    The east–west component of magnetic field variation (∆D-component) at Davao station (Philippines, geomagnetic latitude: – 2.22˚N) are used to investigate the characteristics of the long-term Inter-Hemispheric Field-Aligned Currents (IHFACs) based on the time-series analysis from August 1998 to July 2020. Recent in situ satellite and ground-based observations have reported that dusk-side current polarity of IHFAC is often opposite to that of the noon IHFAC, being inconsistent with Fukushima\u27s IHFACs model. We investigated the consistency of the dusk-side IHFAC polarity derived from the observations with the polarity expected from Fukushima’s IHFACs model and examined the solar cycle dependence of IHFACs. It was confirmed that the dusk-side IHFACs during June and December solstices flow in the same direction of the noontime IHFACs, which was consistent with the IHFAC polarities suggested by the Fukushima model. The dusk-side IHFACs around March and September–November months disagreed with the Fukushima model. The ∆D variations clearly showed seasonal asymmetry in the dawn and noon sectors, whereas the ∆D variations in the dusk sector demonstrated seasonal symmetry. Solar cycle dependence of IHFACs was exhibited in the dusk sector. For the dawn and noon sectors, the yearly peak-to-peak ∆D amplitude in the later solar cycle SC24 decreased by about 35% in comparison with the earlier solar cycle SC23. In contrast, the dusk-side yearly peak-to-peak ∆D amplitude increased by about 200%. The dusk-side IHFAC yearly amplitude tended to be in inverse proportion to solar activity

    Seasonal dependence of semidiurnal equatorial magnetic variation during quiet and disturbed periods

    The analysis of 20-year long-term semidiurnal lunar tidal variations gave the evidence that the semidiurnal variations are completely different between the magnetic quiet and disturbed periods. This is the first time that the seasonal dependence of disturbance-time semidiurnal variation has been provided from the analysis of the EE-index. We found the Kp dependence of semidiurnal variation: For full and new moon phase, counter troughs are amplified during disturbance time, possibly related to disturbance dynamo. For all moon phase, there are positive enhancements in dawn and strong depressions after sunset, resulting from the penetration of polar electric filed. For Seasonal dependence, semidiurnal variations are divided to three seasonal groups, and characterized as deep trough, enhanced crest and weak structure for D-solstice, Equinoxes and J-solstice, respectively. There is no significant longitudinal difference between Ancon and Davao, except for the amplitude of semidiurnal variations.     Fetal and Neonatal Goiter in Cynomolgus Monkeys Following Administration of the Antithyroid Drug Thiamazole at High Doses to Dams During Pregnancy

    To evaluate morphologic alterations in the thyroid gland in the second generation in cynomolgus monkeys, pregnant dams were exposed to high doses of thiamazole. In Experiment A, dams received thiamazole intragastrically via a nasogastric catheter from gestation day (GD) 50 to GD 150 or on the day before delivery. Initially, the dose level was 20 mg/kg/day (10 mg/kg twice daily); however, the dose level was subsequently decreased to 5 mg/kg/day (2.5 mg/kg twice daily), since deteriorated general conditions were observed in two dams. Six out of seven neonates died on the day of birth. The cause of neonatal death was tracheal compression and suffocation from goiter. The transplacental exposure to thiamazole affected the fetal thyroid glands and induced goiter in all neonates. The surviving neonate was necropsied 767 days after discontinuation of thiamazole exposure and showed reversibility of the induced changes. In Experiment B, dams were intragastrically administered thiamazole at 5 mg/kg/day (2.5 mg/kg twice daily) for treatment periods from GDs 51 to 70, 71 to 90, 91 to 110, 111 to 130 and 131 to 150. All fetuses showed enlarged thyroid glands but were viable. Histopathologically, hypertrophy and/or hyperplastic appearance of the follicular epithelium of the thyroid gland was observed at the end of each treatment period. The most active appearance of the follicular epithelium, consisting of crowded pedunculated structure, was demonstrated at end of the treatment period from GD 131 to 150. This is the first report on the morphology of fetal and neonatal goiter in the cynomolgus monkey

    Tertiary Lymphoid Tissues Are Microenvironments with Intensive Interactions between Immune Cells and Proinflammatory Parenchymal Cells in Aged Kidneys

However, the mechanisms underlying TLT expansion and their effect on renal regeneration remain unclear. The authors report that single-nucleus RNA sequencing and validation experiments demonstrate that TLTs potentially amplify inflammation in aged injured kidneys. Lymphocytes within TLTs promote proinflammatory phenotypes of the surrounding proximal tubules and fibroblasts within the TLTs via proinflammatory cytokine production. These proinflammatory parenchymal cells then interact with immune cells by chemokine or cytokine production. Such cell-cell interactions potentially increase inflammation, expand TLTs, and exacerbate kidney injury. These findings help illuminate renal TLT pathology and suggest potential therapeutic targets. [Background] Ectopic lymphoid structures called tertiary lymphoid tissues (TLTs) develop in several kidney diseases and are associated with poor renal prognosis. However, the mechanisms that expand TLTs and underlie exacerbation of kidney injury remain unclear. [Methods] We performed single-nucleus RNA sequencing (snRNA-seq) on aged mouse kidneys with TLTs after ischemia-reperfusion injury. The results were validated using immunostaining, in situ hybridization of murine and human kidneys, and in vitro experiments. [Results] Using snRNA-seq, we identified proinflammatory and profibrotic Vcam1⁺ injured proximal tubules (PTs) with NFκB and IFN-inducible transcription factor activation. VCAM1⁺ PTs were preferentially localized around TLTs and drove inflammation and fibrosis via the production of multiple chemokines or cytokines. Lymphocytes within TLTs expressed Tnf and Ifng at high levels, which synergistically upregulated VCAM1 and chemokine expression in cultured PT cells. In addition, snRNA-seq also identified proinflammatory and profibrotic fibroblasts, which resided within and outside TLTs, respectively. Proinflammatory fibroblasts exhibited STAT1 activation and various chemokine or cytokine production, including CXCL9/CXCL10 and B cell–activating factor, contributing to lymphocyte recruitment and survival. IFNγ upregulated the expression of these molecules in cultured fibroblasts in a STAT1-dependent manner, indicating potential bidirectional interactions between IFNγ-producing CXCR3⁺ T cells and proinflammatory fibroblasts within TLTs. The cellular and molecular components described in this study were confirmed in human kidneys with TLTs. [Conclusions] These findings suggest that TLTs potentially amplify inflammation by providing a microenvironment that allows intense interactions between renal parenchymal and immune cells. These interactions may serve as novel therapeutic targets in kidney diseases involving TLT formation

    SUDACHITIN IS A SAFE ANTICANCER ADJUVANT TARGETING THE TME

    Sudachitin is a polymethoxylated flavone found in the peel of Citrus sudachi, a unique specialty citrus fruit in Tokushima Prefecture, Japan. Previous reports have demonstrated that sudachitin has anti‑inflammatory and metabolic regulatory activities. However, to the best of our knowledge, no studies have explored whether sudachitin can act as an antitumor therapeutic agent by regulating metabolic functions in the tumor microenvironment. In the present study, cell proliferation and cytotoxicity assays were used to determine whether sudachitin inhibited the in vitro growth of liver cancer and pancreatic carcinoma, cholangiocarcinoma and colorectal cancer cells and to compare its toxicity against normal fibroblasts and induced cancer‑associated fibroblasts (CAFs). Using lactate assays and reverse transcription‑quantitative PCR, the effects of sudachitin on glycolysis in CAFs were investigated. The effects of CAFs on malignant tumor cells were evaluated in vitro using cell proliferation, wound healing and invasion assays. As result, sudachitin inhibited various types of tumor cells with different half‑maximal inhibitory concentrations. Treatment with 50 µM sudachitin for 48 h suppressed tumor and CAFs proliferation but was not cytotoxic against normal fibroblasts. This dose also inhibited glycolysis in CAFs and, thus, diminished their pro‑tumorigenic activities. Overall, the present study revealed that sudachitin has promise as a safe and widely available natural antitumor adjuvant
