Association between the c.*229C>T polymorphism of the topoisomerase IIb binding protein 1 (TopBP1) gene and breast cancer
Topoisomerase IIb binding protein 1 (TopBP1)
is involved in cell survival, DNA replication, DNA damage
repair and cell cycle checkpoint control. The biological
function of TopBP1 and its close relation with BRCA1
prompted us to investigate whether alterations in the
TopBP1 gene can influence the risk of breast cancer.
The aim of this study was to examine the association
between five polymorphisms (rs185903567, rs116645643,
rs115160714, rs116195487, and rs112843513) located in
the 30UTR region of the TopBP1 gene and breast cancer
risk as well as allele-specific gene expression. Five hundred
thirty-four breast cancer patients and 556 population controls
were genotyped for these SNPs. Allele-specific Top-
BP1 mRNA and protein expressions were determined by
using real time PCR and western blotting methods,
respectively. Only one SNP (rs115160714) showed an
association with breast cancer. Compared to homozygous
common allele carriers, heterozygous and homozygous for
the T variant had significantly increased risk of breast
cancer (adjusted odds ratio = 3.81, 95 % confidence
interval: 1.63–8.34, p = 0.001). Mean TopBP1 mRNA and
protein expression were higher in the individuals with the
CT or TT genotype. There was a significant association
between the rs115160714 and tumor grade and stage. Most
carriers of minor allele had a high grade (G3) tumors
classified as T2-T4N1M0. Our study raises a possibility
that a genetic variation of TopBP1 may be implicated in
the etiology of breast cancer
Galectin-3 in Cardiac Remodeling and Heart Failure
Galectin-3 is a member of the galectin family, which consists of animal lectins that bind β-galactosides. Recently, a role for galectin-3 in the pathophysiology of heart failure has been suggested. It was observed that galectin-3 is specifically upregulated in decompensated heart failure compared with compensated heart failure in animal models of heart failure. This has been associated with activation of fibroblasts and macrophages, which are a hallmark of cardiac remodeling. Therefore, galectin-3 may be a culprit biomarker in heart failure. Initial clinical observations indicate that galectin-3 may be a useful biomarker for decompensated heart failure, with incremental value over well-used “pressure-dependent” biomarkers, such as B-type natriuretic peptide. Future studies should focus on galectin-3 biology to better address the usefulness of galectin-3 as a biomarker and probe the usefulness of anti-galectin-3 therapy in treating heart failure
Gene and protein expression of O-GlcNAc-cycling enzymes in human laryngeal cancer
Aberrant protein O-GlcNAcylation may contribute to the development and malignant behavior of many cancers. This modification is controlled by O-linked β-N-acetylglucosamine transferase (OGT) and O-GlcNAcase (OGA). The aim of this study was to determine the expression of O-GlcNAc cycling enzymes mRNA/protein and to investigate their relationship with clinicopathological parameters in laryngeal cancer. The mRNA levels of OGT and MGEA5 genes were determined in 106 squamous cell laryngeal cancer (SCLC) cases and 73 non-cancerous adjacent laryngeal mucosa (NCLM) controls using quantitative real-time PCR. The level of OGT and OGA proteins was analyzed by Western blot. A positive expression of OGT and MGEA5 transcripts and OGT and OGA proteins was confirmed in 75.5 and 68.9 % and in 43.7 and 59.4 % samples of SCLC, respectively. Higher levels of mRNA/protein for both OGT and OGA as well as significant increases of 60 % in total protein O-GlcNAcylation levels were noted in SCLC compared with NCLM (p < 0.05). As a result, an increased level of OGT and MGEA5 mRNA was related to larger tumor size, nodal metastases, higher grade and tumor behavior according to TFG scale, as well as incidence of disease recurrence (p < 0.05). An inverse association between OGT and MGEA5 transcripts was determined with regard to prognosis (p < 0.05). In addition, the highest OGT and OGA protein levels were observed in poorly differentiated tumors (p < 0.05). No correlations with other parameters were noted, but the results showed a trend of more advanced tumors to be more frequently OGT and OGA positive. The results suggest that increased O-GlcNAcylation may have an effect on tumor aggressiveness and prognosis in laryngeal cancer.This work was supported, in part, by a grant

REACH Regulation in the context of ethanol registration and other relevant products in fermentation processes of spirituous industry
Alkohol etylowy (etanol), nr CAS1: 64-17-5, nr WE2: 200-578-6, jest substancją chemiczną znajdującą zastosowanie przede wszystkim w przemyśle spożywczym - do produkcji napojów alkoholowych w branży spirytusowej, a także jako dodatek do wyrobu niektórych słodyczy, ciast, tortów oraz zapachów spożywczych. Taki kierunek wykorzystania etanolu dominuje w Polsce. Nie należy jednak zapominać, że etanol stanowi jednocześnie jeden z najważniejszych podstawowych rozpuszczalników, zarówno substancji organicznych jak i nieorganicznych, stosowanych w przemyśle chemicznym, kosmetycznym i farmaceutycznym, w tym ostatnim jako surowiec do produkcji wielu leków. Jego dodatek do benzyny znajduje także zastosowanie jako paliwo do silników. Podstawowy problem prawny, który bezpośrednio lub pośredni dotyka branży spirytusowej w ostatnich latach polega na tym, że etanol podobnie jak inne substancje chemiczne, podlega obowiązkom rejestracyjnym wynikającym z postanowień rozporządzenia REACHEthyl alcohol (ethanol), CAS No1: 64-17-5, EC No2: 200-578-6, is a chemical substance applied first of all in the food industry for the vodka and liquors manufacturing in the spirituous branch, and also as an additive for making some selected sweetmeats, cakes, layer cakes and food fragrances. This way of use prevails in Poland. It should be not forgotten that ethanol belongs simultaneously to the most important basic solvents for both organic and inorganic substances used in the chemical, cosmetic and pharmaceutical industries, in this latter as a raw material for production of numerical medicaments. As an additive to the gasoline is used as an engine fuel. Basic legal problem which directly or indirectly concerns spirituous branch in the last years relies on fact that ethanol similarly like other chemical substances is subject of registration duty resulted from the provisions of REACH regulation
Spatial evolutionary games and radiation induced bystander effect
We present an application of evolutionary game theory to modeling of some processes important from oncological point of view. A studied phenomenon is a radiation induced bystander effect, in which three different strategies (phenotypes) of cells take part. The proposed payoff table of fitness, related to environment adaptation and genetic cell behavior, contains costs/profits of bystander effect, choice of apoptotic pathway, producing growth factors and resistance against bystander effect. We consider a game theory model including spatial cells allocation (the game is played on lattice). We discuss also different polymorphic equilibrium points dependent on model parameters, types of spatial games and players distribution
REACH Regulation vs. waste – where are the problems embedded in?
W publikacji omówiono główne problemy dotyczące braku
spójności legislacji odpadowej z wymaganiami rozporządzenia REACH.
Wymieniono bardziej istotne zapisy odnoszące się do odpadów
w rozporządzeniu REACH oraz legislacji odpadowej. Następnie
dokonano analizy pod kątem istniejących konsekwencji zapisów
prawa, ze szczególnym uwzględnieniem pojęcia „produktu ubocznego”
oraz „odpadu, który utracił status odpadu” oraz sytuacji energetyki
zawodowej, dotkniętej jako pierwsza branża problemem braku
spójności w istniejących zapisach. Na koniec przedyskutowano
kwestię braku pragmatycznego podejścia do zapisów prawnych
na przykładzie importu pasty ołowiowej, która miała służyć jako
uzupełniający surowiec w produkcji akumulatorów. Artykuł zakończono
wnioskami oraz podsumowaniem w postaci cytatów pochodzących
od respondentów Punktu Konsultacyjnego ds. REACH
i CLP Ministerstwa Gospodarki.The problems concerning lack of the consistency between
waste legislation and REACH regulation requirements have been
discussed. The most important provisions referring to REACH and
waste legislation have been mentioned. Then the assessment of consequences
of legal provisions with particular attention to the term
„by-product” and „waste ceased to be waste” has been made. Also
situation of power industry affected as the first sector by inconsistency
in existing legal provisions has been analyzed. Finally question
of lack of pragmatic approach to the legal provisions has been raised
on the example of import of leaded paste to be used as supplementary
feedstock for the production of car batteries. Article includes
conclusions in the end and summing up in the shape of some quotes
originating from respondents of the REACH & CLP Consulting Point
of the Polish Ministry of Economy
Exposure scenarios : a descriptor system for describing applications

W publikacji omówiono trzy zasadnicze etapy procesu oceny
bezpieczeństwa chemicznego: ocenę zagrożeń, ocenę narażenia
oraz charakterystykę ryzyka. W ramach oceny zagrożeń podano
i omówiono m.in. poziomy progowe narażenia uznawane za bezpieczne.
Przy dyskusji dotyczącej oceny narażenia omówiono rolę
scenariuszy narażenia oraz sposób dochodzenia do rzeczywistych
poziomów narażenia człowieka oraz przewidywanego stężenia
w środowisku jako wartości odniesienia do poziomów progowych
nie powodujących szkodliwych skutków. Następnie przedyskutowano
sposób przeprowadzania charakterystyki ryzyka oraz cykliczne
dochodzenie do akceptowalnego poziomu ryzyka, a co za tym
idzie, do opracowania ostatecznego scenariusza narażenia, który
ma służyć dalszym użytkownikom w łańcuchu dostaw do stworzenia
warunków bezpiecznego postępowania z substancją.

wymagań w zakresie informacji i oceny bezpieczeństwa chemicznego”
oraz „Poradnika w pigułce – Ocena bezpieczeństwa chemicznego”.Three basic stages of the chemical safety assessment process
have been discussed in the publication: hazard assessment, exposure
assessment and risk characterization. In the framework of hazard
assessment among others the threshold levels for exposure considered
as safe have been mentioned and discussed. By discussion concerning
exposure assessment, the role of the exposure scenarios and the
way of approach to the real human exposure levels and predicted
environmental concentration as the reference values to the no harmful
effect threshold levels have been described. Then the way of risk
characterization and cyclic achievement of acceptable risk level have
been talked over and, by extension elaboration of the final exposure
scenario which may be applied for downstream users in the supply
chain to create safe substance handling conditions – has been discussed.

Legal obligation to notify to the European Chemicals Agency (ECHA) an information on classification and labelling
W artykule przytoczono w większości w oryginalnym brzmieniu odpowiednie fragmenty wzajemnie się przenikających przepisów: o rozporządzenia Parlamentu Europejskiego i Rady (WE) nr 1272/2008 (CLP) z dn. 16 grudnia 2008 r. w sprawie klasyfikacji, oznakowania i pakowania substancji i mieszanin o rozporządzenia Parlamentu Europejskiego i Rady (WE) nr 1907/2006 (REACH) z dn. 18 grudnia 2006 r., odnoszące się do obowiązku zgłoszenia klasyfikacji i oznakowania do Europejskiej Agencji Chemikaliów (ECHA). Obowiązku tego należy dokonać przed 1 grudnia 2010 r. dla wszystkich substancji wprowadzonych do obrotu przed tą datą - stąd tak istotne jest uświadomienie tego faktu przedsiębiorstwom, aby go nie przeoczyły. Następnie zwrócono uwagę na możliwość dokonania powiadomienia w przypadku importerów substancji przez jeden podmiot w imieniu całej grupy, zwłaszcza w tym przypadku, kiedy eksporter substancji do UE nie jest zainteresowany w ujawnianiu pełnej informacji na temat jej składu ze względu na tajemnicę handlową. Zwrócono także uwagę na fakt, że wymieniony obowiązek w niektórych przypadkach może dotyczyć takich substancji, jak odpady (o ile są wykorzystywane gospodarczo będąc przekazywanymi do zagospodarowania stronom trzecim) oraz wybrane polimery. Na zakończenie stwierdzono, że na podstawie przedłożonych informacji Agencja utworzy wykaz klasyfikacji i oznakowania, który będzie udostępniony publicznie na stronach internetowych ECHA.In the article original fragments of relevant provisions interpenetrating each other have been quoted from the following regulations: o Regulation (EC) No 1272/2008 of the European Parliament and of the Council of 16 December 2008 on classification, labelling and packaging of substances and mixtures (CLP) o Regulation (EC) No 1907/2006 of the European Parliament and of the Council of 18 December 2006 (REACH), referring to the legal obligation to notify to the European Chemicals Agency (ECHA) an information on classification and labelling. This obligation should be made before 1 December 2010 for all substances placed on the market before this date - that's why awareness of this fact is so essential for the companies to avoid this deadline missing by them. Then in the case of more importers the possibility of notification by one enterprise in the name of the whole group has been mentioned, particularly in case of substance exporter to the EC, not interested in disclosure full information about substance composition for the sake of business confidentiality. Attention has been paid to fact that aforementioned legal obligation may refer to such substances like waste (if they are economically utilized being transferred to other entrepreneurs for further disposal) and selected polymers. Finally, it has been stated that on the basis of received information the Agency will establish and maintain a classification and labelling inventory in the form of a database available on the ECHA website
