Enrichment and Resistance Mechanism of Aegiceras corniculatum to Gas Elemental Mercury
红树林是生长在热带、亚热带地区的木本植物群落,具有极为重要的生态价值和经济价值。红树植物具有较高的富集汞的能力,其叶与大气之间存在持续的汞交换过程。目前,关于红树植物对离子汞耐受性的研究已有很多,而有关红树植物对气态汞胁迫的响应及其耐受性机制的研究却鲜有报道。因此,本研究以桐花树(Aegicerascorniculatum)幼苗为材料,利用封闭通量箱测定其在饱和气态汞(Hg0)胁迫下的生理、解剖结构和亚细胞结构的变化,探究桐花树叶片与大气汞交换的昼间变化规律及其影响因素。探讨其对汞的富集和耐受机制。研究结果表明: (1)桐花树幼苗叶片对气态汞胁迫的响应及其各部位对汞的吸收和累积结果显示:Hg...Mangroves are woody plant communities, which locate in tropics and subtropics zone, possessing very important ecological and economic value. Physiological Response of Aegiceras corniculatum Leaves to Hg~0 Stress
桐花树是我国主要的广布种红树植物,具有泌盐性。通过胁迫实验,研究桐花树幼苗叶片生理生态指标对气态单质汞(Hg0)胁迫的响应,以期了解红树植物吸收和富集气态汞的机制。结果表明:气态汞胁迫造成桐花树叶片可溶性糖含量、过氧化物酶(POD)活性下降;而丙二醛(MDA)、脯氨酸、超氧化物歧化酶(SOD)等指标上升;叶绿素含量、过氧化氢酶(CAT)活性出现先升高后降低的趋势,还原型谷胱甘肽(GSH)含量则先降低后升高。总之,虽然Hg0胁迫可增加桐花树幼苗叶的膜质过氧化程度,影响其生理代谢活动,但其叶片可通过调节抗氧化酶活性及还原型谷胱甘肽含量来提高自身抗氧化能力,进而提高对Hg0的抗性。As one of the main widespread mangrove plants in China, Aegiceras corniculatum can tolerate salt by excreting salt. Mangroves are facing more and more serious heavy metal pollution, especially the mercury contamination. To investigate the mechanism of mercury absorption and tolerance of mangrove, stress experiments were conducted with Aegiceras corniculatum seedlings under Hg0 stress. In this experiment, physiological characteristics such as the contents of chlorophyll, soluble sugars, malondialdehyde( MDA), proline( Pro), and reduced glutathione(GSH), activities of superoxide dismutase(SOD), peroxidase(POD) and catalase(CAT) of Aegiceras corniculatum seedlings' leaves were measured. Results showed as follows: the content of soluble sugars and the activity of POD gradually decreased under Hg0stress; while MDA content,Pro content and SOD activity significantly increased; the chlorophyll content and CAT activity increased firstly and then decreased, and on the contrary, the content of GSH decreased initially and then increased. The findings from above study demonstrate that although Hg0 tress can lead to damage of metabolism by causing membrane lipid peroxidation in seedlings 'leaves, but the seedlings could change activities of the antioxidative enzymes and the content of GSH to enhance the antioxidant abilities, and then strengthenorganism resistance to mercury.国家自然科学基金(No.41176090
便携拉曼检测集成装置与芯片式卡盒相结合用于食品添加剂的快速检测
针对食品安全体系拉曼光谱检测的自动化前处理问题,研制了一台便携拉曼检测集成装置,能与芯片式卡盒相结合以实现食品添加剂的前处理及检测。集成装置主要包括触控屏、控制板、通讯模块、蠕动泵及小型拉曼光谱仪。蠕动泵对芯片式卡盒所需的溶剂进行程序驱动,自动完成检测前的前处理工作;小型拉曼光谱仪对前处理收集液进行检测;控制板对蠕动泵及拉曼光谱仪的参数进行设定监控;通讯模块则可把检测数据传输到WIFI设备或云数据中心,并接受指令。将集成装置应用于糖果中食用色素的检测,结果表明日落黄与柠檬黄的检测限均可达到10 mg/L。该装置体积小、检测过程快,适合用于食品安全体系的现场快速检测。国家自然科学基金(NO.U1505243);;福建省自然科学基金(NO.2015J01064);;福建省高校青年自然基金重点项目(NO.JZ160404);;国家重大科学仪器设备开发专项(2011YQ030124)资
星系中心大质量黑洞及潮汐瓦解恒星事件
黑洞潮汐瓦解恒星事件(Tidal Disruption Events,TDE)是星系中心黑洞瓦解进入其潮汐瓦解半径内的恒星并吸积恒星碎片物质而产生的一种剧烈辐射耀发现象.TDE的能谱和光变特征中蕴含了中心黑洞和被瓦解的恒星的信息,为我们证实和普查宁静星系中的黑洞,研究其参数、吸积过程和喷流产生、以及核区星际介质等提供了可能.TDE还可能提供中等质量黑洞和双黑洞存在的证据.TDE的观测和理论已成为一个新开辟的天体物理研究领域,但目前的进展受制于探测到事件太少(尤其是在X射线波段),且观测数据普遍质量不高.TDE的发生率很低,要探测大样本的事例需要监测足够大的空间体积.爱因斯坦探针卫星(Einstein Probe,EP)覆盖了0.5–4 keV的软X射线波段(接近TDE耀发时的辐射峰值能段),具有大视场以及高灵敏度,非常利于对TDE的探测.预期爱因斯坦探针卫星每年可以发现约几十至上百例TDE,其中有约10例或更多具有相对论性喷流特征.这将使我们可以获得较为完备、具有统计意义的TDE的样本,为进一步研究黑洞的存在和统计性质、增长和演化、发现中等质量黑洞和大质量双黑洞等提供了新的途径.中国科学院空间科学战略性先导科技专项(编号:XDA15052100);;北京大学“985工程”建设项目“星团环境对双黑洞形成演化过程的干扰及其对引力波探测的影响”资
Matrix effect and retention efficiency of hydrophilic-lipophilic balance cartridges in multi-residual determination of veterinary drugs in river water
基质效应是液相色谱-质谱联用(lC-MS)定量分析中的重要干扰因素。以反渗透水为空白对照进行lC-MS/MS检测,在500倍浓缩条件下考察了33种兽药分别经WATErS、SuPElCO和CnW等3种亲水亲脂平衡(HydrOPHIlIC-lIPOPHIlIC bAlAnCE,Hlb)固相萃取柱富集后的基质效应及其保留效果。结果表明,以反渗透水为基质时3种Hlb柱均表现出外源性基质效应,对大部分喹诺酮类和四环素类兽药呈现促进效应,有两种Hlb柱对雌激素类兽药等呈现抑制效应,对磺胺类兽药的基质效应不显著,有一种Hlb柱对氯霉素类兽药呈现抑制效应;以河水为基质时,由于外源性杂质与内源性杂质共同作用,对大部分喹诺酮类和四环素类兽药呈现促进效应,对氯霉素类和雌激素类兽药呈现抑制效应,对磺胺类兽药基质效应不显著。与外标法相比较,基质匹配标准校正法可有效消除基质效应对检测结果的影响。在50 ng/l和200 ng/l两个加标水平下,WATErS、SuPElCO和CnW 3种Hlb柱对河水中33种目标兽药的校正后的回收率分别为40.3%~146.0%、37.8%~104.2%和52.9%~150.1%;rSd(n=4)为0.2%~14.6%。相同实验条件下3种Hlb柱的外源性基质效应不容忽视。而采用基质匹配标准校正法克服目标物基质效应,保留效果无显著差异,富集效果良好,为河水样品中兽药多残留分析方法中固相萃取柱的选择提供了科学依据。Matrix effect is an important interfering factor in LC-MS quantitative analysis.In this paper,matrix effects and retention efficiencies of 33 veterinary drugs spiked in river water were studied on hydrophilic-lipophilic balance( HLB) cartridges of 3 brands( Waters,Supelco,and CNW),using LC-MS /MS for detection and reverse osmosis( RO) water as the control under 500-fold concentration.In RO water,only the exogenous matrix effects were observed on three brands of HLB cartridges.Most quinolones and tetracyclines showed positive matrix effects.Estrogens showed negative matrix effects on two brands of HLB cartridges.Sulfonamides were not obviously affected by matrix effects.Chloramphenicols showed negative matrix effects on one brand of HLB cartridge.In river water,matrix effects were different from those of the RO water due to the combined exogenous and endogenous interfering substances.Sulfonamides showed slight matrix effects as those in RO water.Most quinolones and tetracyclines showed positive matrix effects.Chloramphenicols and estrogens showed negative matrix effects.Compared to the external standard method,matrix matched calibration method effectively overcame the matrix effects with better quantitative results.The recoveries of 33 target veterinary drugs spiked in river water at 50 ng / L and 200 ng / L levels were in the ranges of 40.3%- 146.0%( Waters),37.8%- 104.2%( Supelco),and 52.9%- 150.1%( CNW) with RSDs( n = 4) of 0.2%- 14.6%.The results indicated that there was no significant difference in the retention efficiency between the 3 HLB cartridges with the matrix matched calibration method.This study provided supporting data for the HLB cartridge selection in multiresidual determination of the veterinary drugs in river water samples.福建省自然科学基金项目(2012J01048); 中国科学院城市环境研究所开放基金项目(KLUEH201009

口服益生菌改善铝诱发的小鼠海马炎症
【目的】探讨口服益生菌能否改善铝暴露诱发的小鼠海马炎症。【方法】通过给予小鼠口服氯化铝,建立海马炎症模型,将24只8周龄雄性小鼠C57BL/6随机分成对照(CON)组、氯化铝处理(Al)组、益生菌处理(PO)组和联合处理(Al+PO)组,每组各6只,分别持续给予口服氯化铝和(或)益生菌6周。海马神经炎症的评估,采用免疫荧光染色法检测小鼠海马组织中激活的小胶质细胞(Iba-1+/CD68+)数量以及脑源性神经营养因子(BDNF)的表达量,酶联免疫吸附测定法检测血清和海马组织中白介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)的含量。【结果】免疫荧光染色法研究结果显示:与CON组相比,Al组海马组织中Iba-1+/CD68+细胞数量显著增加(<0.01),而BDNF表达量显著减少(<0.01);与Al组相比,Al+PO组海马组织中Iba-1+/CD68+细胞数量显著减少(<0.01),而BDNF表达量增加(<0.01)。酶联免疫吸附测定(ELISA)研究结果显示:与CON组相比,Al组血清和海马组织中的IL-1β和TNF-α含量均有显著增加(<0.01);与Al组相比,Al+PO组血清IL-1β含量显著减少(<0.01),海马组织中的IL-1β和TNF-α含量均显著减少(<0.01)。【结论】口服益生菌可改善铝暴露诱发的小鼠海马炎症
以β-淀粉样蛋白为靶的单价及二价真核表达载体的构建和表达
【目的】构建以β-淀粉样蛋白为靶单价及二价真核表达载体pcDNA3.1-Aβ1-42、pcDNA3.1-AB42x,并在真核细胞中表达目的蛋白。【方法】PCR法扩增编码B一淀粉样蛋白的目的基因,利用基因克隆技术构建单价及二价真核表达载体;应用酶切及测序鉴定真核表达载体构建成功后,用Western blot和细胞免疫组化染色检测其在真核细胞中的表达。【结果】相应的双酶酶切能够获得插入的目的基因片段(分别为137bp和269bp),测序未发现突变;Western blot结果可见两条约为4ku和8ku的条带,细胞免疫组化染色可见阳性细胞。【结论】单价及二价真核表达载体构建成功;真核表达载体能在真核细胞中表达出目的蛋白
