    Preparing and characterizing Fe3O4@cellulose nanocomposites for effective isolation of cellulose-decomposing microorganisms

    This study developed Fe3O4@cellulose nanocomposites by co-precipitation synthesis for bacteria capture and isolation. By surface modification with cellulose, the Fe3O4@cellulose nanocomposites have 20 nm average particle size and 3.3–24.9 emu/g saturation magnetization. Living bacteria could be captured by the Fe3O4@cellulose nanocomposites and harvested by magnetic field, with high efficiency (95.1%) and stability (>99.99%). By metabolizing cellulose and destroying the Fe3O4@cellulose@bacteria complex, cellulose-decomposing microorganisms lost the magnetism. They were therefore able to be isolated from the inert microbial community and the separation efficiency achieved over 99.2%. This research opened a door to cultivate the uncultivable cellulose-decomposing microorganisms in situ and further characterize their ecological functions in natural environment

    Theileria annulata Cyclophilin1 (TaCyp1) Interacts With Host Cell MED21

    Host cells infected by Theileria annulata schizonts show the character of permanent proliferation in vitro, also named transformation. To explore the molecular mechanism a T. annulata Cyp1 (TaCyp1) protein potentially involved in regulating cell transformation was used as bait to screen for its interacting proteins by yeast-two-hybrid assay. Additional GST-pull down experiments confirmed that only MED21 specifically interacted with TaCyp1. Moreover, the distribution of TaCyp1 around T. annulata schizonts facilitated interaction with host cell MED21. As a component of mediator complex, MED21 is normally involved in regulating the transcription of nearly all RNA polymerase II-dependent genes. Therefore, to explore its influence on NF-κB signaling MED21 RNA interference and parasite killing with BW720c treatment were performed. Knock down of MED21 resulted in a significant decrease in NF-κB1/2 mRNA expressions, but no significant change in P105, P52 levels, nor detectable alteration in levels of phosphorylated IκBα/β. By contrast, BW720c treatment induced an obvious decrease in the phosphorylation status of P52 and IκBα/β, but no obvious change in that of P105. This suggests that BW720c-induced parasite death had a significant negative influence on NF-κB signaling, whereas knock down of MED21 had no obvious effect on NF-κB signaling. Characterization of TaCyp1 provides information on the function of parasite cyclophilins and leads to a better understanding of the interactions between T. annulata and its host leukocytes

    Breast cancer survival analysis with molecular subtypes : an initial step

    As a predominant threat to women's health world-wide, breast cancer has become increasingly important in on-cology research. The discovery of molecular subtypes of breast cancer has led to more subtype oriented treatment and prognosis prediction. Effective prognosis models help to estimate the recurrence as well as the quality and duration of survival, leading to more personalized treatments. However, most traditional prognostic models either ignore molecular subtypes or only make limited use of them. The roles of molecular subtypes in the development and treatment of breast cancer have not been fully revealed. With the over 1200 cases collected by Sir Run Run Shaw Hospital of Zhejiang University in the past two decades, we aim to improve understanding of molecular subtypes and their impacts on the prognosis via data analysis in the long run. As the initial stage, this short paper presents our preliminary work of logistic regression experiments with the data. Though molecular subtypes have not been included the tentative model, they are to be explored further in following stages

    Testicular Characteristics and the Block to Spermatogenesis in Mature Hinny

    Most hinnies (female donkey×male horse) and mules (female horse×male donkey) are sterile with few reports of equine fertile hybrids. The main cause of this sterility is thought to be a meiotic block to spermatogenesis and oogenesis. This study compared the developmental features of the testes and a histological analyses of spermatogenesis in a male hinny with those of a normal, fertile stallion and Jack donkey. Hinny testes showed a thicker tunica albuginea, fewer blood vessels and more connective tissue in the testis parenchyma than those of the stallion and Jack donkey. Although the mean number of seminiferous tubules was significantly higher in stallion and hinny than Jack donkey (p<0.01), the mean proportion of seminiferous tubules was lower in the hinny (p<0.01) which resulted in a smaller diameter of seminiferous tubules. The mean number of spermatogonia and spermatocytes per unit area were significantly lower in hinny testis (p<0.01) and no spermatids or mature spermatozoa cells were found during immunofluorescent analyses. These results indicated that defects in seminiferous tubule development and structure occur in the testis of hinnies. Furthermore, most spermatogonia and spermatocytes cease development in synapsis during mid-meiosis of spermatocytes, which results in a block to spermatogenesis that prevents the formation of spermatids and matured spermatozoa during meiosis in male hinnies

    Prevalence of and Risk Factors for Peripheral Neuropathy in Chinese Patients With Diabetes: A Multicenter Cross-Sectional Study

    Diabetic peripheral neuropathy (DPN) is the most common complication of diabetes, and its progression significantly worsens the patient's quality of life. This study investigated the prevalence and risk factors associated with DPN in a large sample of Beijing individuals with type 1 and 2 diabetes, as well as compared the diagnostic methods for DPN. A total of 2,048 diabetic patients from 13 centers in Beijing were assessed for DPN through questionnaires and examination. Patients were divided into DPN group and suspected DPN/non-DPN group. The demographic, clinical and biological characteristics between the two groups were compared. Binary logistic regression analysis was performed to identify potential variables associated with DPN in diabetic patients. The diagnostic methods for DPN were also compared. Among the 2,048 diabetic patients, 73 cases of type 1 diabetes mellitus, 1,975 cases of type 2 diabetes were included in this study. Among them, 714 (34.86%) were identified with DPN, 537 (26.22%) were suspected of having DPN, and 797 (38.92%) were identified without DPN. Patient's age, duration of diabetes, and diabetic retinopathy were the significant independent risk factor for DPN among patients with type 2 diabetes. The odds ratio (OR) was 1.439 (95% confidence interval (CI): 1.282–1.616, P &lt; 0.001), 1.297 (95% CI: 1.151–1.462, P &lt; 0.001), and 0.637 (95% CI: 0.506–0.802, P &lt; 0.001), respectively. Ankle reflex, temperature sensation plus vibration sensation are the best screening test for patients with type 1 and 2 diabetes. The Youden indexes were 62.2 and 69.8%, respectively. The prevalence rates of DPN in the Chinese patients with type 1 and type 2 diabetes in Beijing were 21.92 and 35.34%, respectively. Patient's age, duration of diabetes, and diabetic retinopathy are the independent risk factors for DPN

    Prediction of Crack Resistance of LFSMA-13 with and without Anti-Rut Agent Using Parameters of FTIR Spectrum under Different Aging Degrees

    This paper aims to better analyze the crack resistance of lignin fiber reinforced SMA-13 (LFSMA-13) asphalt mixtures, with and without polymer anti-rut agent (ARA), under different aging degrees. IDEAL-CT test and Fourier transform infrared (FTIR) spectroscopy were utilized to analyze the relationships between the crack resistance of LFSMA-13, with and without ARA, and the parameters of the FTIR spectrum of the asphalt extracted from the test samples. A convenient testing method to predict the anti-crack ability of the mixtures in a road was also derived in this study. The test samples were prepared using the specifications listed by AASHTO. The fracture formation work (Winitial) and cracking index (CTIndex) in the IDEAL-CT test were adopted to reflect the cracking ability of the asphalt mixtures in both the crack formation stage and the crack propagation stage. The peak areas of the FTIR spectrum were utilized to reveal the chemical properties of the asphalt material inside the SMA-13 asphalt mixtures, with and without ARA under different aging degrees. Grey correlation analysis was adopted to choose the most suitable FTIR spectrum parameters to derive the prediction models of Winitial and CTIndex under different aging degrees. After conducting a series of tests, the results showed that the aging process could well affect the crack resistance of the test samples and the peak areas of the asphalt extracted from the mixtures. The FTIR parameters selected from the grey correlation analysis could be used to well predict the anti-crack ability of the asphalt mixtures

    Design of a Variable Stiffness Gecko-Inspired Foot and Adhesion Performance Test on Flexible Surface

    Adhesion robots have broad application prospects in the field of spacecraft inspection, repair, and maintenance, but the stable adhesion and climbing on the flexible surface covering the spacecraft has not been achieved. The flexible surface is easily deformed when subjected to external force, which makes it difficult to ensure a sufficient contact area and then detach from it. To achieve stable attachment and easy detachment on the flexible surface under microgravity, an adhesion model is established based on the applied adhesive material, and the relationship between peeling force and the rigidity of the base material, peeling angle, and working surface stiffness is obtained. Combined with the characteristics of variable stiffness structure, the adhesion and detachment force of the foot is asymmetric. Inspired by the adhesion-detachment mechanism of the foot of the gecko, an active adhesion-detachment control compliant mechanism is designed to achieve the stable attachment and safe detachment of the foot on the flexible surface and to adapt to surfaces with different rigidity. The experimental results indicate that a maximum normal adhesion force of 7.66 N can be generated when fully extended, and the safe detachment is achieved without external force on a flexible surface. Finally, an air floating platform is used to build a microgravity environment, and the crawling experiment of a gecko-inspired robot on a flexible surface under microgravity is completed. The experimental results show that the gecko-inspired foot with variable stiffness can satisfy the requirements of stable crawling on flexible surfaces

    Screening and identification of host proteins interacting with Theileria annulata cysteine proteinase (TaCP) by yeast-two-hybrid system

    Abstract Background Theileria annulata can infect monocytes/macrophages and B lymphocytes and causes severe lymphoproliferative disease in ruminants. Meanwhile, infection by T. annulata leads to the permanent proliferation of cell population through regulating signaling pathways of host cells. Cysteine proteinases (CPs) are one kind of protein hydrolase and usually play critical roles in parasite virulence, host invasion, nutrition and host immune response. However, the biological function of T. annulata CP (TaCP) is still unclear. In this study, a yeast-two-hybrid assay was performed to screen host proteins interacting with TaCP, to provide information to help our understanding of the molecular mechanisms between T. annulata and host cells. Methods The cDNA from purified bovine B cells was inserted into pGADT7-SfiI vector (pGADT7-SfiI-BcDNA, Prey plasmid) for constructing the yeast two-hybrid cDNA library. TaCP was cloned into the pGBKT7 vector (pGBKT7-TaCP) and was considered as bait plasmid after evaluating the expression, auto-activation and toxicity tests in the yeast strain Y2HGold. The yeast two-hybrid screening was carried out via co-transforming bait and prey plasmids into yeast strain Y2HGold. Sequences of positive preys were analyzed using BLAST, Gene Ontology, UniProt and STRING. Results Two host proteins, CRBN (Bos taurus cereblon transcript variant X2) and Ppp4C (Bos indicus protein phosphatase 4 catalytic subunit) were identified to interact with TaCP. The results of functional analysis showed that the two proteins were involved in many cellular processes, such as ubiquitylation regulation, microtubule organization, DNA repair, cell apoptosis and maturation of spliceosomal snRNPs. Conclusions This study is the first to screen the host proteins of bovine B cells interacting with TaCP, and 2 proteins, CRBN and Ppp4C, were identified using yeast two-hybrid technique. The results of functional analysis suggest that the two proteins are involved in many cellular processes, such as ubiquitylation regulating, microtubule organization, DNA repair, cell apoptosis and maturation of spliceosomal snRNPs. The interaction with CRBN and Ppp4C indicate that TaCP possibly is involved in regulating signaling pathways and cell proliferation, which is helpful for understanding the interaction between T. annulata and host cells
