Genetic and Metabolic Controls for Sulfate Metabolism in \u3cem\u3eNeurospora crassa\u3c/em\u3e: Isolation and Study of Chromate-Resistant and Sulfate Transport-Negative Mutants

Mutants of Neurospora resistant to chromate were selected and all were found to map at a single genetic locus designated as cys-13. The chromate-resistant mutants grow at a wild-type rate on minimal media but are partially deficient in the transport of inorganic sulfate, especially during the conidial stage. An unlinked mutant, cys-14, is sensitive to chromate but transports sulfate during the mycelial stage at only 25% of the wild-type rate; cys-14 also grows at a fully wild-type rate on minimal media. The double-mutant strain, cys-13;cys-14, cannot utilize inorganic sulfate for growth and completely lacks the capacity to transport this anion. The only biochemical lesion that has been detected for the double-mutant strain is its loss in capacity for sulfate transport. Neurospora appears to possess two distinct sulfate permease species encoded by separate genetic loci. The transport system (permease I) encoded by cys-13 predominates in the conidial stage and is replaced by sulfate permease II, encoded by the cys-14 locus, during outgrowth into the mycelial phase. Narrative Generation in Entertainment: Using Artificial Intelligence Planning

From the field of artificial intelligence (AI) there is a growing stream of technology capable of being embedded in software that will reshape the way we interact with our environment in our everyday lives. This ‘AI software’ is often used to tackle more mundane tasks that are otherwise dangerous or meticulous for a human to accomplish. One particular area, explored in this paper, is for AI software to assist in supporting the enjoyable aspects of the lives of humans. Entertainment is one of these aspects, and often includes storytelling in some form no matter what the type of media, including television, films, video games, etc. This paper aims to explore the ability of AI software to automate the story-creation and story-telling process. This is part of the field of Automatic Narrative Generator (ANG), which aims to produce intuitive interfaces to support people (without any previous programming experience) to use tools to generate stories, based on their ideas of the kind of characters, intentions, events and spaces they want to be in the story. The paper includes details of such AI software created by the author that can be downloaded and used by the reader for this purpose. Applications of this kind of technology include the automatic generation of story lines for ‘soap operas’

New Frontiers in the Relationship Between National and European Courts

The central question pervading this discussion is simply stated: whether and to what extent the European Court of Justice (“Court of Justice” or “Court”) (and the European institutions more generally) can count on the courts of the Member States to perform their judicial tasks in ways that are faithful to the ground rules of European Union (“EU”) law, be those rules substantive or procedural in character. Looking back over the past five decades, I am struck by the succession of different forms this question of national court “fidelity” to Community law, for lack of a better term, has taken. (The quoted term is meant to evoke precisely the general duty of loyal cooperation *526 imposed on Member States under Article 10 of the current EC Treaty.) I would suggest that we have witnessed essentially three generations of such “fidelity challenges.” In the early years, the fidelity challenge--or “infidelity risk,” if you prefer--ran something like this: Will national courts accept and conform to the Grundnorms of European constitutional law, that is to say, the principles of supremacy and direct effect expounded by the Court of Justice? Or, to take a second example, will national courts genuinely make preliminary references to the Court of Justice in accordance with the criteria for the making of preliminary references that have been established in the Treaty and in the case law of the Court? Considering the indispensable role that preliminary references and preliminary rulings play in the development of EU law, the importance of these ground rules should not be underestimated
