    A role for NRAGE in NF-κB activation through the non-canonical BMP pathway

    <p>Abstract</p> <p>Background</p> <p>Previous studies have linked neurotrophin receptor-interacting MAGE protein to the bone morphogenic protein signaling pathway and its effect on p38 mediated apoptosis of neural progenitor cells via the XIAP-Tak1-Tab1 complex. Its effect on NF-κB has yet to be explored.</p> <p>Results</p> <p>Herein we report that NRAGE, via the same XIAP-Tak1-Tab1 complex, is required for the phosphorylation of IKK -α/β and subsequent transcriptional activation of the p65 subunit of NF-κB. Ablation of endogenous NRAGE by siRNA inhibited NF-κB pathway activation, while ablation of Tak1 and Tab1 by morpholino inhibited overexpression of NRAGE from activating NF-κB. Finally, cytokine profiling of an NRAGE over-expressing stable line revealed the expression of macrophage migration inhibitory factor.</p> <p>Conclusion</p> <p>Modulation of NRAGE expression revealed novel roles in regulating NF-κB activity in the non-canonical bone morphogenic protein signaling pathway. The expression of macrophage migration inhibitory factor by bone morphogenic protein -4 reveals novel crosstalk between an immune cytokine and a developmental pathway.</p

    Creating restoration landscapes: partnerships in large-scale conservation in the UK

    It is increasingly recognized that ecological restoration demands conservation action beyond the borders of existing protected areas. This requires the coordination of land uses and management over a larger area, usually with a range of partners, which presents novel institutional challenges for conservation planners. Interviews were undertaken with managers of a purposive sample of large-scale conservation areas in the UK. Interviews were open-ended and analyzed using standard qualitative methods. Results show a wide variety of organizations are involved in large-scale conservation projects, and that partnerships take time to create and demand resilience in the face of different organizational practices, staff turnover, and short-term funding. Successful partnerships with local communities depend on the establishment of trust and the availability of external funds to support conservation land uses. We conclude that there is no single institutional model for large-scale conservation: success depends on finding institutional strategies that secure long-term conservation outcomes, and ensure that conservation gains are not reversed when funding runs out, private owners change priorities, or land changes hands

    Tracking random finite objects using 3D-LIDAR in marine environments

    This paper presents a random finite set theoretic formulation for multi-object tracking as perceived by a 3D-LIDAR in a dynamic environment. It is mainly concerned with the joint detection and estimation of the unknown and time varying number of objects present in the environment and the dynamic state of these objects, given a set of measurements. This problem is particularly challenging in cluttered dynamic environments such as in urban settings or marine environments, because, given a measurement set, there is absolutely no knowledge of which object generated which measurement, and the detected measurements are indistinguishable from false alarms. The proposed approach to multi-object tracking is based on the rigorous theory of finite set statistics (FISST). The optimal Bayesian multi-object tracking is not yet practical due to its computational complexity. However, a practical alternative to the optimal filter is the probability hypothesis density (PHD) filter, that propagates the first order statistical moment of the full multi-object posterior distribution. In contrast to classical approaches, this random finite set framework does not require any explicit data associations. In this paper, a Gaussian mixture approximation of the PHD filter is applied to track variable number of objects from 3D-LIDAR measurements by estimating both the number of objects and their respective locations in each scan.     Innate lymphoid cells (ILCs) are tissue-resident sentinels that are essential for early host protection from pathogens at initial sites of infection. However, whether pathogen-derived antigens directly modulate the responses of tissue-resident ILCs has remained unclear. In the present study, it was found that liver-resident type 1 ILCs (ILC1s) expanded locally and persisted after the resolution of infection with mouse cytomegalovirus (MCMV). ILC1s acquired stable transcriptional, epigenetic and phenotypic changes a month after the resolution of MCMV infection, and showed an enhanced protective effector response to secondary challenge with MCMV consistent with a memory lymphocyte response. Memory ILC1 responses were dependent on the MCMV-encoded glycoprotein m12, and were independent of bystander activation by proinflammatory cytokines after heterologous infection. Thus, liver ILC1s acquire adaptive features in an MCMV-specific manner
