Using tactical and operational factors to assess strategic alignment: an SME study
The strategic use of Information Technology (IT), better known as strategic alignment, has significantly increased, as a result of the strong dependence of organisational activity on Information Systems (IS) and their related technologies. Strategic alignment is considered as a key element to improve performance on organisations, enhance efficiency and allow organisations to be more competitive in their respective industry. One of the first steps towards achieving strategic alignment is to have adequate means to measure it. Current assessment approaches, though, are mainly focused at the strategic level but provide little insight at tactical and operational levels, which are recognized as important areas for achieving strategic alignment. Furthermore, most of the existing approaches are tested in large organisations and there is little research on assessing the effectiveness of these approaches for Small and Medium Enterprises (SMEs). This paper proposes an alternative instrument that rather than focusing only at the strategic level it aims to have a better understanding by measuring alignment at tactical and operational levels. Training senior employees for ICT skills enhancement through ârefocusâ: The EUROPEAN project
Declining fertility rates and the ageing of the European population represents a major challenge to many governments within the European Union (EU) and its neighbouring realms, in the present and for the forthcoming decades. The increasing ageing workforce within several United Kingdom (UK) and the EU private and public organisations is an evident manisfestation. The workforce comprised of senior employees are often seen by employers as less mobile and flexible with low performance and thus, less able to deal with the rapid changes occurring in their organisations; as rapid developments require flexible and responsive workforce. The mobility of the senior employees is therefore a topical theme in the workforce market. The necessary knowledge and skills required for the senior employees can be taught through various Information and Communication Technologies (ICT) related training systems within an organisation. However, it appears that the development of training systems required for senior employees need careful investigation into the requirements of the problem situation. The reason is that the design of senior employees training systems should be more carefully planned, as literature indicates that it is significantly time consuming and is harder to train senior employees on ICT applications. This paper presents a European project â REFOCUS (oldeR Employees training on inFOrmation and CommUnication technologieS), an initaitive undertaken by the EU for enhancing the ICT skills of the ageing workforce. This project also aims at designing a new electronic learning (e-Learning) method that focuses on the needs of senior employees of 40 years of age and over â who can become a strategic resource for the European economy
An Integer Programming Approach to the Student-Project Allocation Problem with Preferences over Projects
The Student-Project Allocation problem with preferences over Projects (SPA-P) involves sets of students, projects and lecturers, where the students and lecturers each have preferences over the projects. In this context, we typically seek a stable matching of students to projects (and lecturers). However, these stable matchings can have different sizes, and the problem of finding a maximum stable matching (MAX-SPA-P) is NP-hard. There are two known approximation algorithms for MAX-SPA-P, with performance guarantees of 2 and 32 . In this paper, we describe an Integer Programming (IP) model to enable MAX-SPA-P to be solved optimally. Following this, we present results arising from an empirical analysis that investigates how the solution produced by the approximation algorithms compares to the optimal solution obtained from the IP model, with respect to the size of the stable matchings constructed, on instances that are both randomly-generated and derived from real datasets. Our main finding is that the 32 -approximation algorithm finds stable matchings that are very close to having maximum cardinality
Popularity versus Similarity in Growing Networks
Popularity is attractive -- this is the formula underlying preferential
attachment, a popular explanation for the emergence of scaling in growing
networks. If new connections are made preferentially to more popular nodes,
then the resulting distribution of the number of connections that nodes have
follows power laws observed in many real networks. Preferential attachment has
been directly validated for some real networks, including the Internet.
Preferential attachment can also be a consequence of different underlying
processes based on node fitness, ranking, optimization, random walks, or
duplication. Here we show that popularity is just one dimension of
attractiveness. Another dimension is similarity. We develop a framework where
new connections, instead of preferring popular nodes, optimize certain
trade-offs between popularity and similarity. The framework admits a geometric
interpretation, in which popularity preference emerges from local optimization.
As opposed to preferential attachment, the optimization framework accurately
describes large-scale evolution of technological (Internet), social (web of
trust), and biological (E.coli metabolic) networks, predicting the probability
of new links in them with a remarkable precision. The developed framework can
thus be used for predicting new links in evolving networks, and provides a
different perspective on preferential attachment as an emergent phenomenon
Super-stability in the Student-Project Allocation Problem with Ties
The Student-Project Allocation problem with lecturer preferences over Students ( Open image in new window ) involves assigning students to projects based on student preferences over projects, lecturer preferences over students, and the maximum number of students that each project and lecturer can accommodate. This classical model assumes that preference lists are strictly ordered. Here, we study a generalisation of Open image in new window where ties are allowed in the preference lists of students and lecturers, which we refer to as the Student-Project Allocation problem with lecturer preferences over Students with Ties ( Open image in new window ). We investigate stable matchings under the most robust definition of stability in this context, namely super-stability. We describe the first polynomial-time algorithm to find a super-stable matching or to report that no such matching exists, given an instance of Open image in new window . Our algorithm runs in O(L) time, where L is the total length of all the preference lists. Finally, we present results obtained from an empirical evaluation of the linear-time algorithm based on randomly-generated Open image in new window instances. Our main finding is that, whilst super-stable matchings can be elusive, the probability of such a matching existing is significantly higher if ties are restricted to the lecturersâ preference lists
Who settles for less? Subjective dispositions, objective circumstances, and housing satisfaction
In recent years there has been growing interest in individualsâ self-perceptions of their wellbeing on the grounds that these complement well-established objective indicators of welfare. However, individualsâ assessments depend on both objective circumstances and subjective, idiosyncratic dispositions, such as aspirations and expectations. We add to the literature by formulating a modelling strategy that uncovers how these subjective dispositions differ across socio-demographic groups. This is then tested using housing satisfaction data from a large-scale household panel survey from Australia. We find that there are significant differences in the way in which individuals with different characteristics rate the same objective reality. For instance, male, older, migrant, and Indigenous individuals rate equal housing conditions more favourably than female, younger, Australian-born, and non-Indigenous individuals. These findings have important implications for how self-reported housing satisfaction, and wellbeing data in general, are to be used to inform evidence-based policy
Phenotypic redshifts with self-organizing maps: A novel method to characterize redshift distributions of source galaxies for weak lensing
Wide-field imaging surveys such as the Dark Energy Survey (DES) rely on
coarse measurements of spectral energy distributions in a few filters to
estimate the redshift distribution of source galaxies. In this regime, sample
variance, shot noise, and selection effects limit the attainable accuracy of
redshift calibration and thus of cosmological constraints. We present a new
method to combine wide-field, few-filter measurements with catalogs from deep
fields with additional filters and sufficiently low photometric noise to break
degeneracies in photometric redshifts. The multi-band deep field is used as an
intermediary between wide-field observations and accurate redshifts, greatly
reducing sample variance, shot noise, and selection effects. Our implementation
of the method uses self-organizing maps to group galaxies into phenotypes based
on their observed fluxes, and is tested using a mock DES catalog created from
N-body simulations. It yields a typical uncertainty on the mean redshift in
each of five tomographic bins for an idealized simulation of the DES Year 3
weak-lensing tomographic analysis of , which is a
60% improvement compared to the Year 1 analysis. Although the implementation of
the method is tailored to DES, its formalism can be applied to other large

Activity of the antiarrhythmic drug amiodarone against Leishmania (L.) infantum: an in vitro and in vivo approach
<div><p>Abstract Background: Considering the high toxicity and limited therapies available for treating visceral leishmaniasis (VL), the drug repositioning approach represents a faster way to deliver new therapies to the market. Methods: In this study, we described for the first time the activity of a potent antiarrhythmic, amiodarone (AMD), against L. (L.)infantum and its in vitro and in vivo activity. Results: The evaluation against promastigotes has shown that amiodarone presents leishmanicidal effect against the extracellular form, with an IC50 value of 10 ÎŒM. The activity was even greater against amastigotes in comparison with promastigotes with an IC50 value of 0.5 ÎŒM. The selectivity index in relation to the intracellular form demonstrated that the antiparasitic activity was approximately 56 times higher than its toxicity to mammalian cells. Investigation of the in vivo AMD activity in the L. infantum-infected hamster model showed that 51 days after the initial infection, amiodarone was unable to reduce the parasite burden in the spleen and liver when treated for 10 consecutive days, intraperitoneally, at 50 mg/kg/day, as determined by qPCR. Although not statistically significant, AMD was able to reduce the parasite burden by 20% in the liver when treated for 10 consecutive days, orally, at 100 mg/kg/day; no reduction in the spleen was found by qPCR. Conclusions: Our findings may help further drug design studies seeking new AMD derivatives that may provide new candidates with an in vitro selectivity close to or even greater than that observed in the prototype delivering effectiveness in the experimental model of VL.</p></div
Bright Field Microscopy as an Alternative to Whole Cell Fluorescence in Automated Analysis of Macrophage Images
Fluorescence microscopy is the standard tool for detection and analysis of cellular phenomena. This technique, however, has a number of drawbacks such as the limited number of available fluorescent channels in microscopes, overlapping excitation and emission spectra of the stains, and phototoxicity.We here present and validate a method to automatically detect cell population outlines directly from bright field images. By imaging samples with several focus levels forming a bright field -stack, and by measuring the intensity variations of this stack over the -dimension, we construct a new two dimensional projection image of increased contrast. With additional information for locations of each cell, such as stained nuclei, this bright field projection image can be used instead of whole cell fluorescence to locate borders of individual cells, separating touching cells, and enabling single cell analysis. Using the popular CellProfiler freeware cell image analysis software mainly targeted for fluorescence microscopy, we validate our method by automatically segmenting low contrast and rather complex shaped murine macrophage cells.The proposed approach frees up a fluorescence channel, which can be used for subcellular studies. It also facilitates cell shape measurement in experiments where whole cell fluorescent staining is either not available, or is dependent on a particular experimental condition. We show that whole cell area detection results using our projected bright field images match closely to the standard approach where cell areas are localized using fluorescence, and conclude that the high contrast bright field projection image can directly replace one fluorescent channel in whole cell quantification. A RAC-GEF network critical for early intestinal tumourigenesis.
RAC1 activity is critical for intestinal homeostasis, and is required for hyperproliferation driven by loss of the tumour suppressor gene Apc in the murine intestine. To avoid the impact of direct targeting upon homeostasis, we reasoned that indirect targeting of RAC1 via RAC-GEFs might be effective. Transcriptional profiling of Apc deficient intestinal tissue identified Vav3 and Tiam1 as key targets. Deletion of these indicated that while TIAM1 deficiency could suppress Apc-driven hyperproliferation, it had no impact upon tumourigenesis, while VAV3 deficiency had no effect. Intriguingly, deletion of either gene resulted in upregulation of Vav2, with subsequent targeting of all three (Vav2-/- Vav3-/- Tiam1-/-), profoundly suppressing hyperproliferation, tumourigenesis and RAC1 activity, without impacting normal homeostasis. Critically, the observed RAC-GEF dependency was negated by oncogenic KRAS mutation. Together, these data demonstrate that while targeting RAC-GEF molecules may have therapeutic impact at early stages, this benefit may be lost in late stage disease
