    Evaluation of Antibacterial and Antifungal Properties of Alchornea laxiflora

    Alchornea laxiflora leaf extract was tested against a range of microorganisms using standard microbiological methods for antimicrobial activities. The extract inhibited the growth of all the bacterial and 15 fungal isolates tested. The zones of inhibition exhibited against the test bacteria ranged between 12 mm and 24 mm and between 11 mm and 24 mm for the extract and the antibiotic streptomycin, respectively. The zones of inhibition observed against the fungal isolates by the extract ranged between 12 mm and 23 mm. The minimum inhibitory concentrations (MICs) and the minimum bactericidal concentrations (MBCs) exhibited by the extract against test bacteria ranged between 0.78 mg/mL–25 mg/mL and 1.56 mg/mL–25 mg/mL, respectively, while the MICs and minimum fungicidal concentrations (MFCs) values for the test fungi ranged between 8.75 mg/mL–35.00 mg/mL and 8.75 mg/mL–35.00 mg/L, respectively. The preliminary phytochemical screening of the extract revealed the presence of alkaloids, tannins, flavonoids, saponins, and reducing sugars as major phytoconstituents in the extract. A. laxiflora leaf extract is a potent source of antibacterial and antifungal compounds; further studies on the extract are ongoing in our laboratories to elucidate the probable mechanism(s) of action on bacteria and fungi found to be susceptible to the extract

    RADIOACTIVITY CONCENTRATION AND DOSE ASSESSMENT OF SOIL SAMPLES IN CEMENT FACTORY AND ENVIRONS IN OGUN STATE, NIGERIA

    The main natural radioactive materials that contribute externally and internally to man and his environs are radionuclides emanating from gamma series of 238U, 232Th, as well as 40K isotope, alpha and beta sources. Also, human activities such as mining and milling of uranium and phosphate, tobacco smoking, oil exploration, air transportation, coal-fired power station, and so on can trigger The technologically Enhanced Naturally Occurring Radioactive Materials (TENORMs). It is imperative to monitor the terrestrial background radiation of radionuclides emanating from these sources in order maintain a safe environment for humans. This study measured the activity concentration of 238U, 40K and 232Th in soil samples from cement factory and environs using the NaI(TI) detector, in three zones in Ogun State, Nigeria. The activity concentrations obtained for the three zones are in the order 238U < 232Th < 40K respectively. The radiological parameters estimated from the activity measured were all within the recommended permissive limit except for the annual gonadal doses from the cement factory and its environs, which are higher than the global standard by the factors of 1.03 and 1.07 respectively. This study therefore concludes that people living and working in these areas might be exposed to high radiation burden as a result of cumulative effect of the emission from the factory

    Drug Use and High-risk Sexual Behavior among School-Going Adolescents in Nigeria

    BACKGROUND: Sexual risk behavior and drug abuse among adolescents and youths remained perpetual topical issues of focus in most developmental programs related to developing countries, especially in Nigeria, where the school-going adolescents constitute more than half of the youths. The high level of teenage pregnancy and sexual violence such as abuses and other harmful trajectories including STIs and HIV is increasingly reported than the pre-2000s. AIM: This study focuses on underscoring the variation in risky sexual behavior among school-going users and non-users of drugs. It also analyzed the predisposing factors of drug use among school-going adolescents in Nigeria. METHODS: In combination with problem behavior theory, the research draws data (n = 11,799) from the 2012 National HIV and AIDS and Reproductive Health Survey (NARHS Plus II) collated by the Federal Ministry of Health in Nigeria with support from the Department for International Development and United States Agency for International Development, to underscore the self-reported sexual risk behavior among students who are users and non-users of drugs. RESULTS: The result revealed that 32.5% (male) and 33.4% (female) use drugs. More than half of the respondents reported that they have engaged in sexual intercourse, 27.3% (male) and 31.8% (female) have had ≥2 lifetime sexual partners. There is higher odds ratio (OR) of risky sexual behavior among students that have ever used drugs or taken alcohol (OR = 2.2, 95% CI [1.8–2.8]) for male and (OR = 2.1, 95% CI [0.83–2.03]) for female. CONCLUSION: The study concludes that continued exposure of school-going youths to drugs or alcohol may pose serious challenge of risky sexual behavior and also severe threat to initiatives on zero new HIV infections or zero new AIDS death in Nigeria. The authors recommend that campaign to discourage drug or alcohol use should be intensified and introduced to all schools

    Cortical AAV-CNTF gene therapy combined with intraspinal mesenchymal precursor cell transplantation promotes functional and morphological outcomes after spinal cord injury in adult rats

    Ciliary neurotrophic factor (CNTF) promotes survival and enhances long-distance regeneration of injured axons in parts of the adult CNS. Here we tested whether CNTF gene therapy targeting corticospinal neurons (CSN) in motor-related regions of the cerebral cortex promotes plasticity and regrowth of axons projecting into the female adult F344 rat spinal cord after moderate thoracic (T10) contusion injury (SCI). Cortical neurons were transduced with a bicistronic adeno-associated viral vector (AAV1) expressing a secretory form of CNTF coupled to mCHERRY (AAV-CNTFmCherry) or with control AAV only (AAV-GFP) two weeks prior to SCI. In some animals, viable or nonviable F344 rat mesenchymal precursor cells (rMPCs) were injected into the lesion site two weeks after SCI to modulate the inhibitory environment. Treatment with AAV-CNTFmCherry, as well as with AAV-CNTFmCherry combined with rMPCs, yielded functional improvements over AAV-GFP alone, as assessed by open-field and Ladderwalk analyses. Cyst size was significantly reduced in the AAV-CNTFmCherry plus viable rMPC treatment group. Cortical injections of biotinylated dextran amine (BDA) revealed more BDA-stained axons rostral and alongside cysts in the AAV-CNTFmCherry versus AAV-GFP groups. After AAV-CNTFmCherry treatments, many sprouting mCherry-immunopositive axons were seen rostral to the SCI, and axons were also occasionally found caudal to the injury site. These data suggest that CNTF has the potential to enhance corticospinal repair by transducing parent CNS populations

    Convalescent plasma in patients admitted to hospital with COVID-19 (RECOVERY): a randomised controlled, open-label, platform trial

    SummaryBackground Azithromycin has been proposed as a treatment for COVID-19 on the basis of its immunomodulatoryactions. We aimed to evaluate the safety and efficacy of azithromycin in patients admitted to hospital with COVID-19.Methods In this randomised, controlled, open-label, adaptive platform trial (Randomised Evaluation of COVID-19Therapy [RECOVERY]), several possible treatments were compared with usual care in patients admitted to hospitalwith COVID-19 in the UK. The trial is underway at 176 hospitals in the UK. Eligible and consenting patients wererandomly allocated to either usual standard of care alone or usual standard of care plus azithromycin 500 mg once perday by mouth or intravenously for 10 days or until discharge (or allocation to one of the other RECOVERY treatmentgroups). Patients were assigned via web-based simple (unstratified) randomisation with allocation concealment andwere twice as likely to be randomly assigned to usual care than to any of the active treatment groups. Participants andlocal study staff were not masked to the allocated treatment, but all others involved in the trial were masked to theoutcome data during the trial. The primary outcome was 28-day all-cause mortality, assessed in the intention-to-treatpopulation. The trial is registered with ISRCTN, 50189673, and ClinicalTrials.gov, NCT04381936.Findings Between April 7 and Nov 27, 2020, of 16 442 patients enrolled in the RECOVERY trial, 9433 (57%) wereeligible and 7763 were included in the assessment of azithromycin. The mean age of these study participants was65·3 years (SD 15·7) and approximately a third were women (2944 [38%] of 7763). 2582 patients were randomlyallocated to receive azithromycin and 5181 patients were randomly allocated to usual care alone. Overall,561 (22%) patients allocated to azithromycin and 1162 (22%) patients allocated to usual care died within 28 days(rate ratio 0·97, 95% CI 0·87–1·07; p=0·50). No significant difference was seen in duration of hospital stay (median10 days [IQR 5 to >28] vs 11 days [5 to >28]) or the proportion of patients discharged from hospital alive within 28 days(rate ratio 1·04, 95% CI 0·98–1·10; p=0·19). Among those not on invasive mechanical ventilation at baseline, nosignificant difference was seen in the proportion meeting the composite endpoint of invasive mechanical ventilationor death (risk ratio 0·95, 95% CI 0·87–1·03; p=0·24).Interpretation In patients admitted to hospital with COVID-19, azithromycin did not improve survival or otherprespecified clinical outcomes. Azithromycin use in patients admitted to hospital with COVID-19 should be restrictedto patients in whom there is a clear antimicrobial indication

    Effects of different fertilizer sources and rates on proximate and phytochemical composition of three cucumber varieties (Cucumis sativus L.)

    Proximate and phytochemical analysis of three cucumber varieties (Poinsett, Marketer and Marketmore) as influenced by the application of fertilizer types and rates was carried out at the Department of Biochemistry and Molecular Biology of the Obafemi Awolowo University, Ile-Ife. Cucumber was subjected to four fertilizer types: Organic fertilizer, Liquid organic fertilizer, Urea fertilizer and NPK fertilizer applied at the rate of 0, 75 and 120kg N/ha. Proximate compositions showed several disparities with the different treatments. Poinsett, Marketer and Marketmore varieties responded well to Liquid organic fertilizer treatment at all rates to produce significantly (p&lt;0.05) high percentage carbohydrate contents and dry matter contents. Higher rates of Urea and Organic fertilizer favoured the production of crude protein. The qualitative phytochemical analysis showed that Marketer and Marketmore varieties accounted for the highest secondary metabolites. Phenols were readily available in all varieties regardless of the nutrient source. NPK and Urea fertilizer produced more phenols and at higher rates, accounted for more flavonoids in Marketer and Marketmore varieties. 120 Kg N NPK increased the percentage carbohydrate in Poinsett, Markerter and Marketmore by 48%, 5% and 1% respectively. Also, 120 Kg N NPK accounted for 30% and 2 % increase in proximal fruit dry matter for both Poinsett and Marketmore varieties respectively whereas 120 Kg N Liquid Organic fertilizer boosted fruit dry matter in Marketer by only 10%. Results showed that organic fertilizer improved the nutritional values in cucumber fruit compared with inorganic fertilizer but increased the perishability by increasing the moisture content. However, there is the need to further quantify the concentration of the secondary metabolites in cucumber fruits for nutritional values.Keywords: Cucumber, Fertilizer, Phytochemicals, Proximate Compositio

    Preliminary Phytochemical Screening and Antibacterial Properties of Crude Stem Bark Extracts and Fractions of Parkia biglobosa (Jacq.)

    A methanolic crude extract of Parkia biglobosa was prepared and later partitioned in succession with different solvents of increasing polarity ranging from n-hexane, chloroform and ethyl acetate to butanol. Phytochemical screening of the extract revealed the presence of alkaloids, tannins, saponins, flavonoids, steroids, glycoside and sugars. The inhibition zones exhibited by the extract against the tested bacteria ranged between 14 ± 0.00 mm (against Escherichia coli) and 28 ± 0.71 mm (against Pseudomonas aeruginosa). The MIC of the methanolic extract of P. biglobosa against isolates ranged between 0.63 mg/mL and 5 mg/mL, while the MIC values exhibited by the n-hexane and aqueous fractions ranged between 0.63 mg/mL and 10 mg/mL. Overall the extract and fractions of P. biglobosa used in this work were found to possess antimicrobial properties which compared favourably with those of streptomycin. These observations make this plant a potential source of bioactive compounds that can be used in management of bacterial infections. The use of this plant as herbal medicaments in African countries and the reports on the toxicity of the plant further show that the plant is non-toxic to humans
