바실러스 세레우스균에서 유래한 MazF 단백질의 구조 및 기능 연구

These systems are known to regulate cellular growth of bacteria in response to stress and other circumstances. The importance of TA systems is largely magnified as various functions have been revealed. In this paper, I presented the crystal structure of BC0266 which is known to as a putative mRNA interferase. And I also reported it as a type II MazF toxin. The MazF toxin is activated during stressful conditions. It cleaves mRNA in a sequence-specific and ribosome-independent manner. Its activated state causes toxic results and may lead to bacterial death. In this study, I experimented structural and functional studies of Bacillus cereus and presented the first toxin structure in the TA system of B.cereus. Especially, B.cereus MazF adopts a PemK-like fold which is a subfamily of MazF. BC0266 also has an RNA recognizing loops which are called β1–β2 and β3–β4 loops. Key residues of ribonuclease activity are proposed by in vitro assay and mutational studies. It demonstrates the ribonucleic activity and the active sites which are essential for its cellular toxicity.Toxin-Antitoxin(TA) 시스템은 박테리아에 만연하며 스트레스에 반응하여 세포의 성장을 조절한다. TA system에 관련되서 다양한 기능이 알려짐에 따라 그 중요성이 점차 부각되고 있는 추세이다. 여기서 우리는 mRNA interferase의 기능을 가지는 BC0266의 크리스탈 구조를 보여주고 그것을 typeII 톡신 MazF라고 부른다. MazF 톡신은 스트레스 상황에서 ribonuclease 기능이 활성화되는데 이는 ribosome-independent하게 그리고 sequence-specific하게 작용한다. 이것의 지속적인 활동은 독성결과를 유발하고 결국 박테리아 사멸로 이어진다. 이 연구에서 바실러스 세레우스에서 유래한 MazF의 구조와 기능 연구를 했으며 최초로 바실러스 세레우스의 MazF 톡신 구조를 구했다. 특히 바실러스 MazF는 PemK와 같은 fold와 유사성을 지니며 RNA-substrate recognizing loop을 가지고 있는데 이는 매우 높은 resolution으로 관측되었다. 바실러스 세레우스의 MazF 주요 잔기는 촉매활성을 가지고 있으며 mutation 실험을 통해 in vitrto assay를 진행한 결과 이 잔기들이 ribonucleic activity를 가지고 있고 celluar toxicity에 매우 중요한 인자들이라는 것을 알 수 있었다.I.Introduction(1) 1.1. 20세기 아르헨티나 소설에 나타난 정체성 탐구

인류 역사를 통해서 인간이란 무엇인가라는 질문은 가장 근본적이면서도 가장 오래 동안 지속된 물음이었으면서도 아직도 정확한 답을 찾지 못한 질문이다. 게다가 근대 국가가 성립된 이후 또 다른 정체성에 대한 물음이 추가되었다. 나는 어떤 나라의 국민인가? 어느 국민의 특질 혹은 속성은 무엇인가? 어느 국가의 문화적 성격은 어떻게 규정될 수 있는가? 따위의 질문들이 이어진다. 오랜 세월 동안 혈통적, 언어적 동질성을 유지해온 국가에 있어서 국가적 정체성을 발견하거나 정체성에 대한 국민적 공감대를 확립하는 것은 별로 어려운 문제가 아니다. 하지만 중남미처럼 불과 수백 년 전에 완전히 이질적인 두 문화가 급격하게 충돌하고 한 문화가 다른 문화를 강제적으로 지배, 대체하려 하였고, 19세기 독립 이후 각 지역마다 정치, 경제, 사회, 문화적 발전과정이 상이한 경우 그곳의 한 개인, 집단, 사회, 국가의 정체성을 정의한다는 것은 실로 어렵고도 긴요한 문제로 부각될 수밖에 없다..

인터뷰 - 엔리케 두셀을 만나다

지난 8월 초에 개최된 세계철학자대회 참석차 방한한 엔리케 두셀 (Enrique Dussel)은, 서울대학교 라틴아메리카연구소 주최의 <오르비 스 테르티우스(Orbis Tertius): 라틴아메리카 석학에게 듣는다>에서 유럽중심적 사유에 대한 현대 라틴아메리카 지성인의 비판적 관점을 소개하여 청중들로부터 뜨거운 반응을 얻었다. 이 글은 서울대학교 라틴아메리카연구소 소장 김창민 교수가 8월 7일 엔리케 두셀을 인터뷰한 전문이다. 인터뷰 내용의 일부는 8월 11자에 소개되었다

Estudio Comparativo de Hijo de Hombre(1960) de Augusto Roa Bastos e Hijo de Hombre(1979) de Mun-Yol Lee: En torno al Tema de la Salvación

Este estudio tiene como objetivo comparar dos novelas homónimas de Paraguay y Corea. Además del título idéntico, tienen varios aspectos comunes: el tema, el fondo y la estructura. A pesar de coincidir en dichos aspectos, son muy diferentes en cuanto al entendimiento del Dios cristiano y de la salvación. La obra de Roa Bastos insinúa que la salvación del ser humano podría realizarse en este mundo a través del comportamiento humanitario y sacrificado del ser humano, como el que se manifiesta en los personajes de Gaspar Mora, Casiano Jara y Cristóbal Jara. Por eso, éste último llega a decir: Lo que no puede hacer el hombre, nadie más puede hacer. Así, el camino de la salvación se presenta más abierto. La obra de Lee, una novela policiaca, comprende mayormente los vagabundeos y razonamientos teológicos de Min (estudiante rebelde de teología), de Yio (seguidor de Min) y de Ahas Pertz (en la escritura de Min) sobre el Salvador. Al final, Min regresa al Dios cristiano del que se había apartado, y Yio, al darse cuenta de la traición de su maestro, lo mata y se suicida. En la novela coreana, nunca se muestra suficiente confianza en la capacidad del pueblo de salvarse a sí mismo, ni se piensa en las virtudes divinas del ser humano que, según Jesús, Dios le ha otorgado desde siempre

People's Response to Policy Making for Radioactive Waste Disposal

이 연구의 목적은 지역에 대한 국가의 정책 결정에 대해 지역 주민들이 어떻게 대응하는지를 민족지적으로 분석하는 것이다. 국가와 지역은 지배와 피지배의 구도 속에 존재하고 있기는 하지만 지역이 항상 국가 지배에 대해 순응하기만 하는 것은 아니다. 이런 사실은 정책의 집행과정에서 지역 주민들이 대항하거나 저항하는 모습을 통해서 나타난다. 지역은 국가에 대한 대항권력의 지위를 지니고 있으며, 양자 간 상호작용과 긴장 그리고 균형에 의해 국가와 지역 사이의 관계가 정해질 수 있다. 이 연구에서 분석하고자 하는 국가의 정책은 위도를 대상으로 한 방사능 폐기물 처리장 건립이다. 여러 차례의 타당성 조사와 건립 시도 끝에 정부는 2003년에 부안군의 신청을 받아 위도에 방사능 폐기물 처리장을 건립하기로 결정하였다. 그러나 이 결정에 대해 부안군 주민들은 반대 의견을 표시하였으며, 전면적인 반대 운동을 전개하였다. 정작 당사자인 위도 주민들은 유치 신청서에 서명을 하였지만 부안군민들의 거센 반대 운동과 국가의 일관성 없는 정책 추진으로 찬성과 반대 어느 입장도 공개적으로 지지하지 못하는 입장이 되었다. 위도에 방사능 폐기물 처리장을 세우기로 한 정책 결정은 전국적인 관심사가 되었고 부안군은 이 논쟁 속에서 2년의 세월을 보내야 했다. This study aims to describe the process of debate on radioactive waste management in Wido and to present an analysis of some of the social and political dimensions of a environmentalism. Radioactive waste management has been one of the main issues of environment movements in Korea. Governments of Korea decided to build radioactive waste installations in Wido. But Buan county people opposed the decision violently. So, opinion about radioactive waste installations has been divided between Wido and Buan County. This dispute is not simple problem to accept or oppose to radioactive waste installations. This dispute contains knowledge as social process, the tension between 'nuclear' and 'people' perspectives, the tradition of compensation in Buan country, and local level politics. I understand the Buan dispute as a discursive field where different perspectives engage and compete. Change of Relationship Between Jeju Residents Society in Japan and Jeju Village: 1930-2000

지금까지 재일교포에 대한 연구는 많이 이루어졌으나 대부분의 연구는 재일교포들의 사회문화적 실상을 다룬 것이다(이광규 1983: 1987. 이문웅 1988: 1989 등). 이 연구들은 문화변동의 관점에서 재일교포를 분석하여 일본문화와 한국문화가 어떻게 접변하였으며, 현재 재일교포들의 문화가 어떻게 일본문화화 되어 가는지를 다루었다. 한편 제주도 마을의 지역 개발에 대한 연구(전경수, 한상복 1996: 1999)는 정부의 역할에 초점이 맞추어졌다. 정부가 지역개발에서 한 역할의 중요성은 매우 크다. 그러나 제주도의 경우에는 다른 지역과 달리 재일교포가 지역 개발 과정에 적극 참여하였고, 이것이 제주도 지역개발의 특징이라고 할 수 있을 정도이다. 그러나 기존의 연구들에서는 재일교포의 역할이 크게 주목 받지 못하였다. 이 연구는 제주의 마을과 그 마을 출신 재일교포사회 사이의 관계를 분석하는 것이 목적이다. 이 관계를 분석함으로써 궁극적으로 제주 마을과 제주사람들의 문화를 이해하는 것을 지향한다. 구체적으로 이 연구의 목적을 나열하면 다음과 같다. 첫째, 제주 출신 재일 교포 사회의 형성과 변화 과정을 분석한다. 둘째, 재일 교포와 제주 마을 사이의 인적, 물적 교류 유형을 역사적으로 분석한다. This study aims to analysis the change of relationship between Jeju residents society in Japan and Jeju village. For this aim, I have done anthropological field research in Jeju village and Jeju resident society in Japan. The relationship between Jeju resident society in Japan and Jeju village has been changed in accordance with socio-economic conditions of Jeju villagers and Jeju residents in Japan. Under the rule of Japanese Imperialism when both Jeju islanders and residents in Japan had hard time economically, the relationship was most amicable. After Liberation when the economy of the residents in Japan were prosperous and that of Jeju villagers were bad, the relationship was patron-client relationship. But after 1980, the economy of Jeju villagers was boom up, the relationship became alienated and personalized. The Influence of Computer-Assisted Instruction Emphasizing the Particulate Nature of Matter on High School Students' Conceptual Understanding in Chemistry

This study examined the influences of computer-assisted instruction (CAI) upon high school students' conceptual understanding about gas laws. The CAI programs were designed to supply animated molecular motions for emphasizing the particulate dynamic nature of matter. The CAI and the control groups (2 classes) were selected from a girls high school in Seoul, and taught about gas laws for four class hours. After the instructions, students' conceptions were measured by the Chemistry Conceptions Test (CCT). The Kinship Structure in Huksan Islands: 'Kunjip' and 'Nerong'

이 연구는 흑산도의 한 어촌마을을 대상으로 하여 친척을 연구한 민속지 연구다- 연구자는 조사 마을의 친척을 통하며 친척조직의 지역적 한 유혈을 파악함으로써 한국 친척 연구의 일반화를 위한 새로운 사례를 제공하고자 하였다. 연구자는 생태적 경제적 조건에 대한 적응의 결과물로서 조사마을의 친척조직을 파악하고자 하였다 이 연구에서 얻은 결과물은 다음과 같다. 첫째, 연구마을에서는 제사가 장남에게 상속도|지 않고 마을에 남아 부모와 함께 생활한 아들에게 상속된다는 점이다. 조상의 신위인 기성은 큰집 에만 모셔지기 때문에 장남이라도 분가하면 제사를 모실 수 없게 된다. 장남 분가는 연구 마을의 생태적 환경의 결과물이다. 열악한 경제환경으로 연구마을은 많은 수의 인구를 부양할 수 없어서 인구 유출 압력 크다. 뿐만 아니라 마을 공동재산에 대한 권리 확보를 위해서 장남 분가를 하는 것이 더 적응적이다. 이런 조건 때문에 장남은 분가하여 마을을 떠나고 남은 아들이 남아 가계를 계승하는 것이 일반화되어 있다. 둘째, 연구마을에서는 내롱 이라고 하는 친족조직이 중요한 사회 단위로 기능하고 있다. 한 아들만 남고 나머지 아들은 분가하여 마을을 떠나는 관행으로 마을에 남아 없는 사람들의 친척관계는 상당히 멀다. 8촌 이내의 혈족의 수가 적기 때문에 당내를 구성하는 것은 매우 어려우며, 종손이나 종가의 개념도 없어 문중이 형성되지도 못하고 없다. 이 상황에서 같은 파에 속한 혈족들로 구성된 내롱 은 먼 친척을 가깝게 인식하게 하며 친척집단이 수행해야 하는 여러 역할을 담당하는 단위로 기능하고 있다 내롱의 가장 중요한 기능은 시제를 모시는 것이다 문중이 없기 때문에 시제는 내롱을 단위로 순환하여 모신다. 그리고 개인 책임을 지는 제사는 점차 내롱이 책임을 지는 시제로 전환되어 일찍 제사를 중단하는 경향도 나타나고 있다. 즉, 큰집의 중요성은 줄어드는 반면 내롱의 역할은 증대하고 있다. 셋째, 연구마을에서는 작숙, 오춘과 같이 먼 친척을 가까운 친척으로 인식하게 하는 문화적 장치들도 발달해 있다. 이는 내롱이 먼 친척을 가까운 친척으로 인식하게 하는 경향과 같은 맥락이다. 나아가 작숙은 친척이 반드시 혈족만으로 구성되지 않고 인척관계에 있는 사람까지 포함하고 있음을 보여주는 사례이다. 마을에 살고 있는 혈족의 수는 적은 반면 인척의 수는 많기 때문에 이들과 협력관계를 유지하기 위하여 이러한 관행이 발전한 것으로 보인다. This article is an ethnographic study of a village in Huksan Islands to analyze kinship structure. This study aims to offer a case for generalizing Korean kinship system by understanding a local type kinship system in my village. I think of kinship system of my village as a result of ecological and economical adaption. The findings of this study can be drawn as follow. First of all, Jesa which is ancestor worship serviced annually at the death day is not inherited to one's eldest son but to one of his son who has lived with him. Because memorial tablets are enshrined in ancestor worship room which is located only 'Kunjip' , inherited house, the eldest son who set up a separated family can not hold Jesa. The fact that the eldest son set up a new separated family is a result of ecological adaptation. There are small land and scarce resources, the village could not support many dwellers and it is adaptable to leave elder son his parent family early. Secondly, 'Nerong', which is one sort of kin groups works as a very important social unit. The size of patrilineal descent group is very small and kinship relation is relatively far, neither Dandnae nor Munjung is organized in my village. In this situation Nmmg which is composed of the members of a branch of Munjung functions as a kin group. The most important function of Nerong is to serve Sije. Thirdly, relatively far removed kin are regarded as near of kin by kinship terms like 'Jaksuk', 'Ochun'. It is same context of 'Nerong'. Nerong also regards far removed kin as near of kin. And Jaksuk is a case to insist that kinship is not consist of consanguine only but consist of relatives also. In my village, the number of consanguines are small but relatives are abundant, so it is necessary to obtain cooperation from relatives. In this context, it is appreciated that such cultural elements like Jaksuk are cultural adaptation.이 논문은 2000년도 학술진흥재단의 중점연구소 지원사업의 일환으로 이루어졌음
