mechanism underlying the uncertainty in the modeled sea level pressure. For OJ explosive cyclones, the spread of adiabatic warming causes substantial sea level pressure spreading southwest of the center of the cyclones. For PO explosive cyclones, the latent heat release causes substantial sea level pressure spreading around the cyclone center

    日本における水球競技は、他の多くのスポーツ競技同様に、クラブ活動を中心とした学校教育システムの中で発展を見せてきた。しかしながら、このシステムには若年代からシニア選手に至る長期的な指導理念に基づいた具体的施策が不足していることが浮き彫りにされた。一方諸外国及び日本国内でも、長期育成型の一貫指導プログラムによって成功を収める競技が多く存在している。日本代表チームが五輪出場を目指す上で長期的な視野に立った指導プログラムの存在は欠かせない。こうした状況を背景に財団法人日本水泳連盟水球委員会では、水球競技選手の国際的競技能力の向上を目指した長期的な育成計画として「水球一貫指導プログラム」を開発し、その運用に着手した。本プログラムにおける今後の課題は、世界最先端の水球競技における技術と戦術に常に照らし合わせつつ内容を柔軟に対応させることと、競技への導入段階である最も初心者レベルにおけるより効果的なプログラムを充実させることにある。The coaching system of Japan Water polo has developed by the school education program. However, this coaching system does not have philosophy of long term athlete development and priority policies for it. On the contrary, national sports federation which has established the long term athlete development program (LTAD) has continued to rise and achieve a successful outcome over the world. LTAD will be one of the keywords of success in which Japan water polo participate the Olympic. Japan Swimming Federation Water Polo Committee has developed "LTAD system for Water polo" to solve this problem. This system has two action assignments. One is that it must modify to follow the frontier of world water polo scene and the other is to make some solution for the beginner of water polo

    Optimization of quantum noise in space gravitational-wave antenna DECIGO with optical-spring quantum locking considering mixture of vacuum fluctuations in homodyne detection

    Quantum locking using optical spring and homodyne detection has been devised to reduce quantum noise that limits the sensitivity of DECIGO, a space-based gravitational wave antenna in the frequency band around 0.1 Hz for detection of primordial gravitational waves. The reduction in the upper limit of energy density ΩGW{\Omega}_{\mathrm{GW}} from 2×10152{\times}10^{-15} to 1×10161{\times}10^{-16}, as inferred from recent observations, necessitates improved sensitivity in DECIGO to meet its primary science goals. To accurately evaluate the effectiveness of this method, this paper considers a detection mechanism that takes into account the influence of vacuum fluctuations on homodyne detection. In addition, an advanced signal processing method is devised to efficiently utilize signals from each photodetector, and design parameters for this configuration are optimized for the quantum noise. Our results show that this method is effective in reducing quantum noise, despite the detrimental impact of vacuum fluctuations on its sensitivity.Comment: 12 pages, 5 figure

    First-step experiment in developing optical-spring quantum locking for DECIGO: sensitivity optimization for simulated quantum noise by completing the square

    DECi-hertz Interferometer Gravitational Wave Observatory (DECIGO) is a future mission for a space-borne laser interferometer. DECIGO has 1,000-km-long arm cavities mainly to detect the primordial gravitational waves (PGW) at lower frequencies around 0.1 Hz. Observations in the electromagnetic spectrum have lowered the bounds on the upper limit of PGW energy density (Ωgw10151016\Omega_{\rm gw} \sim 10^{-15} \to 10^{-16}). As a result, DECIGO's target sensitivity, which is mainly limited by quantum noise, needs further improvement. To maximize the feasibility of detection while constrained by DECIGO's large diffraction loss, a quantum locking technique with an optical spring was theoretically proposed to improve the signal-to-noise ratio of the PGW. In this paper, we experimentally verify one key element of the optical-spring quantum locking: sensitivity optimization by completing the square of multiple detector outputs. This experiment is operated on a simplified tabletop optical setup with classical noise simulating quantum noise. We succeed in getting the best of the sensitivities with two different laser powers by the square completion method.Comment: 10 pages, 14 figure

    LDL-C/HDL-C Ratio Predicts Carotid Intima-Media Thickness Progression Better Than HDL-C or LDL-C Alone

    High-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C) are strong predictors of atherosclerosis. Statin-induced changes in the ratio of LDL-C to HDL-C (LDL-C/HDL-C) predicted atherosclerosis progression better than LDL-C or HDL-C alone. However, the best predictor of subclinical atherosclerosis remains unknown. Our objective was to investigate this issue by measuring changes in carotid intima-media thickness (IMT). A total of 1,920 subjects received health examinations in 1999, and were followed up in 2007. Changes in IMT (follow-up IMT/baseline IMT × 100) were measured by ultrasonography. Our results showed that changes in IMT after eight years were significantly related to HDL-C (inversely, P < 0.05) and to LDL-C/HDL-C ratio (P < 0.05). When the LDL-C/HDL-C ratios were divided into quartiles, analysis of covariance showed that increases in the ratio were related to IMT progression (P < 0.05). This prospective study demonstrated the LDL-C/HDL-C ratio is a better predictor of IMT progression than HDL-C or LDL-C alone

    Identification of novel proteins differentially expressed in pluripotent embryonic stem cells and differentiated cells

    Mammalian pluripotent stem cells possess properties of self-renewal and pluripotency. These abilities are maintained by the strict regulation of pluripotent stem cell-specific transcription factor network and unique properties of chromatin in the stem cells. Although these major signaling pathways robustly control the characteristics of stem cells, other regulatory factors, such as metabolic pathways, are also known to modulate stem cell proliferation and differentiation. In this study, we fractionated protein samples from mouse embryonic stem (ES) cells cultured with or without the leukemia inhibitory factor (LIF). Protein expression was quantified by 2-dimensional differential gel electrophoresis (2D-DIGE). In total, 44 proteins were identified as being differentially expressed in the pluripotent stem cells and the differentiated cells. Surprisingly, half of the identified proteins were the proteins localized in mitochondria, which supply cellular energy and regulate cell cycle, development, and cell death. Some of these identified proteins are involved in the metabolic function and the regulation of pluripotency. Further analysis of the identified proteins could provide new information for the manipulation of pluripotency in ES cells
