    HIV-1 Vpr Causes Synaptodendritic Damage in Neurons

    HIV weakens the immune system by infecting and destroying T-cells, leaving the body vulnerable to infection and the development of AIDS. Conventional treatments for HIV, such as combined anti-rectroviral therapy (cART), fail to prevent the development of HIV-associated neurocognitive disorder (HAND). Neurological dysfunction has been directly related to the invasion of HIV in the central nervous system (CNS). HIV produces neurotoxic proteins, such as the Viral Protein R (Vpr), which contribute to HAND. Astrocytes are the most abundant cells in the brain and an important HIV target. We hypothesize that astrocytes expressing Vpr will cause neuronal damage in our co-culture system. Primary astrocytes were transfected with Vpr plasmid or control (pEGFP or mock) using electroporation. Astrocytes were then co-cultured with cortical neurons. At 48 and 72 hours we collected the primary astrocytes to confirm the Vpr expression via western blot analysis. We then measured structural damage in the neurons using immunofluorescence for cytoskeletal (MAP2, f-actin) and synaptic (synaptophysin) damage. Preliminary results showed strong staining of filamentous actin and MAP2 with weak detection of synaptophysin. The positive control for neurotoxicity (2.8µM acrylamide) showed substantial damage to the cellular structure. Results for Vpr expression are pending. After confirming that the immunofluorescence assays are working with our controls, we expect to detect any synaptodendritic damage in the neurons caused by Vpr in our upcoming experiments

    Coupled-channels effects in elastic scattering and near-barrier fusion induced by weakly bound nuclei and exotic halo nuclei

    The influence on fusion of coupling to the breakup process is investigated for reactions where at least one of the colliding nuclei has a sufficiently low binding energy for breakup to become an important process. Elastic scattering, excitation functions for sub-and near-barrier fusion cross sections, and breakup yields are analyzed for 6,7^{6,7}Li+59^{59}Co. Continuum-Discretized Coupled-Channels (CDCC) calculations describe well the data at and above the barrier. Elastic scattering with 6^{6}Li (as compared to 7^{7}Li) indicates the significant role of breakup for weakly bound projectiles. A study of 4,6^{4,6}He induced fusion reactions with a three-body CDCC method for the 6^6He halo nucleus is presented. The relative importance of breakup and bound-state structure effects on total fusion is discussed.Comment: 29 pages, 9 figure

    Intestinal colonization due to Escherichia coli ST131: Risk factors and prevalence

    Background Escherichia coli sequence type 131 (ST131) is a successful clonal group that has dramatically spread during the last decades and is considered an important driver for the rapid increase of quinolone resistance in E. coli. Methods Risk factors for rectal colonization by ST131 Escherichia coli (irrespective of ESBL production) were investigated in 64 household members (18 were colonized) and 54 hospital contacts (HC; 10 colonized) of 34 and 30 index patients with community and nosocomial infection due to these organisms, respectively, using multilevel analysis with a p limit of < 0.1. Result Colonization among household members was associated with the use of proton-pump inhibitors (PPI) by the household member (OR = 3.08; 95% CI: 0.88–10.8) and higher age of index patients (OR = 1.05; 95% CI; 1.01–1.10), and among HC, with being bed-ridden (OR = 21.1; 95% CI: 3.61–160.0) and having a urinary catheter (OR = 8.4; 95% CI: 0.87–76.9). Conclusion Use of PPI and variables associated with higher need of person-to-person contact are associated with increased risk of rectal colonization by ST131.     Descendants of the Jurassic turiasaurs from Iberia found refuge in the Early Cretaceous of western USA

    A new, largely complete eusauropod dinosaur with cranial and postcranial elements from two skeletons, Mierasaurus bobyoungi gen. nov., sp. nov. from the lower Yellow Cat Member (Early Cretaceous) of Utah (USA), is the first recognized member of Turiasauria from North America. Moreover, according to our phylogenetic results, Moabosaurus utahensis from the lower Yellow Cat Member of Utah (USA) is also a member of this clade. This group of non-neosauropod eusauropods, which now includes five genera (Losillasaurus, Turiasaurus, Mierasaurus, Moabosaurus and Zby), was previously known only from the Jurassic of Europe. These recent discoveries in Utah suggest that turiasaurs as a lineage survived the Jurassic-Cretaceous extinction boundary and expanded their known range, at least, into western North America. The revised spatiotemporal distribution of turiasaurs is consistent with the presence of a land connection between North America and Europe sometime during the late Tithonian to Valanginian (c.147-133 Ma). Mierasaurus and Moabosaurus are the only non-neosauropod eusauropods known from North America, despite being younger than the classic neosauropods of the Morrison Formation (c.150 Ma)

    Strong Water Absorption in the Dayside Emission Spectrum of the Planet HD 189733b

    Recent observations of the extrasolar planet HD 189733b did not reveal the presence of water in the emission spectrum of the planet. Yet models of such 'Hot Jupiter' planets predict an abundance of atmospheric water vapour. Validating and constraining these models is crucial for understanding the physics and chemistry of planetary atmospheres in extreme environments. Indications of the presence of water in the atmosphere of HD 189733b have recently been found in transmission spectra, where the planet's atmosphere selectively absorbs the light of the parent star, and in broadband photometry. Here we report on the detection of strong water absorption in a high signal-to-noise, mid-infrared emission spectrum of the planet itself. We find both a strong downturn in the flux ratio below 10 microns and discrete spectral features that are characteristic of strong absorption by water vapour. The differences between these and previous observations are significant and admit the possibility that predicted planetary-scale dynamical weather structures might alter the emission spectrum over time. Models that match the observed spectrum and the broadband photometry suggest that heat distribution from the dayside to the night side is weak. Reconciling this with the high night side temperature will require a better understanding of atmospheric circulation or possible additional energy sources.Comment: 11 pages, 1 figure, published in Natur

    Species Composition, Distribution, Biomass Trends and Exploitation of Dominant Fish Species in Manila Bay using Experimental Trawl Survey

    An experimental trawl fishing survey was conducted in Manila Bay from March 2014 to October 2015 at sixteen (16) pre-established dragging stations adapted from an earlier study (MADECOR, 1995). Using a commercial otter trawl, the average trawling speed during fishing operations was 6-7 km/hour. Analysis of catches focused on biomass trends, species composition, distribution and exploitation of dominant species. A total of 146 fish and invertebrate species belonging to 48 families were recorded during the survey period wherein most of the catches were dominated by small pelagic species such as anchovies and sardines. Exploitation rates (E) for the six (6) dominant species (Sardinella gibbosa, Sardinella fimbriata, Valamugil seheli, Mugil cephalus, Encrasicholina devisi and Stolephorous commersonnii) shows signs of overfishing. The estimated demersal fish biomass of the bay revealed that the relative decline was about 90% from the 1947 baseline study
