    THE ANTIBACTERIAL ACTIVITY OF CLAUSENA ANISATA HOOK, A SOUTH AFRICAN MEDICINAL PLANT

    Background: Clausena anisata Hook also known as Iperepesi in Xhosa language is a medicinal plant used traditionally for the treatment of various ailments and some opportunistic infections associated with tuberculosis (TB). Patients in South Africa based on the phytotherapeutic information on this species in the Eastern Cape, use this medicinal plant. Hence, the antibacterial activity of various solvent extracts of the leaves and barks were respectively, evaluated using selected bacterial strains. Method: The leaves and stem bark were tested against 10 selected strains of Gram - positive and Gram - negative bacteria through the agar dilution method. Acetone, dichloromethane and water extracts were used for the extraction. MIC was determined at different concentrations (0.1mg/ml, 0.5mg/ml, 1mg/ml and 5mg/ml) and the results obtained were compared to that of standard antibiotics. Result: The acetone extract of the leaves were more active against both Gram-positive and Gram –negative bacteria with MIC ranging from 0.1 mg/ml - 0.5 mg / ml. The dichloromethane extract of the bark showed appreciable activities against Staphylococcus aureus (ATCC 6538) (MIC: 0.1mg /ml) Escherichia coli and, Streptococcus pyogenes with an MIC of 5mg/ml respectively. On the other hand, the aqueous extract of the leaves showed no activity against the tested organisms with the exception of the aqueous bark extract which inhibited Staphylococcus aureus (MIC: 0.5mg/ml) and Pseudomonas aeruginosa (MIC: 5mg/ml). Conclusion: This study confirmed the antibacterial activities of acetone extract of the leaves of Clausena anisata. The capability of this extract to inhibit both Gram positive and negative bacteria is an indication that the extract is a potential broad spectrum antibacterial. The result of this study further justified its indigenous use for the treatment of bacteria commonly associated with TB especially among the people of Nkonkobe Municipality

    Assessment of the medicinal potentials of the methanol extracts of the leaves and stems of Buddleja saligna

    <p>Abstract</p> <p>Background</p> <p><it>Buddleja saligna </it>Willd (Loganiaceae) is a small to medium-sized evergreen tree; trunk short, often gnarled and crooked; crown dense, rounded or domed-shaped; foliage greyish green. The wild olives are traditionally used to lower blood pressures in many parts of the world. In southern Africa, bark and leaf decoctions are used to treat colic, coughs, colds, sore eyes, urinary problems and as purgatives.</p> <p>Methods</p> <p>The antibacterial, antioxidant activities and phenolic contents of the methanol extracts of the leaves and stems of <it>Buddleja saligna </it>were evaluated using <it>in vitro </it>standard methods. Spectrophotometry was the basis for the determinations of total phenol, total flavonoids, flavonols, and proanthocyanidins. Tannins, quercetin and catechin equivalents were used for these parameters. The antioxidant activities of the leaves and stem extracts of <it>Buddleja saligna </it>were determined by ABTS, DPPH, and ferrous reducing antioxidant property (FRAP) methods. Laboratory isolates of 10 bacteria species which included five Gram-positive and five Gram-negative strains were used to assay for antibacterial activity of this plant.</p> <p>Results</p> <p>The antioxidant activities of the leaves as determined by the ABTS and DPPH were similar to that of the stem. The flavonoids and the flavonols contents of the leaves were higher than that of the stem but the total phenols, proanthocyanidins and FRAP activities were higher in the methanol extracts of the stem. The extracts did show activity against both Gram-positive and Gram-negative bacteria. For instance, while the methanol extract of the leaves showed good activities on all the organisms except <it>Serratia marcescens </it>and <it>Pseudomonas aeruginosa </it>at MICs of between 2.5 and 5.0 mg/ml, the extract of the stem only showed activities on <it>Bacillus cereus, Streptococcus pyrogens </it>and <it>Pseudomonas aeruginosa </it>at the same concentration.</p> <p>Conclusion</p> <p>The results from this study indicate that the leaves and stem extracts of <it>Buddleja saligna </it>possess antioxidant properties and could serve as free radical inhibitors or scavenger or, acting possibly as primary antioxidants. Although, the antibacterial properties of <it>Buddleja saligna </it>are not as effective as the standard drugs-Chloramphenicol and Streptomycin, they still possess some activity against bacterial strains used in this study. <it>Buddleja saligna </it>may therefore be a good candidate for functional foods as well as pharmaceutical plant-based products.</p

    Phenolic content and antioxidant property of the bark extracts of <it>Ziziphus mucronata </it>Willd. subsp. <it>mucronata </it>Willd

    <p>Abstract</p> <p>Background</p> <p>Several plants traditionally used in treatment of a variety of infections in South Africa are reported in ethnobotanical surveys. Many of these plants including <it>Ziziphus mucronata </it>subsp. <it>mucronata </it>lack scientific reports to support their medicinal importance.</p> <p>Methods</p> <p>The antioxidant activities and phenolic contents of the acetone, ethanol and aqueous extracts of the stems of <it>Z. mucronata </it>subsp. <it>mucronata </it>were evaluated using <it>in vitro </it>standard methods. The total phenol, total flavonoids and proanthocyanidin content were determined spectrophotometrically. Quercetin, Tannic acid and catechin equivalents were used for these parameters. The antioxidant activities of the stem bark extracts of this plant were determined by ABTS, DPPH, and ferrous reducing antioxidant property (FRAP) methods.</p> <p>Results</p> <p>The quantity of the phenolic compounds, flavonoids and proanthocyanidins detected differ significantly in the various extracts. The phenolics were significantly higher than the flavonoids and proanthocyanidin contents in all the extracts investigated. The ferric reducing ability and the radical scavenging activities of the extracts were very high and dose-dependent. The ethanol extract had the highest antioxidant activity, followed by the acetone extract while the aqueous extract was the least active. Reacting with ABTS, the 50% inhibitory concentrations (IC<sub>50</sub>) were (0.0429 ± 0.04 mg/ml) for aqueous, (0.0317 ± 0.04 mg/ml) for acetone and (0.0306 ± 0.04 mg/ml) for ethanol extracts while they inhibited DPPH radical with 50% inhibitory concentration (IC<sub>50</sub>) values of 0.0646 ± 0.02 mg/ml (aqueous), 0.0482 ± 0.02 mg/ml (acetone) and 0.0422 ± 0.03 mg/ml (ethanol).</p> <p>Conclusions</p> <p>A correlation between the antioxidant activity and the total phenolic contents of the extracts indicated that phenolic compounds were the dominant contributors to the antioxidant activity of the plant. This study, therefore, demonstrated that <it>Z. mucronata </it>subsp<it>. mucronata </it>has strong antioxidant property and free radical scavenging capability.</p
