    TFRC–RNA interactions show the regulation of gene expression and alternative splicing associated with IgAN in human renal tubule mesangial cells

    Introduction: IgA nephropathy (IgAN) is the most common primary glomerular disease (PGD) which could progress to renal failure and is characterized by aberrant IgA immune complex deposition. Transferrin receptor1 (TFRC), an IgA receptor, is a potential RNA binding protein (RBP) which regulates expression of genes positively associated with the cell cycle and proliferation and is involved in IgAN. Molecular mechanisms by which TFRC affects IgAN development remain unclear.Methods: In this study, TFRC was overexpressed in human renal tubular mesangial cells (HRMCs) and RNA-sequencing (RNA-seq) and improved RNA immunoprecipitation sequencing (iRIP-seq) were performed. The aim was to identify potential RNA targets of TFRC at transcriptional and alternative splicing (AS) levels.Results: TFRC-regulated AS genes were enriched in mRNA splicing and DNA repair, consistent with global changes due to TFRC overexpression (TFRC-OE). Expression of TFRC-regulated genes potentially associated with IgAN, including CENPH, FOXM1, KIFC1, TOP2A, FABP4, ID1, KIF20A, ATF3, H19, IRF7, and H1-2, and with AS, CYGB, MCM7 and HNRNPH1, were investigated by RT-qPCR and iRIP-seq data analyzed to identify TFRC-bound RNA targets. RCC1 and RPPH1 were found to be TFRC-bound RNA targets involved in cell proliferation.Discussion: In conclusion, molecular TFRC targets were identified in HRMCs and TFRC found to regulate gene transcription and AS. TFRC is considered to have potential as a clinical therapeutic target

    Nurse educators perceptions of simulation teaching in Chinese context: benefits and barriers

    Background Although simulated teaching was introduced to China in the 1990s, it remains underused in nursing education. Determining how Chinese nurse educators feel about using simulation in their institutions is very important for faculty training and has the potential to influence simulation implementation. Method This cross-sectional descriptive study was undertaken to identify the nurse educators’ experiences in the use of simulation from various regions of China. One hundred and thirty-six nurse educators provided demographic data and information about simulation implementation within their institutions and explored the perceived barriers and benefits of simulation usage. Results The survey data shows that 108 participants have used simulation in their work, but less than 92 (67.6%) of the respondents had used this teaching strategy more than ten times in last year. The study identified four factors hindering nurse faculty from simulation adoption: (1) concerns with student readiness; (2) the need for faculty team-building for simulation teaching; (3) lack of adequate simulation resources; and (4) thoughtful integration of simulation into nursing curricula. Conclusions Study data suggest that faculty training programs for simulation should be based on the nurse educators’ training needs, including systematically designed training topics, and the provision of hands-on learning simulation activities with expert feedback to help nurse educators achieve the competencies required for effective simulation-based education

    Hexokinase1: A glucose sensor involved in drought stress response and sugar metabolism depending on its kinase activity in strawberry

    Hexokinase1 (HXK1) is a bifunctional enzyme that plays indispensable roles in plant growth, nitrogen utilization, and stress resistance. However, information on the HXK family members of strawberries and their functions in glucose sensing and metabolic regulation is scarce. In the present study, four HXKs were firstly identified in the genome of Fragaria vesca and F. pentaphylla. The conserved domains of the HXK1s were confirmed, and a site-directed mutation (S177A) was introduced into the FpHXK1. FpHXK1, which shares the highest identity with the AtHXK1 was able to restore the glucose sensitivity and developmental defects of the Arabidopsis gin2-1 mutant, but not its kinase-activity-impaired mutant (FpHXK1S177A). The transcription of FpHXK1 was dramatically up-regulated under PEG-simulated drought stress conditions. The inhibition of the HXK kinase activity delayed the strawberry plant’s responses to drought stress. Transient overexpression of the FpHXK1 and its kinase-impaired mutant differentially affected the level of glucose, sucrose, anthocyanins, and total phenols in strawberry fruits. All these results indicated that the FpHXK1, acting as a glucose sensor, was involved in drought stress response and sugar metabolism depending on its kinase activity

    In order to further explore the pollution characteristics of volatile organic compounds in ambient air in winter and spring in Shijiazhuang City, the pollution characteristics of 62 volatile organic compounds (VOCs), monthly and quarterly variation, the correlation between VOCs and PM2.5, and the main sources of VOCs are investigated by using EPA TO-15 method. It shows that 40 organic compounds of the 64 VOCs have been quantitatively determined in winter and spring in the city, which are mainly acetone, benzene, carbon tetrachloride, dichloromethane, toluene, ethyl acetate, etc.. In the no-quantitatively determined components, higher ethanol, butyl acetate, butane etc. are detected. The VOCs concentration has positive correlation with the PM2.5 concentration during haze days

    Human action recognition using 3D pose data has gained a growing interest in the field of computer robotic interfaces and pattern recognition since the availability of hardware to capture human pose. In this paper, we propose a fast, simple, and powerful method of human action recognition based on human kinematic similarity. The key to this method is that the action descriptor consists of joints position, angular velocity and angular acceleration, which can meet the different individual sizes and eliminate the complex normalization. The angular parameters of joints within a short sliding time window (approximately 5 frames) around the current frame are used to express each pose frame of human action sequence. Moreover, three modified KNN (k-nearest-neighbors algorithm) classifiers are employed in our method: one for achieving the confidence of every frame in the training step, one for estimating the frame label of each descriptor, and one for classifying actions. Additional estimating of the frame's time label makes it possible to address single input frames. This approach can be used on difficult, unsegmented sequences. The proposed method is efficient and can be run in real time. The research shows that many public datasets are irregularly segmented, and a simple method is provided to regularize the datasets. The approach is tested on some challenging datasets such as MSR-Action3D, MSRDailyActivity3D, and UTD-MHAD. The results indicate our method achieves a higher accuracy

    Ultrastructure characteristics and quality changes of low-moisture Chilgoza pine nut (Pinus gerardiana) during the near-freezing-temperature storage

    The effects of thermal treatment on the ultrastructure and quality of Chilgoza pine nuts stored at near-freezing temperature were investigated. The moisture content of pine nuts was adjusted to 13.3%, compared to the initial 17.3% moisture content. Thermal treatment improved the storage stability. The hydrolysis of lipids was delayed in low-moisture pine nuts, resulting in a significant inhibition of free fatty acid accumulation in pine nuts. Low-moisture pine nuts also showed lower peroxide and thiobarbituric acid (TBA)​ values compared with the control. Ultrastructure characterization revealed that thermal treatment maintained the cell integrity, deferred the degradation of the plasmalemma and protected the internal lipid droplet. Thermal treatment better retained the antioxidant components, including total phenolics and vitamin E, and reduced the activities of lipase and lipoxygenase in pine nuts. These results indicated that thermal treatment could retard the senescence and deterioration of quality in pine nuts stored at a near-freezing temperature.Se investigaron los efectos del tratamiento térmico en la ultrastructura y la calidad del piñón de pino Chilgoza almacenado a una temperatura cercana a congelación. El contenido de humedad de los piñones de este pino fue de 13,3%, en comparación con el contenido de humedad inicial de 17,3%. El tratamiento térmico mejoró la estabilidad de almacenamiento. La hidrólisis de los lípidos se retardó en los piñones de pino de baja humedad, resultando en una inhibición significativa de la acumulación de ácidos grasos libres en los piñones. Los piñones de pino de baja humedad también mostraron unos índices bajos de peróxido y TBA en comparación con la muestra control. La caracterización de la ultrastructura reveló que el tratamiento térmico mantuvo la integridad de las células, atrasó la degradación de la plasmalema y protegió la gota lipídica interna. El tratamiento térmico retuvo de forma más favorable los componentes antioxidantes, como el total fenólico y la vitamina E, además redujo la actividad de la lipasa y la lipoxigenasa en los piñones. Estos resultados indicaron que el tratamiento térmico podría retardar la senescencia y la deterioración de la calidad de los piñones almacenados a una temperatura cercana a la congelación

    Abstract Lung cancer, a prevalent and aggressive disease, is characterized by recurrence and drug resistance. It is essential to comprehend the fundamental processes and discover novel therapeutic objectives for augmenting treatment results. Based on our research findings, we have identified a correlation between methylation of cg09897064 and decreased expression of ZBP1, indicating a link to unfavorable prognosis in patients with lung cancer. Furthermore, these factors play a role in macrophage polarization, with ZBP1 upregulated in M1 macrophages compared to both M0 and M2 polarized macrophages. We observed cg09897064 methylation in M2 polarization, but not in M0 and M1 polarized macrophages. ATACseq analysis revealed closed chromatin accessibility of ZBP1 in M0 polarized macrophages, while open accessibility was observed in both M1 and M2 polarized macrophages. Our findings suggest that ZBP1 is downregulated in M0 polarized macrophages due to closed chromatin accessibility and downregulated in M2 polarized macrophages due to cg09897064 methylation. Further investigations manipulating cg09897064 methylation and ZBP1 expression through overexpression plasmids and shRNAs provided evidence for their role in modulating macrophage polarization and tumor growth. ZBP1 inhibits M2 polarization and suppresses tumor growth, while cg09897064 methylation promotes M2 polarization and macrophage-induced tumor growth. In mechanism investigations, we found that cg09897064 methylation impairs CEBPA binding to the ZBP1 promoter, leading to decreased ZBP1 expression. Clinical experiments were conducted to validate the correlation between methylation at cg09897064, ZBP1 expression, and macrophage M2 polarization. Targeting these factors may hold promise as a strategy for developing innovative checkpoint inhibitors in lung cancer treatment
