Impact of Renal of Lymphatic Hyperplasia on Blood Pressure Regulation in Male Mice

Chronic high blood pressure, or hypertension, is identified as a risk factor for heart disease, stroke, and chronic kidney disease. The primary cause of most hypertension is increased peripheral vascular resistance that is controlled, in large part, by the kidney’s water handling. In the kidney, specific immune cell subsets and overall renal inflammation have both been identified as drivers of hypertension in preclinical models. Lymphatic vessels serve as a route of both fluid and immune cell clearance and their expansion, lymphangiogenesis, is necessary for the resolution of tissue inflammation. We hypothesized that by increasing renal lymphangiogenesis, renal inflammation would be reduced and blood pressure would be normalized during salt-sensitive hypertension. To investigate the role of renal immune cell trafficking in instances of blood pressure challenge, we employed a murine model of inducible renal lymphatic expansion. Mice expressing a lymphatic growth factor, vascular endothelial growth factor (VEGF)-D, under the control of a TRE-promoter were crossed with mice expressing a Kidney Specific Protein-regulated transactivator (rtTA). Upon administration of doxycycline, the rtTA-dox complex binds to the TRE promoter region, causing transcription of VEGF-D only in renal tubular epithelial cells. The resultant kidney-specific VEGF-D overexpression caused expansion of the existing renal lymphatic network in addition to generating de novo lymphangiogenesis in the cortex of “Kid-VD” mice. We then utilized the Kid-VD mouse model during an established rodent hypertension regimen of nitric oxide inhibition and high salt diet loading to identify the impact – through weekly blood pressure measures - and mechanism – by cellular, protein, and RNA analysis – of expanded renal lymphatics on blood pressure regulation. Lymphatic circulation may thus provide a new target for the treatment of chronic hypertension and its associated co-morbidities

A Novel Approach in Pigment Printing Using Nano-keratin Based Binder

    Get PDF
This study is devoted to preparation and characterization of nano-keratin based binder (NKBB) from cheap renewable natural resources, namely coarse Egyptian wool or feather. The prepared NKBB is utilized as a biodegradable, environment-friendly and relatively cheap binder in textile pigment printing of polyester, pure polyacrylic, viscose, polyester/viscose, and polyester/acrylic fabrics. Different concentrations of the prepared NKBB as well as its mixture with commercially produced one are used in the pigment printing paste. The colour strength of the printed fabrics as well as their fastness properties to light, washing, and perspiration are evaluated. The effect of the used binder on the bending stiffness of the printed fabrics is also assessed. Results show that the NKBB gives almost the same colour strength and fastness properties as the commercial binder with improved stiffness of the printed samples in relation to that printed with commercial one

Bone metastatic prostate cancer and resistance to tyrosine kinase inhibitors: an intimate relationship between loss of miR-203 and up-regulation of EGFR signaling: DOI: 10.14800/rd.345

A tumor suppressor role for miR-203 in RAS-dependent prostate cancer metastasis has been described recently by our group. We have explored the regulatory mechanisms by which miR-203 is being regulated through EGFR signaling. We investigated the molecular mechanism of metastasis and identified novel roles of genes that interact with miR-203 downstream of activated Ras. We showed an amplifying regulatory loop involving the direct interaction of miR-203 with the EGFR ligands, AREG, EREG, and TGFA 3’UTR. Using clinical specimens and database analysis, our data suggested that decreasing miR-203 and increasing EGFR ligands, AREG, EREG, and TGFA expressions are correlated with prostate cancer progression. Since tyrosine kinase inhibitors (TKIs) have been shown to inhibit tyrosine phosphorylation of EGFR in a dose-dependent manner, we examined a role for miR-203 in TKIs-induced apoptosis in RAS-activated prostate cancer. We investigated the mechanisms by which miR-203 overexpression contributes to TKIs-resistant RAS-activated prostate cancer cells apoptosis. We have shown indications for candidate miR-203 targets that are either influenced by anti-apoptotic proteins (e.g. API5, BIRC2, and TRIAP1) or positively influenced by a novel NF-?B-inducible oncogenic molecule, TNFAIP8. These observations suggest that the latter category may be synergistically affected by the regulatory loop of miR-203 depletion and anti-apoptotic proteins overexpression. Our results provided evidence showing a role of miR-203 in regulating the expression of EGFR signaling genes in response to TKIs-resistance during prostate cancer progression

Flexural resistance of the polypropylene fibres reinforced cement mixes with waste material

    Get PDF
The polypropylene (PP) fibers in shotcrete has been used for ground support and building strengthening, since several decades. However, the recent trend is to use the waste material in cementbased mixes to produce an eco-friendly material. Such waste material is the incineration fly ash (FA) that is classified as a hazardous product. This study is intended to establish the mechanical properties of fiber reinforced mortar in addition to cement or sand partial replacement by fly ash, in terms of flexural strength testing. The mechanical properties reflect the influence of the dosage of fiber content and the proportion of the fly ash on the flexural strength. The percentage of cement or sand was replaced by 0, 10, 20, and 30% fly ash. The dosage of fibers was 0, 0.6, 1.2, and 1.8 kg/m3. This green mix with fibers provides a partial substitute of cement as it is cheaper, by incorporating waste product, and saving energy consumption in the production. Due to growing interest in sustainable construction, engineers and architects are motivated to choose such materials which are more sustainable

REVISITING PREHISTORIC ARCHEOLOGICAL SITES: ENVISIONING FIRST BUILT ENVIRONMENTS TO REPOSSESS GEOGRAPHICALLY SPECIFIC APPROACHES IN ARCHITECTURE

    Get PDF
Since Prehistoric times, architecture had been a human response to an occurring natural setting. Starting from places of dwelling to buildings that no longer only serve physical requirements for survival. Architectural languages were approached initially as an expression of culture, evolution, and growth of a community within a natural setting. This response resulted in the creation of built environments, humanity’s decision to become sedentary. This decision took place in the Late Stone age, a key phase in our timeline. First built environments were born in a time known as the Neolithic revolution, which shown itself as humans transitioned from hunter-gatherer societies to agrarian based ones. Once Nature shaped man, now man shapes nature. We observe the dynamic created between man the hosting setting. While observing this two sided complex operating system through revisiting prehistoric archeological sites, we can notice how various geographic zones birthed a diversity of built environments. However, by observing the paradigm of duality in our modern world, we can see neglected neighborhoods and cities, and more importantly that we failed to fulfill our fair part of shaping our natural setting and this reflected on the state of our communities as well. By repossessing how to be geographically specific in architecture, we can set the parameters to architectural planning that includes nature as a co-partner and as a result attempt to improve the well-being of our neglected communities
