    Identification, Structural Characterization and Molecular Simulation of ACE Inhibitory Peptides from the Hydrothermal Vent Mussel

    The objective of this study was to prepare an angiotensin I-converting enzyme (ACE)-inhibitory peptide from the hydrothermal vent mussel, Gigantidas vrijenhoeki. The G.vrijenhoeki protein was hydrolyzed by various hydrolytic enzymes. The peptic hydrolysate exhibited the highest ACE-inhibitory activity and was fractionated into four molecular weight ranges by ultrafiltration. The <1 kDa fraction exhibited the highest ACE inhibitory activity and was found to have 11 peptide sequences. Among the analyzed peptides, KLLWNGKM exhibited stronger ACE inhibitory activity and an IC50 value of 0.007 μM. To investigate the ACE-inhibitory activity of the analyzed peptides, a molecular docking study was performed. KLLWNGKM exhibited the highest binding energy (-1,317.01 kcal/mol), which was mainly attributed to the formation of hydrogen bonds with the ACE active pockets, zinc-binding motif, and zinc ion. These results indicate thatG. vrijenhoeki-derived peptides can serve as nutritional and pharmacological candidates for controlling blood pressure.2

    Sargassum species are representative brown algae found on the coasts of the world, with 27 species native to the coast of Jeju Island, South Korea. Fucoidan was derived from brown algae and was evaluated variety of biological activities. Analysis of monosaccharide composition is inevitable for in-depth study of fucoidan. However, the quantitative analysis method of fucoidan with complex structures according to species and habitat is insufficient. In this study, the validated HPLC method using the PMP derivatization has a short running time and can be economically repeated using UV detection. In addition, the quantification was verified by comparing it with the quantitative results of commercial fucoidan obtained using HPAEC-PAD. The analysis method was verified by establishing system suitability, precision, accuracy, linearity, LOD, and LOQ, and the results were satisfactory. Furthermore, we evaluated the antioxidant effect of the polysaccharide extract of Sargassum species in Jeju Island. The SCpol and SApol showed excellent activity and, as a result of zebrafish experiment, it was shown that all species-derived extracts had antioxidant potential. These results were presented as a way to indicate to regional economic revitalization by increasing the value of industrial applications and utilizing its resources.2

    Reversed Approach through Lymphocele/ Lymphatic Fluid Collection for Glue Embolization of Injured Lymphatic Vessels

    A total of 9 glue embolization procedures of injured lymphatic vessels with a reversed approach from the lymphocele/lymphatic fluid collection in 8 patients were retrospectively reviewed. The approach routes were via the indwelling chest tube for pleural effusion (n ¼ 2), the drainage catheter tract for abdominopelvic lymphocele/lymphatic fluid collection/thigh lymphocele (n ¼ 4), and the direct puncture of the lymphatic fluid collection at the operation bed (n ¼ 2). All the procedures were technically successful without complications. The mean daily leakage rate decreased from 465 mL/d before the procedure to 42 mL/d after the procedure, and the drainage catheters could be removed after 8 procedures, achieving a clinical success rate of 88.9% (8 of 9 procedures)

    Brown algae are known to contain rich anti-inflammatory compounds. The objective of this study was to identify anti-inflammatory compound in the Sargassum macrocarpum and anti-inflammatory effect of tuberatolide B (TTB) was investigated. TTB significantly suppressed the production of NO and pro-inflammatory cytokines such as interleukin (IL)-6 and IL-1β. In addition, the mechanism involved in the inhibition of the inflammatory effect of LPS by TTB was investigated. The results showed that TTB aforementioned this effect through NF-κB and MAPKs phosphorylation pathways. We also assessed the anti-inflammatory activity of TTB using in vivo zebrafish model. In LPS-stimulated zebrafish, TTB enhanced survival and significantly inhibited the NO production and mRNA expression of inducible NO synthase. Therefore, anti-inflammatory activity of TTB against LPS in RAW264.7 cells and the zebrafish model was determined. These finding suggest that TTB may be used as functional anti-inflammatory foods and nutraceuticals2

    Skin-moisturizing Effects of Abalone By-products Viscera Extract in HDF and HaCaT Cells by Regulating MAPKS and PI3K/AKT/NF-kB Signaling Pathways

    Abalone, a nutritional value marine edible gastropod, is a popular seafood delicacy worldwide, especially in Asia. However, viscera by-products are generally discarded during processing. Therefore, we investigated the skin-moisturizing effects of abalone viscera ultrasonic extract (AVU) in human dermal fibroblasts (HDF) and human keratinocyte (HaCaT) cells. AVU did not detect microorganisms. It also showed valuable protein contents, indicating that it is a worthy and safe material for industrial application. AVU increased collagen synthesis production and messenger RNA (mRNA) expression of Collagen Type I Alpha 1, 2, and 3 Chains through the transforming growth factor beta/ Suppressor of mother against decapentaplegic pathway in HDF cells. AVU also increased hyaluronic acid production, upregulated Hyaluronan Synthases 1, 2, and 3, filaggrin and aquaporin3 mRNA levels, and downregulated hyaluronidase mRNA levels in HaCaTcells. Furthermore, mechanistic studies showed that AVU increased the phosphorylation of extracellular signal-regulated kinase, p38, and cyclic AMP response-binding protein activation. AVU activated the transcription factors, phosphoinositide 3-kinase, protein kinase B, and nuclear factor kappa B cell p65 and downregulated the degranulation of inhibitory kappa B in HaCaT cells. These results demonstrated that AVU diverted from by-products has a potential moisturizing property that can be used as a functional food and cosmetics ingredient.2

    Anti-Zika Virus Potential of Chlorophyll Extracts from Tetraselmis sp., a Marine Microalga

    Recent advancements in the large-scale cultivation of Tetraselmis sp. in Korea have enabled year-round production of this marine microalgae. This study explores the potential industrial applications of Tetraselmis sp. biomass by investigating the antiviral properties of its extracts and primary components. The antiviral effects of Tetraselmis sp. extracts were evaluated in Zika virus (ZIKV)-infected cells. Following extensive isolation and purification, the main compounds were characterized using liquid chromatography–mass spectrometry (LC-MS) and nuclear magnetic resonance (NMR) analyses. Their antiviral activities were confirmed using in vitro and in silico tests. Tetraselmis sp. extracts reduced infectious viral particles and non-structural protein 1 mes-senger RNA levels in ZIKV-infected cells without inducing cytotoxicity. Additionally, they modulated the interferon-mediated immune system responses. Tetraselmis sp. extracts are composed of four main chlorophylls: chlorophyll a, chlorin e6-131-152-dimethyl-173-phytyl ester, hydroxychlorophyll a, and hydroxypheophytin a. Among them, chlorophyll a, e6-131-152-dimethyl-173-phytyl ester, and hydroxypheophytin showed the antiviral activities in ZIKV-infected cells and molecular docking simulations predicted interactions between these chlorophylls and ZIKV. Our findings suggest that Tetraselmis sp. chlorophyll extracts exert antiviral effects against ZIKV and could serve as potential therapeutic candidates against ZIKV infection.2
