    鉴定AP-4是在非激素依赖型前列腺癌中上调L-plastin表达的转录因子

    【目的】鉴定AP-4是在非激素依赖型前列腺癌中调控L-plastin表达的转录因子,进一步阐明非激素依赖型前列腺癌的发病机制。【方法】在鉴定L-plastin启动子近3’端-216到1序列片段存在明显的转录活性的基础上,利用TFSEARCH软件分析该片段的序列,寻找可能调控L-plastin表达的非甾体激素类转录因子,并用凝胶滞后实验和超滞后实验证实结合于该片段的转录因子,利用PCR定点突变法构建删除该转录因子结合位点的重组子,并检测切除该片段序列后荧光素酶活性,研究该转录因子在非激素依赖型前列腺癌中调控L-plastin表达的作用。【结果】TFSEARCH软件分析表明在距转录起始点-199-194bp处含有AP-4转录因子结合位点CAGCTG,凝胶滞后实验和超滞后实验表明AP-4是结合于L-plastin启动子3’端序列CAGCTG的转录因子,删除AP-4结合位点后荧光素酶活性下降。【结论】AP-4是促进L-plastin在非激素依赖型前列腺癌中表达的重要调控因子

    两种不同入路的腹腔镜肾上腺嗜铬细胞瘤切除术

    【目的】探讨两种不同入路的腹腔镜肾上腺嗜铬细胞瘤切除术。【方法】本组25例,男性14例,女性11例,平均53.3岁。左侧15例,右侧10例。术前均用苯苄胺等药物准备2~4周。经腹腔入路施行腹腔镜肾上腺嗜铬细胞瘤切除术11例,经腹膜后入路14例。在手术刚开始、建立气腹后、分离肿瘤时、切除肿瘤后分别记录心率、平均动脉压、中心静脉压的变化。【结果】1例早期经腹腔入路手术的病例在术中发现肿瘤粘连紧密改为开放性手术,其余24例在腹腔镜下顺利切除肿瘤。术后病理检查证实均为肾上腺嗜铬细胞瘤。术中脉搏血压没有剧烈波动。术后血压稳定,疼痛较轻,术后5~7d出院。经腹腔入路组与经腹膜后入路组在手术中心率、平均动脉压、中心静脉压以及手术时间和出血量等方面均没有显著差异(P>0.05)。无气胸、腹腔内脏器损伤、肠粘连等并发症。【结论】两种不同的入路施行腹腔镜肾上腺嗜铬细胞瘤切除术都是安全可行,具有操作精细、创伤小、恢复快等优点

    考察了邻羟基苯甲酸和邻氨基苯甲酸类化合物的荧光自猝灭现象,研究表明,较高浓度的邻羟基苯甲酸和邻氨基苯甲酸等一类物质,在光激发下,通过分子间氢键形成激-基二聚体,使荧光性的单体量减少,从而产生自猝灭.此自猝灭荧光探针可指示体系微环境的变化,并成功地应用于dnA的定量测定.In pH 10.50 wide buFFer solution, nitrazepam under the conditions of the irradiation of ultraviolet light and the presence of H 2O 2 produces a Fluorescent product ( λ ex =355 nm, λ em =465 nm) .A new method For the Fluorimetric determination of nitrazepam was developed .The Fluorescence intensity is linearly increased with the concentration of nitrazepam over the range of 0.25 ̄3.00 mg·L -1 .The relative standard deviation and detectable limit are 2.3% and 0.13 mg·L -1 respectively .This method has been applied to the determination of nitrazepam in tablets.国家教委博士点基金;福建省自然科学基
