[์˜๋ฌธ] In Order to Compare the growth of various cultivable Mycobacteria in cholesterol or hyaluronic acid supplemented media with that in ordinary Dubos liquid medium, some species of Mycobacteria , including human pathogens and non-pathogens(M. tuberculosis, B.C.G., M. vaccae, M. kansasii, M. scrofulaceum) were cultivated in three different media : Dubos liquid medium, KI-l medium containing 100 ใŽ/ml cholesterol and 10 % sheep serum (Kato & Ishaque, 1978), and modified LA-3 medium containing 0.1 % hyaluronic acid and 1 % bovine serum albumin (Skinsnes, 1974). The cultivation of Mycobacteria in the three different media were compared by measurement of dry weight of bacterial growth. The effect of bovine serum albumin on cholesterol utilization by Mycobacteria were also examined by modifying the constituents of KI-l medium. The results are summarized as follows : 1. All of above Mycobacteria examined in this study grew better in KI-1 medium than in others. This was especially true when bacterial growth in KI-1 medium was compared with that in Dubos medium : 4 and 3 fold increase in M. tuberculosis were observed in KI-1 medium within 4 to 6 weeks, 10 and 6 fold in B.C.G., 1.5 and 1.2 fold in M. vaccae, 2.4 and 2.1 fold in M. kansasii, and 5.2 and 3.5 fold in M. scrofulaceum. 2. The various Mycobacteria grew better in cholesterol supplemented Dubos medium than in ordinary Dubos medium: 1.5-2.1 fold increase bacterial growth in M. tuberculosis were observed in cholesterol supplemented Dubos medium compared with that in Dubos medium within 1-4 weeks, 1.5-1.8 fold in B.C.G., 1.6-1.8 fold in M. kansasii, 1.3-1.6 fold in M. vaccae, 1.3-1.5 fold in M. scrofulaceum. 3. In the requirement of the additional effects of bovine serum albumin on cholesterol supplemented medium, there was a 1.5-3 fold increases of bacterial growth in KI basal medium enriched by both cholesterol and bovine serum album in when compared with that in KI basal medium itself or enriched by cholesterol or bovine serum albumin alone. Therefore, mycobacterial growth appeared to be increased by adding cholesterol or hyaluronic acid to the medium, especially, cholesterol with bovine serum albumin.restrictio

Antitumer Aetivity of Lactobacillus casei on the Growth of Sarcoma-180YS in Mice Chang Hee Kim Department of Medical Science The Graduate School, Yonsei University (Directed by Professor Joo Deuk Kim, M.D.) Microorganisms which have been uses for clinical and experimental immunotherapy of turmers included BCG (Zbar et at.,1972 Bast et al., 1974), Corynebacterium parvum(Mills et at.,1981), Bordeteita pertussis(Purnell et at., 1975, Likhite, 1974), Toxoplasma gondii(Krahenbuhl & Remington,1974) and Listeria monocytogenes (Bast et al., 1975a,b). In addition to being potent reticuloendothelial system stimulants, many of theme agents evoke an acquired cellular resistance in the host, which to a great measure nonspecifically inhibits tumor growth. However, it is necessary to pay careful attention to the side effects in clinical applications for human cancer therapy since most of these bacteria are pathogenic. Lactobacilli, a gram-positive and non-pathogenic organism which inhabits the animal intestine, have been used for food such as yogurt all over the world from ancient times. There has been few reports on antitumor activity of lactobacilli. The present study wart undertaken to examine the antitumor activity of Lactobacillus casei(L. casei) and to elucidate the underlying mechanisms and the role of macrophages in the antitumor activity of L. casei. The results of this study are summerized as follows: 1. A group of mice received an intraperitoneal injection of sarcoma-180YS (S-l80) cells, followed by a series of 5 intraperitoneal injection of L. casei at 1 day intervals. The control group received only S-180 cells. The survival rate of the experimental group at 25 days was 67% as opposed to 33% for the control group. 2. Mice which received an intraperitoneal injection of S-180 cells admixed with 1ร—10**6 viable L. casei had a survival rate of 83% at 25 days. Mice had received S-180 admixed with 1ร—10**7 heat killed L. casei had a survival rate of 67%. The control group had survival rate of 33%. 3. S-180 inoculated subcutaneously was affected by pre-and post L. casei immunization in the following way. The inhibition rate of S-180 cell growth at 25 days after inoculation was 64% of control in the pre-immunized group and 58% of control in the post-immunized group. In additional experiment, the intralesional inoculation of L. casei in the first 15 days caused inhibition rates of 58โˆผ75%, but, as the size of the tumor increased(after 16 days, mean tumor weight:490ยฑ43mg) the inhibition rate decreased. 4. When L. casei and S-180 were inoculated subcutaneously at different sites, no inhibition of tumor growth was observed. 5. When L. casei admixed with S-180 were inoculated subcutaneously, the inhibition rates in the group given 1ร—10**6 viable bacilli were 74% and 58% of control in the group given 1ร—1O**7 heat killed bacilli. 6. The inhibitory effect of L. casei on S-180 growth was eliminated by pretreatment of the mice with carrageenan. 7. L. casei increased the delayed type hypersensitivity response to SRBC. S-180 growth was inhibited by both viable and heat killed L. casei. Macrophages appear to have an important role in this process, as suggested by inhibition of macrophage function with carrageenan.
