    Optimal Imprisonment with General Enforcement of Law

    We study the optimal use of imprisonment when enforcement efforts are general (i.e. when the probability of detection is common for a range of acts). In contrast to the conventional wisdom that optimal imprisonment rises with the act's harmfulness and is equal to the maximum level only for the most harmful acts, we show that - when the distribution of criminal benefits exhibits a standard monotone hazard rate - optimal imprisonment can only be zero or maximal. Thus, having general as opposed to specific enforcement effort does not alter the fact that only extreme sanction levels should optimally be employed

    Prognosis: Closed-box analysis of network protocol implementations

    We present Prognosis, a framework offering automated closed-box learning and analysis of models of network protocol implementations. Prognosis can learn models that vary in abstraction level from simple deterministic automata to models containing data operations, such as register updates, and can be used to unlock a variety of analysis techniques - model checking temporal properties, computing differences between models of two implementations of the same protocol, or improving testing via model-based test generation. Prognosis is modular and easily adaptable to different protocols (e.g. TCP and QUIC) and their implementations. We use Prognosis to learn models of (parts of) three QUIC implementations - Quiche (Cloudflare), Google QUIC, and Facebook mvfst - and use these models to analyse the differences between the various implementations. Our analysis provides insights into different design choices and uncovers potential bugs. Concretely, we have found critical bugs in multiple QUIC implementations, which have been acknowledged by the developers

    Effects of Mindfulness Training on School Teachers\u2019 Self-Reported Personality Traits As Well As Stress and Burnout Levels

    Among a sample of only female school teachers, we compared a mindfulness meditation (MM) training group (n = 19) with a waiting-list control group (n = 20) on several participant-completed questionnaires: the Five Facet Mindfulness Questionnaire, the Big Five Personality Inventory, the Teacher Stress Inventory, and the Maslach Burnout Inventory. With these measures, we assessed the participants\u2019 dispositional mindfulness, personality styles, and their stress and burnout. Following mindfulness training, teachers in the MM group showed higher trait mindfulness and conscientiousness and lower neuroticism and stress and burnout levels than teachers in the waiting-list control group. These results support the beneficial role of MM in individuals\u2019 effective management of stressful conditions in the workplace

    Fragile X mental retardation protein (FMRP) and metabotropic glutamate receptor subtype 5 (mGlu5) control stress granule formation in astrocytes

    Fragile X syndrome (FXS) is a common form of intellectual disability and autism caused by the lack of Fragile X Mental Retardation Protein (FMRP), an RNA-binding protein involved in RNA transport and protein synthesis. Upon cellular stress, global protein synthesis is blocked and mRNAs are recruited into stress granules (SGs), together with RNA-binding proteins including FMRP. Activation of group-I metabotropic glutamate (mGlu) receptors stimulates FMRP-mediated mRNA transport and protein synthesis, but their role in SGs formation is unexplored. To this aim, we pre-treated wild type (WT) and Fmr1 knockout (KO) cultured astrocytes with the group-I-mGlu receptor agonist (S)-3,5-Dihydroxyphenylglycine (DHPG) and exposed them to sodium arsenite (NaAsO2), a widely used inducer of SGs formation. In WT cultures the activation of group-I mGlu receptors reduced SGs formation and recruitment of FMRP into SGs, and also attenuated phosphorylation of eIF2α, a key event crucially involved in SGs formation and inhibition of protein synthesis. In contrast, Fmr1 KO astrocytes, which exhibited a lower number of SGs than WT astrocytes, did not respond to agonist stimulation. Interestingly, the mGlu5 receptor negative allosteric modulator (NAM) 2-methyl-6-(phenylethynyl)pyridine (MPEP) antagonized DHPG-mediated SGs reduction in WT and reversed SGs formation in Fmr1 KO cultures. Our findings reveal a novel function of mGlu5 receptor as modulator of SGs formation and open new perspectives for understanding cellular response to stress in FXS pathophysiology

    Application of single molecule technology to rapidly map long DNA and study the conformation of stretched DNA

    Herein we describe the first application of direct linear analysis (DLA) to the mapping of a bacterial artificial chromosome (BAC), specifically the 185.1 kb-long BAC 12M9. DLA is a single molecule mapping technology, based on microfluidic elongation and interrogation of individual DNA molecules, sequence-specifically tagged with bisPNAs. A DNA map with S/N ratio sufficiently high to detect all major binding sites was obtained using only 200 molecule traces. A new method was developed to extract an oriented map from an averaged map that included a mixture of head-first and tail-first DNA traces. In addition, we applied DLA to study the conformation and tagging of highly stretched DNA. Optimal conditions for promoting sequence-specific binding of bisPNA to an 8 bp target site were elucidated using DLA, which proved superior to electromobility shift assays. DLA was highly reproducible with a hybridized tag position localized with an accuracy of ±0.7 µm or ±2.1 kb demonstrating its utility for rapid mapping of large DNA at the single molecule level. Within this accuracy, DNA molecules, stretched to at least 85% of their contour length, were stretched uniformly, so that the map expressed in relative coordinates, was the same regardless of the molecule extension

    Does the mind map learning strategy facilitate information retrieval and critical thinking in medical students?

    <p>Abstract</p> <p>Background</p> <p>A learning strategy underutilized in medical education is mind mapping. Mind maps are multi-sensory tools that may help medical students organize, integrate, and retain information. Recent work suggests that using mind mapping as a note-taking strategy facilitates critical thinking. The purpose of this study was to investigate whether a relationship existed between mind mapping and critical thinking, as measured by the Health Sciences Reasoning Test (HSRT), and whether a relationship existed between mind mapping and recall of domain-based information.</p> <p>Methods</p> <p>In this quasi-experimental study, 131 first-year medical students were randomly assigned to a standard note-taking (SNT) group or mind map (MM) group during orientation. Subjects were given a demographic survey and pre-HSRT. They were then given an unfamiliar text passage, a pre-quiz based upon the passage, and a 30-minute break, during which time subjects in the MM group were given a presentation on mind mapping. After the break, subjects were given the same passage and wrote notes based on their group (SNT or MM) assignment. A post-quiz based upon the passage was administered, followed by a post-HSRT. Differences in mean pre- and post-quiz scores between groups were analyzed using independent samples <it>t</it>-tests, whereas differences in mean pre- and post-HSRT total scores and subscores between groups were analyzed using ANOVA. Mind map depth was assessed using the Mind Map Assessment Rubric (MMAR).</p> <p>Results</p> <p>There were no significant differences in mean scores on both the pre- and post-quizzes between note-taking groups. And, no significant differences were found between pre- and post-HSRT mean total scores and subscores.</p> <p>Conclusions</p> <p>Although mind mapping was not found to increase short-term recall of domain-based information or critical thinking compared to SNT, a brief introduction to mind mapping allowed novice MM subjects to perform similarly to SNT subjects. This demonstrates that medical students using mind maps can successfully retrieve information in the short term, and does not put them at a disadvantage compared to SNT students. Future studies should explore longitudinal effects of mind-map proficiency training on both short- and long-term information retrieval and critical thinking.</p

    Simultaneous Detection of FISH Signals and Bromo-Deoxyuridine Incorporation in Fixed Tissue Cultured Cells

    FISH (Fluorescence in situ hybridization) is a powerful technique that detects and localises specific DNA sequences on metaphase chromosomes, interphase nuclei or chromatin fibres. When coupled to BrdU (5-Bromo 2-deoxy-uridine) labeling of newly replicated DNA, the replication properties of different DNA sequences can be analysed. However, the technique for the detection of BrdU incorporation is time consuming, and relies on acidic pH buffer treatments, that prevent use of pH sensitive fluorochromes such as FITC (Fluoro-isothiocianate) during FISH. In this work, we describe a simplified protocol that allows the simultaneous detection of FISH signals and BrdU incorporation. Since the technique does not involve paraformaldehyde for cell fixation, or formamide for denaturation of the target DNA and in post-hybridisation washes, it represents a safer alternative to classical FISH techniques

    A New Approach to Spin and Statistics

    We give an algebraic proof of the spin-statistics connection for the parabosonic and parafermionic quantum topological charges of a theory of local observables with a modular PCT-symmetry. The argument avoids the use of the spinor calculus and also works in 1+2 dimensions.
